BACKGROUND: Swab cytology of ear canals is one of the most useful and rapid methods to assess the presence of external ear infections. Smears are generally stained with rapid Romanowsky-type stains, with or without prior heat fixation. OBJECTIVES: The aim of this study was to compare 4 different methods of fixation and staining of ear swab cytology samples from dogs. METHODS: Eight dogs with otitis externa were selected from a dermatology referral population. A cotton swab was used to obtain ceruminous material from 12 ear canals. Four smears of each swab were prepared on glass slides (randomly identified as A, B, C, or D) and air-dried for cytologic examination. Samples marked A were stained with Dip Quick (Jorgensen Laboratories Inc, Loveland, CO, USA) after heat fixation; samples marked B were stained without heat fixation; samples marked C were heat-fixed and dipped only in the counterstain (the blue reagent) of Dip Quick; and samples marked D were dipped only in the counterstain, without heat fixation. Ten high-power fields (hpf; X100 oil immersion objective) in each slide were evaluated by 2 observers, and total numbers of keratinocytes, yeast, bacteria, and neutrophils were counted. Statistical comparison was performed using an ANOVA model applied after verifying the normal distribution of the data, and using nonparametric sign tests and Wilcoxon signed rank tests. RESULTS: No statistically significant differences were observed in the numbers of keratinocytes, yeast, bacteria, or neutrophils among the 4 staining methods (P > .05), although significant interobserver differences were found. CONCLUSION: We conclude that heat fixation does not improve the quality of ceruminous ear swab samples for cytologic evaluation, and propose a 1-step dip in the blue reagent alone as a rapid method of staining samples from canine ear canals.  相似文献

Dysferlin (DYSF) is involved in the membrane-repair process, in the intracellular vesicle system and in T-tubule development in skeletal muscle. It interacts with mitsugumin 53, annexins, caveolin-3, AHNAK, affixin, S100A10, calpain-3, tubulin and dihydropyridine receptor. Limb-girdle muscular dystrophy 2B (LGMD2B) and Miyoshi myopathy (MM) are muscular dystrophies associated with recessively inherited mutations in the DYSF gene. The diseases are characterized by weakness and muscle atrophy that progress slowly and symmetrically in the proximal muscles of the limb girdles. LGMD2B and MM, which are collectively termed "dysferlinopathy", both lead to abnormalities in vesicle traffic and membrane repair at the plasma membrane in muscle fibers. SJL/J (SJL) and A/J mice are naturally occurring animal models for dysferlinopathy. Since there has been no an approach to therapy for dysferlinopathy, the immediate development of a therapeutic method for this genetic disorder is desirable. The murine models are useful in verification experiments for new therapies and they are valuable tools for identifying factors that accelerate dystrophic changes in skeletal muscle. It could be possible that the genetic or immunological background in SJL or A/J mice could modify muscle damage in experiments involving these models, because SJL and A/J mice show differences in the progress and prevalent sites of skeletal muscle lesions as well as in the gene-expression profiles of their skeletal muscle. In this review, we provide up-to-date information on the function of dysferlin, the development of possible therapies for muscle dystrophies (including dysferlinopathy) and the detection of new therapeutic targets for dysferlinopathy by means of experiments using animal models for dysferlinopathy.  相似文献

The objective was to evaluate effects of feeding level of milk replacer on body growth, plasma metabolite and insulin concentrations, and allometric growth of visceral organs in suckling calves. Holstein bull calves (n = 8; 3–4 days of age) were fed either a low amount (average 0.63 kgDM/day, LM) or high amount (average 1.15 kgDM/day, HM) of high protein milk replacer until they were slaughtered at 6 weeks of age. Body weight (BW) at 4, 5, and 6 weeks of age, feed intake, average daily gain, and feed efficiency were higher in the HM than LM calves. The HM group had higher plasma glucose at 3 and 4 weeks of age and insulin levels after the age of 4 weeks compared with LM calves whereas no effect was detected on plasma nonesterified fatty acid or urea nitrogen concentrations. The HM calves had greater empty body weight (EBW), viscera‐free BW and most of the organs dissected than LM calves. Relative weights (% of EBW) of liver, spleen, kidneys, and internal fat were higher, whereas head and large intestine was lower in HM than LM calves. The results suggest that increased milk feeding levels would accelerate the growth of the body and specific organs.  相似文献

