Protein and RNA induction during the germination of Inubie (Echinochloa oryzicola) seeds pretreated with CaCN2 was studied. It was observed that the germination of Echinochloa oryzicola seeds was delayed after pretreatment with a high concentration of CaCN2 for 18 h. A considerable difference in the protein patterns was detected in the SDS-PAGE gel between the control (H2O) and CaCN2 treatment, particularly a 48 kilodalton (kD) protein band disappeared in CaCN2 treatment. At the tested three levels of CaCN2, the induction of this 48 kD protein was apparently inhibited during the germination process even when the duration of the pretreatment was as short as 2 h. When imbibed with water, the amount of the 48 kD protein increased rapidly within 1 h from the trace level in the dry seeds, and reached the maximum level after about 1–2 d. The results showed that the CaCN2 treatment also decreased the total RNA level in the germinating seeds. The relation of this protein induction and RNA decline with the beginning of Echinochloa oryzicola germination was discussed.  相似文献

To improve embryo development in bovine separated blastomeres, we evaluated applicability of co‐culture with intact embryos. The morphological quality of blastocysts derived from separated blastomeres and rate of blastocyst formation were only slightly increased when the cells were co‐cultured with intact embryos, which did not provide significant differences when statistically analyzed. However, the cell count of inner cell mass (ICM), trophectoderm (TE) and total number of cells in Day 8 blastocysts were significantly higher when the cells were co‐cultured with the intact embryos than those with the cells cultured individually (P < 0.05). Transfer of four monozygotic pairs of blastocysts derived from the cells co‐cultured with intact embryos led to three pregnancies even when the blastomeres were produced by in vitro maturation and in vitro fertilization of oocytes collected by ovum pick‐up from elite cows. These results suggest that co‐culturing with intact embryos may enhance development of bovine separated blastomere.  相似文献

The purpose of this study was to examine sequential changes in the immunologic parameters of perinatal mares and neonatal foals of the heavy draft horse. Blood samples were collected from clinically healthy pregnant mares and their newborn foals every week from 1 month before the expected foaling date, and 1 hour, 1 day (24-48 hours), and 1, 2, 3, and 4 weeks after foaling. Peripheral blood samples were used to examine total leukocyte counts (n = 20), differential leukocyte counts (n = 20), lymphocyte subpopulations (n = 13), lymphocyte responses to mitogens (n = 10), neutrophil phagocytic function (n = 12), and serum immunoglobulin G (IgG) concentrations (n = 10). In perinatal mares, remarkable changes observed included increased neutrophils, decreased lymphocytes, decreased CD4+ T lymphocytes, and decreased lymphocyte responses to mitogens at delivery. These changes were speculated to be the result of physical stress associated with delivery. In neonatal foals, increase in the phagocytic function of neutrophils, and increase in serum IgG concentration after suckling colostrum and increase of lymphocytes accompanied by physiologic growth were observed. Compared to dams, foals showed lower phagocytic function of neutrophils before suckling and fewer lymphocytes and lower lymphocyte responses to mitogens within 1 day after birth. This study revealed immunologic dynamics in perinatal mares and neonatal foals. Immunologic functions are suppressed in foaling mares and are immature in neonatal foals, especially before colostral intake. We expect these data will be useful for further studies in the field of clinical immunology, and preventive medicine.  相似文献

ABSTRACT:   The self-feeding rhythms of ayu Plecoglossus altivelis altivelis were examined. Individual ayu (mean body weight 40 g) were held in 60-L glass tanks equipped with self-feeders. Six of 14 fish learned self-feeding during the experiment. Under two different light–dark (LD) conditions (16 h:8 h and 8 h:16 h LD), self-feeding was synchronized to the LD cycle, and feeding occurred almost exclusively during the light phase. During exposure to constant light (LL), circadian feeding rhythms were observed. These results indicate self-feeding rhythms in ayu are restricted to the light phase under LD conditions and are controlled by the circadian clock under LL conditions.  相似文献

Drifting gill-nets were for a long time the main research gear for investigating the distribution of Pacific saury Cololabis saira. However, it has proven to be difficult to estimate the absolute abundance of saury using drifting gill-nets due to a lack of knowledge on the survey areas swept by these gill-nets. A mid-water trawl stock abundance estimation has the advantage of being able to estimate the absolute stock abundance before the commercial fishing season begins, whereas the conventional stock abundance estimation based on catch per unit efforts of commercial fisheries must be done after the commercial fishing season has ended. To confirm the practicality of a mid-water trawl to estimate the stock abundance of saury, we conducted a comparative survey in the northwestern Pacific Ocean using both a mid-water trawl and drifting gill-nets. We found that the geographic distribution pattern of catch per unit effort for both the mid-water trawl and driftnets was similar. Using the area-swept method and data on the fishing efficiency of the mid-water trawl obtained in a previous study, we were able to estimate the stock abundance plus confidence limits within the research areas. Our results suggest that the size composition of saury sampled by the mid-water trawl can be considered to approximate the actual size composition of saury.  相似文献

We compared the diet of Ommastrephes bartramii paralarvae with morphological changes in their beaks and proboscis (fused tentacles). The paralarvae were collected north of the Hawaiian Islands during 2001 and 2002 and ranged in mantle length (ML) from 1.1 to 13.2 mm. They fed on crustaceans, including copepods (copepodite stage) and amphipods. The rostral tips of upper and lower beaks began to protrude anteriorly at around 3–4 mm ML, and the smallest paralarva with identifiable prey in its digestive tract was 4.2 mm ML, which suggests that the paralarvae can masticate prey soon after the beaks protrude. The proboscis separated into two tentacles at 9.3–13.2 mm ML, but the newly formed tentacles were weakly developed even in the largest specimen, suggesting that tentacles do not operate functionally and that the arms are used to capture prey.  相似文献

