Low erucic acid (LEA) rapeseed, which has accumulated mutant fatty acid elongase genes at the BnFAE1.1 and BnFAE1.2 loci of the A- and C-genome, respectively, is an important oilseed crop. Short growing turnip rape (B. rapa) is also important as a catch crop in the continuous cropping of rice in Asia but there is no LEA B. rapa cultivar for cultivation in South Asia. In order to develop LEA turnip rape cultivars, high erucic acid turnip rape cultivars were interspecifically crossed as recurrent parents to a canola quality rapeseed. In the meantime, we monitored incorporation of the mutant bnfae1.1 (e1) gene into A-genome of turnip rape, using a dCAPS primer pair, which can amplify PCR fragment only for the mutant e1 gene from A-genome. The early backcross progenies showed poor seed set, but which was improved in advanced progenies. Finally, homozygous e1e1 genotypes were established in the selfed progenies of BC2–BC3, and their LEA content was confirmed by gas-chromatography analysis. Our results and promising lines will contribute to LEA-trait selection in turnip rape and rapeseed breeding.  相似文献

The meal value of Soybean for monogastric animals is determined partly by sucrose, raffinose and stachyose. Of these, sucrose is desirable, while raffinose and stachyose are indigestible, causing flatulence and abdominal discomfort. The objective of this study was to identify quantitative trait loci (QTL) controlling seed sucrose, raffinose, and stachyose in a set of 140 SoyNAM (Nested Association Mapping) recombinant inbred lines (RILs), developed from the cross between lines IA3023 and LD02‐4485. A total of 3,038 SNP markers from the Illumina SoyNAM BeadChip SNP were used to map the QTLs for sucrose and the RFOs, raffinose, and stachyose. Significant genotypic differences (p < .001) among RILs were observed for sucrose, raffinose and stachyose contents across years. A 3038 Illumina SoyNAM BeadChip SNPs identified three QTLs for sucrose, one on chromosome 1, explaining 10% variance and two on chromosome 3 each explaining 22%. Raffinose QTL was detected on chromosome 6, explaining 6% variance. The mapped QTLs were novel and spanned regions harbouring candidate genes with roles in plant growth including seed development.  相似文献

Subsurface drip irrigation systems, compared to other irrigation systems, enhance the delivery of water and nutrients directly into the root zone. However, in light-textured soils, certain quantities of water may percolate below the root zone due to the subsurface position of drip lines and/or poor management of irrigation systems. The main objective of this paper is to evaluate three technologies to enhance a spatial distribution of water and solutes in the root zone and to limit downward leaching. The three technologies include (a) a physical barrier, (b) a dual-drip system with concurrent irrigation, and (c) a dual-drip system with sequential irrigation. To achieve this objective, we performed computer simulations using the HYDRUS (2D/3D) software for both bare and vegetated soils. The results indicate that the physical barrier is more efficient than dual-drip systems in enhancing the water distribution in the root zone while preventing downward leaching. On the other hand, the dual-drip system improves water distribution in sandy soils. Additionally, the dual-drip system with sequential irrigation, followed by the dual-drip system with concurrent irrigation, is the most efficient in limiting downward leaching of solutes.  相似文献

Background

The SUV3 (suppressor of Var 3) gene encodes a DNA and RNA helicase, which is localized in the mitochondria. Plant SUV3 has not yet been characterized in detail. However, the Arabidopsis ortholog of SUV3 (AT4G14790) has been shown to be involved in embryo sac development. Previously, we have reported that rice SUV3 functions as DNA and RNA helicase and provides salinity stress tolerance by maintaining photosynthesis and antioxidant machinery. Here, we report further analysis of the transgenic OsSUV3 rice plants under salt stress.

Findings

The transgenic OsSUV3 overexpressing rice T1 lines showed significantly higher endogenous content of plant hormones viz., gibberellic acid (GA3), zeatin (Z) and indole-3-acetic acid (IAA) in leaf, stem and root as compared to wild-type (WT), vector control (VC) and antisense (AS) plants under salt (200 mM NaCl) stress condition. A similar trend of endogenous plant hormones profile was also reflected in the T2 generation of OsSUV3 transgenic rice under defined parameters and stress condition.

Conclusions

In response to stress, OsSUV3 rice plants maintained plant hormone levels that regulate the expression of several stress-induced genes and reduce adverse effects of salt on plant growth and development and therefore sustains crop productivity.  相似文献

This study aimed to evaluate color Doppler sonographic findings in carotid arteries in RA patients under pharmacological treatments and to compare them with normal population. Forty nine patients with late RA and 48 healthy age and sex-matched controls were recruited. The two groups were matched for other known risk factors of atherosclerosis including serum lipid abnormalities, smoking status, diabetes mellitus and hypertension. High resolution B-mode color Doppler ultrasound with a 7 MHZ transducer was used for measuring the Common Carotid Intima-Medial Thickness (CCIMT) in both sides in all subjects. Presence of atherosclerotic plaque was also investigated. The mean left and maximum CCIMT was significantly higher in the case group (0.72 vs. 0.62 mm for the left artery; p < 0.01; 0.72 vs. 0.64 mm for the maximum reading; p = 0.01). No atherosclerotic plaque was found in common carotid arteries. There were 3 (6.1), 7 (14.3) and 9 (18.4%) plaques in left internal carotid artery, right carotid bulb and left carotid bulb in the case group, respectively with no atherosclerotic plaques in the controls (p = 0.24, 0.01 and < 0.001, respectively). Comparing the findings by gender in the case group with the controls, the mentioned significant differences were only between the male patients and the controls. The process of atherosclerosis in RA patients is similar to that in normal population. However, it is apparently accelerated and more advanced in these patients.  相似文献