Nitrate Vulnerable Zones (NVZ) were introduced in response to the Nitrates Directive (91/676/EEC) which states that all EU countries must reduce the nitrate in drinking water to a maximum of 50 mg/l. Farmers within a designated NVZ must adhere to strict rules over the timing and application of nitrogen from organic and inorganic sources. In Scotland, four NVZ regions were designated in 2003, covering around 14% of the land area and affecting over 12,000 farmers.This paper outlines the results of a recent study to understand farmer activities in response to and attitudes towards NVZ regulations in Scotland. A telephone survey was administered, obtaining a response of 184 farmers, supplemented by four workshops held in each NVZ region. This explored, both quantitatively and qualitatively, farmer behaviour and attitudes.The bulk of farmers have made little capital investment since the 2003 designation. Few farmers have invested in increased slurry storage facilities, nor begun to transport more slurry off-farm, claiming to have had enough storage capacity before designation to cover the imposed closed period.Farmer attitudes indicate a mostly negative view towards the perceived environmental benefits, water management and compliance. This can be explained by a number of concerns raised by farmers towards the scientific basis for designations. Furthermore, farmers viewed the restrictions placed on farming practices within NVZs as too inflexible.Given the mostly sceptical perceptions demonstrated by the farmers we call for a more integrated approach to water management at the catchment level. There is a role for policy-makers to provide clearer information over the science and purpose of the designations and also to invest in the transfer of technologies, in particular N-use software which is specifically tailored to NVZ regulations. Also, critical to this would be the development of transparent indicators of water quality. This may start to embed nitrogen pollution impacts within the farmer's cultural framework of decision-making.  相似文献

A dog with purulent vulval discharge was found to have many cysts containing purulent mucus in the uterine cervix. The uterine horns did not contain any pus. Histological examination revealed a cervical adenomyosis infiltrated by many neutrophils. After an ovariohysterectomy including the whole cervix, the dog recovered well.  相似文献

A novel PCR assay was developed in order to examine the prevalence of Haemobartonella felis (H. felis) in Japanese domestic cats and which was able to differentiate of the Ohio strain and the California strain of H. felis. Blood samples from a total of 21 cats suspected of having haemobartonellosis were examined employing a novel PCR assay and demonstrated positive results in 18 cats which was confirmed by cytological examination of blood smears. Four out of 18 positive cats (22%) were infected with the California strain, whilst the other 12 cats (67%) were infected with the Ohio strain and two animals (11%) were infected with both strains. As most of the cats with moderate to severe anemia were infected with the Ohio strain, it is suggested that the most prevalent strain of H. felis in Japanese domestic cats might be the Ohio strain. In the present study, it was thought that molecular detection and characterization of H. felis may provide valuable information regarding the severity and prognosis of this illness.  相似文献

The WS-Y isolate of Watermelon silver mottle virus (WSMoV) causes severe necrosis in Tetragonia expansa. To determine the RNA segment that induces symptoms, genome reassorants between WS-Y and an isolate causing mild mottle, WS-O, were generated. The origin of each segment in the reassortants was identified by RT-PCR and subsequent restriction enzyme analysis of the amplified fragments. Thirty genome reassortants were isolated from co-infected T. expansa plants. The reassortants with the S RNA segment of WS-Y caused severe necrosis, while those with the S RNA segment of WS-O caused a mild mottle; hence, the S RNA determined symptom expression. The incidence of reassortants was disproportional among genotypes. The most frequent genome reassortant possesses the L RNA of WS-Y, the M RNA of WS-O and the S RNA of WS-Y. A similar ratio of genotypes was found in isolates of local lesions on Chenopodium quinoa. These results strongly suggested that competition occurred independently between the individual RNA segments in a co-infected T. expansa plant, not between isolates.  相似文献

OBJECTIVES: The aims of this study were to evaluate the efficacy and tolerability of oral cephalexin given at 30 mg/kg once daily in dogs with superficial pyoderma and to compare them with those of oral cephalexin given at 15 mg/kg twice daily. METHODS: Twenty dogs with superficial pyoderma were treated with cephalexin at 30 to 60 mg/kg orally once daily (group A) and compared with 20 dogs treated at a dose of 15 to 30 mg/kg orally twice daily (group B). Dogs were treated until 14 days after clinical remission. Type and distribution of lesions, pruritus and general health status were assessed every 14 days using a numerical scale until 14 days after treatment discontinuation. Total scores for each evaluation day were compared between the two groups as well as time to obtain resolution and percentage of relapses. RESULTS: Resolution of superficial pyoderma was obtained in all dogs in 14 to 42 days (median 28 days for both groups), with no difference between groups. Six dogs experienced vomiting or diarrhoea but did not require discontinuation of the treatment. Only one dog (in group A) relapsed nine days after treatment discontinuation. CLINICAL SIGNIFICANCE: Once-daily cephalexin is as effective as twice-daily cephalexin in the treatment of canine superficial pyoderma.  相似文献