Flatfishes display a left–right asymmetry that is unique in the animal kingdom. In order to clarify the mechanisms of the asymmetrical development of pigment cells, changes in pigment cell densities were examined in Japanese flounder Paralichthys olivaceus. During development from symmetrical larvae to asymmetrical juveniles, pigment cell densities were monitored on the skin on both the left side (ocular side in juvenile; eventually has two eyes) and the right side (blind side in juvenile; eventually has no eyes). A symmetrical and constant decrease was observed in leucophores and larval type melanophores. A mostly symmetrical (slightly delayed on the blind side) and constant increase in iridophores from metamorphosis was observed. Adult-type melanophores appeared and then increased only after metamorphosis on the ocular side. However, the pattern of xanthophores was complicated: they first existed symmetrically and decreased symmetrically until metamorphosis, and they later increased only on the ocular side. The dual appearance of the xanthophores, as well as the differences between their depths and sizes on the ocular and blind sides, may suggest the presence of two types of xanthophores—just as melanophores are well known to exhibit two types. The ontogenetic study of pigment cells described here is likely to help to elucidate the process of abnormal pigmentation in flatfishes.  相似文献

The physiological functions of pituitary gonadotropins (GtHs) are well established in higher vertebrates, whereas those in teleosts are still poorly understood. To describe the role of GtHs during gonadal development of female chub mackerel Scomber japonicus, changes in follicle-stimulating hormone (FSH) and luteinizing hormone (LH) cells were investigated immunohistochemically during the seasonal reproductive and spawning cycles. FSH and LH cells were identified in the different cell types of the proximal pars distalis (PPD); FSH cells were located in the central PPD, whereas LH cells were localized along the border of the pars intermedia. To examine changes in FSH and LH cells, the percentage of FSH or LH cell-occupying area in the PPD was evaluated and represented as FSHβ-immunoreactive (ir) or LHβ-ir levels, respectively. FSHβ-ir levels increased significantly from immature to the completion of vitellogenesis, whereas LHβ-ir levels were maintained at high levels from early vitellogenesis to post-spawning. During the spawning cycle, which consisted of four stages from just after spawning to the next oocyte maturation, both FSHβ-ir and LHβ-ir levels showed no significant changes among different stages; however, LHβ-ir levels remained relatively high, and FSHβ-ir levels were constantly low. These results suggest that both FSH and LH may be involved in vitellogenesis and LH may act at final oocyte maturation in female chub mackerel, although the role of FSH during the spawning cycle is still unclear.  相似文献

Pacific saury (Cololabis saira) has a short life span of 2 years and tends to exhibit marked population fluctuations. To examine the importance of sea surface temperature (SST) and mixed layer depth (MLD) as oceanographic factors for interannual variability of saury recruitment in early life history, we analyzed the relationship between abundance index (survey CPUE (catch per unit of effort)) of age‐1 fish and the oceanographic factors in the spawning and nursery grounds of the previous year when they were born, for the period of 1979–2006, in the central and western North Pacific. Applying the mixture of two linear regression models, the variability in the survey CPUE was positively correlated with previous year's winter SST in the Kuroshio Recirculation region (KR) throughout the survey period except 1994–2002. In contrast, the survey CPUE was positively correlated with the previous year's spring MLD (a proxy of spring chlorophyll a (Chl‐a) concentration) in the Kuroshio‐Oyashio Transition and Kuroshio Extension (TKE) during 1994–2002. This period is characterized by unusually deep spring MLD during 1994–1997 and anomalous climate conditions during 1998–2002. We suggest that saury recruitment variability was generally driven by the winter SST in the KR (winter spawning/nursery ground), or by the spring Chl‐a concentration (a proxy of prey for saury larvae) in the TKE (spring spawning/nursery ground). These oceanographic factors could be potentially useful to predict abundance trends of age‐1 saury in the future if the conditions leading to the switch between SST and MLD as the key input variable are elucidated further.  相似文献

This article describes the catalytic effect of NaHCO3 on condensation reactions of monomeric hydroxymethylphenols (HMPs) to elucidate the cure-acceleration mechanism. By comparison of the kinetics of self-condensations of HMPs, NaHCO3 was proved to increase the reactivity of para-hydroxymethyl groups. The changes of 13C nuclear magnetic resonance (NMR) chemical shifts on each HMP system with the additive indicated that the addition of NaHCO3 enhanced some molecular interactions between HMPs and NaHCO3, facilitating a resonance effect that might play a similar role in dissociation of the phenolic hydroxyl groups of HMPs. In addition, computational modeling by molecular orbital calculations elucidated that hydrogen carbonate anion (HCO 3 ) forms an interaction between either the para-hydroxymethyl group and the phenolic hydroxyl group or between the two para-hydroxymethyl groups of HMPs by hydrogen bonds. From the experimental results, the authors proposed the mechanism of the catalytic action of NaHCO3: it appears to be due to the delocalization of an electron initiated by the interaction of the para-hydroxymethyl groups and the phenolic hydroxyl of HMPs with HCO 3 through hydrogen bonds, which results in facilitating the formation of active species.  相似文献