As the use of high performance textiles has grown, the need for chemical finishes to provide the fabric properties required in the special applications has grown accordingly. In this project, a series of water dispersible polyurethanes dispersion (CS-PUs) with multipurpose performance profile was developed using isophorone diisocyanate (IPDI), polyethylene glycol (PEG), 2,2-dimethylol propionic acid (DMPA) and chitosan (CS) for textile applications. In two step synthesis process, NCO functional PU prepolymers prepared by reacting IPDI, PEG, and DMPA were extended with varying molar quantities of chitosan followed by structural characterization through FTIR. The prepared CS-PU dispersions were applied onto the dyed and printed poly-cotton blend fabrics. The performance behavior of the treated fabric in terms of crease recovery, tear strength, tensile strength, and antibacterial properties was evaluated by applying standard test methods. These investigations show that the CS-PU dispersions can be applied as antibacterial textile finishes with significant improvement in the physical and mechanical properties of poly-cotton fabrics.  相似文献

Background:Azo dyes are the most widely used synthetic colorants in the textile, food, pharmaceutical, cosmetic, and other industries, accounting for nearly 70% of all dyestuffs consumed. Recently, much research attention has been paid to efficient monitoring of these hazardous chemicals and their related metabolites because of their potentially harmful effect on environmental issues. In contrast to the complex and expensive instrumental procedures, the detection system based on the QDs with the superior optochemical properties provides a new era in the pollution sensing and prevention. Methods:We have developed a QD-enzyme hybrid system to probe MR in aqueous solutions using a fluorescence quenching procedure. Results:The azoreductase enzyme catalyzed the reduction of azo group in MR, which can efficiently decrease the FRET between the QDs and MR molecules. The correlation between the QDs photoluminescence recovery and MR enzymatic decolorization at the neutral phosphate buffer permitted the creation of a fluorescence quenching-based sensor. The synthesized biosensor can be used for the accurate detection of MR in a linear calibration over MR concentrations of 5-84 μM, with the LOD of 0.5 μM in response time of three minutes. Conclusion:Our findings revealed that this fluorometric sensor has the potential to be successfully applied for monitoring a wide linear range of MR concentration with the relative standard deviation of 4% rather than the other method. Background:Hyperuricemia induces nephropathy through the mediation of oxidative stress, tubular injury, inflammation, and fibrosis. The high uric acid level is associated with the reduction of vitamin D levels. However, the reno-protective effects of this vitamin in hyperuricemia condition remain unknown. This study aimed to elucidate calcitriol treatment in a uric acid-induced hyperuricemia mice model. Methods:Uric acid (125 mg/kg BW) was administered intraperitoneally for 7 (UA7) and 14 (UA14) days. Calcitriol (0.5 g/kg BW) was intraperitoneally injected for the following seven days, after 14 days of uric acid induction (UA14VD7 group). The control group received NaCl 0.9%, by the same route. Serum creatinine was measured using calorimetric method, and uric acid levels were assessed using enzymatic calorimetric assay. Tubular injury and fibrosis were assessed using PAS and Sirius red staining. RT-PCR and qRT-PCR were carried out for the analyses of SOD-1, Collagen-1, and TGF-1 mRNA expression in the kidney. Immunostaining of SOD-1 was performed to detect its expression in the kidney. Results:Uric acid and creatinine levels markedly increased in UA14 groups, followed by an exacerbation of tubular injury. RT-PCR revealed the upregulation of Collagen-1 and TGF-1, along with the downregulation of SOD-1. Calcitriol treatment attenuated the injury with reducing uric acid and creatinine levels, as well as tubular injury. This was associated with lower Collagen-1 and TGF-1 mRNA expression compared to the UA7 and UA14 groups. SOD-1 was upregulated in epithelial cells in the UA14VD7 group. Conclusion:Calcitriol treatment after uric acid induction may attenuate kidney injury through upregulation of SOD-1 and downregulation of Collagen-1 and TGF-1 gene expression. Sodium adsorption ratio (SAR) is one of the water quality indexes that whose is important due to reuse or depletion to environment. Solutes in drain water can be controlled by adsorption, chemical or biological reaction, organic envelope of drainage. Rice husk is the common option of drainage envelops in paddy fields. In this study, the ability of reduction of SAR by rice husk was evaluated in batch scale and physical model of drain envelops. In the batch experiments, the adsorption of SAR parameters was investigated by adding 2 g of rice husk into a 100 ml of sodium chloride solution. The results indicated that rice husk absorbed calcium, magnesium and sodium, respectively. By increasing the temperature, contact time and pH, adsorption of calcium, magnesium and sodium was increased; however, the higher concentration of sodium in soil solution reduced the percentage of adsorption. In a more realistic state, physical models of subsurface drainage in the paddy fields were made. Drainage envelope treatments included of rice husk (H), combination of 20 and 60 % of husk with gravel (H20G80 and H60G40) and a pipe without envelope (NE). Due to higher drain discharge and more sodium removal (lower SAR in drain water), treatment H with the discharge of 16.2 ml/min and SAR of 1.27 (meq/l)0.5 was better in comparison with other treatments.  相似文献
