A chickpea F2 population of 593 plants derived from the intraspecific cross ILC3279 × WR315 was genotyped for markers closely linked to
quantitative trait loci (QTLs) for ascochyta blight resistance (QTLAR1 and QTLAR2 located on linkage group 4 and QTLAR3 on linkage group 2). All the markers located on linkage group 4 exhibited strongly distorted segregation with respect to
the expected Mendelian inheritance, towards the male parental line. This skewed segregation was also observed in a second
F2 population of 50 plants derived from the same cross, confirming the presence of a region of distorted segregation on this
linkage group and its heritability. The most skewed markers were SC-Y17 and TA72, which were tightly linked to each other,
indicating that they may both be closely associated with the genetic factor responsible for segregation distortion in chickpea.
To attempt to explain the non-Mendelian segregation, by identifying factors to which it could be attributed, three different
chi-square tests were carried out to test different hypotheses using the data obtained from examining co-dominant markers
associated with segregation distortion. According to our results, the distorted segregation could be caused by gametophytic
factors that affect either male or female gametes. Pollen fertility and meiosis were also analysed to determine their relationship
with segregation distortion; however, these not seem to be inducing factors in the non-Mendelian segregation reported in this

The assay of angiotensin-converting enzyme (ACE) inhibition by food-derived peptides is usually carried out by using soluble ACE in a batch process. The purification of this enzyme from tissues is not an easy task, and the resulting preparation loses activity very fast. In addition, ACE commercial preparations are very expensive. In this work the immobilization of ACE, through lysine amino groups, to 4% beads cross-linked (4 BCL) glyoxyl-agarose is described. The amount of immobilized enzyme increased with increasing concentrations of enzyme and with incubation time until a saturation point was reached at 50 mg protein/mL gel and 3.5 hours, respectively. The IC50 values for a noncompetitive sunflower peptide inhibitor were similar for the soluble (30.56 microM) and immobilized (32.7 microM) enzymes. An immobilized derivative was obtained that was 60 times more stable than the soluble enzyme at 60 degrees C. This procedure yields a derivative that can be reused and has increased thermal stability compared to that of the soluble enzyme. Thus, ACE immobilization is a good alternative to using soluble freshly prepared or commercial preparations because of economical and practical reasons. 相似文献

Reactions between lipid peroxidation products and proteins in foods have detrimental nutritional effects, most importantly, losses of essential amino acids. One of the major products of the reaction of malondialdehyde and alkanals with amino groups are 4-substituted 1,4-dihydropyridine-3,5-dicarbaldehyde derivatives. The product of the reaction of lysine with malondialdehyde and acetaldehyde, N,N'-di-(4-methyl-1,4-dihydropyridine-3,5-dicarbaldehyde)lysine (MDDL), has been synthesized and used for in vitro and in vivo bioavailability studies. Release of free lysine did not occur in incubations of MDDL with tissue homogenates. After oral administration of radioactively labeled MDDL, radioactivity was only recovered in feces. Radioactivity was not incorporated into hepatic microsomes after intraperitoneal administration, which would have indicated release of available lysine. These results show that MDDL is a form of unavailable lysine, because it is not metabolized to free lysine and cannot be absorbed from the gut. Thus, formation of this derivative in foods would result in loss of available lysine. 相似文献

Urea, ammonium, and free amino acid contents were quantified in a must from Vitis vinifera cv. Pedro Ximenez grapes and in fermented wine and after a short aging of this wine by Saccharomyces cerevisiae race capensis yeast under variable oxygen availability conditions. The previous compounds were also determined in a wine in which the nitrogen source was depleted by the same race of flor yeast (old wine) and also following the addition of ammonium ion, L-glutamic acid, and L-proline. Under specific conditions such as low oxygen level and the absence of some nutrients, the yeasts release some amino acids including L-threonine, L-tryptophan, L-cysteine, and L-methionine to the medium. These amino acids must originate primarily in a de novo synthesis from ethanol that regenerates NAD(P)+. On the basis of these results, the yeasts may be able to use amino acids not only as nitrogen sources but also as redox agents to balance the oxidation-reduction potential under conditions of restricted oxygen, when electron transport along the respiratory chain may be hindered or limited. 相似文献

Serum samples from 282 wild carnivores from different regions of Spain were tested for antibodies to Toxoplasma gondii by the modified agglutination test using a cut-off value of 1:25. Antibodies to T. gondii were found in 22 of 27 (81.5%) of Iberian lynx (Lynx pardinus), 3 of 6 European wildcats (Felis silvestris), 66 of 102 (64.7%) red foxes (Vulpes vulpes), 15 of 32 (46.9%) wolves (Canis lupus), 26 of 37 (70.3%) Eurasian badgers (Meles meles), 17 of 20 (85.0%) stone martens (Martes foina), 4 of 4 pine martens (Martes martes), 6 of 6 Eurasian otters (Lutra lutra), 4 of 4 polecats (Mustela putorius), 1 of 1 ferret (Mustela putorius furo), 13 of 21 (61.9%) European genets (Genetta genetta), and 13 of 22 (59.1%) Egyptian mongooses (Herpestes ichneumon). Serological results indicated a widespread exposure to T. gondii among wild carnivores in Spain. The high T. gondii seroprevalence in Iberian lynx and the European wildcat reported here may be of epidemiologic significance because seropositive cats might have shed oocysts. 相似文献

Since 1990 the Institute for Ecology of Industrial Areas, acting as National Focal Center, is actively involved in an international research programme aimed at the calculation and mapping of critical loads of acidifying compounds. Following the methodological guidelines elaborated under the leadership of UN/ECE Task Force on Mapping and Coordination Center for Effects, national maps of critical loads and their exceedances for acidity, sulphur and nitrogen have been produced. These maps have already been utilized in derivation of European maps of critical loads of acidity and sulphur submitted to the UN/ECE LRTAP Convention as scientific input to the negotiations on the Second Sulphur Protocol. The lessons learned from the critical loads mapping exercise can be summarized as follow:

CD10 is an important cell marker in the diagnosis of acute lymphoblastic leukaemia and of breast myoepithelial (ME) cells in humans. The objective of this study was to assess the value of CD10 as a marker of canine ME cells using immunohistochemistry on routinely processed normal, dysplastic and neoplastic mammary tissue. Five different CD10 positive cell types were identified on the basis of cell morphology, pattern of immunoreactivity, and on the co-expression of additional cell lineage-specific markers.Type 1 cells were typical fusiform cells with a ME cell phenotype (calponin- and cytokeratin [CK] 14-positive, CK8/18-negative). Type 2 cells were typical or atypical polyhedral cells with a luminal epithelial (LE) cell phenotype (calponin- and CK14-negative, CK8/18-positive). Type 3 cells had a type 1 phenotype with variable morphology, and type 4 were atypical neoplastic cells with a mixed ME/LE phenotype. Type 5 cells were typical fusiform cells with a stromal phenotype.Type 1 cells were considered normal ME cells and were found in all sample types; type 2 cells were considered normal or neoplastic LE cells and were also found in all sample types; types 3 and 4 cells were restricted to tumour samples and to malignant tumours, respectively, and type 5 cells were found in all sample types, although predominantly in neoplastic tissue. The findings indicate that the CD10 antigen is a sensitive (although not specific) marker of canine ME cells in normal, dysplastic and neoplastic mammary tissue. Differences in the distribution and staining intensity of CD10-positive cells suggest a number of potential roles for this protein in the pathogenesis of canine mammary neoplasia. 相似文献

The grey squirrel is an invasive pest species in the United Kingdom and populations are controlled to protect timber crops and native fauna. Although animals are culled using a variety of methods, there are few data available on the relative efficiency of different trap designs, and in particular spring traps. We compared grey squirrel captures in Magnum 116 spring-traps set within Fineren boxes against captures in a single entry live capture trap design. Trapping was conducted in mature mixed woodland in North Wales. No significant difference in the frequency of grey squirrel captures or body mass of trapped individuals between trap designs was observed. Despite the Fineren box design limiting access by non-target species, by-catch was recorded. We make recommendations for future research to minimise this and also compare our results for Magnum 116 traps with data available on captures in Fenn IV spring traps. The findings have broad relevance to the control of grey squirrels across their wider international invasive range. 相似文献

Gliomatosis cerebri (GC) is a rare, diffusely infiltrating, glial cell tumour of neuroepithelial origin. This report describes a case of oligodendroglial GC in a 6-year-old male Poodle with central nervous system symptoms. Computed tomography revealed anomalous parenchyma density and ventricular asymmetry. Cerebrospinal fluid showed elevated protein (30 mg dL(-1)) and nucleated cell count (20 μL(-1)). Presumptive diagnosis of necrotizing meningoencephalitis was made. Because of rapid deterioration of the general condition of the animal, the dog was euthanized. Histologically there was an infiltration of round or ovoid neoplastic cells in the white matter of the left cerebral hemisphere and in leptomeninges. Immunohistochemistry showed that 80% of the neoplastic cells expressed Olig2 and some 50% expressed glial fibrilary acidic protein. On the basis of clinical, histological and immunohistochemical features, a diagnosis of oligodendoglial GC was done. This case represents the first report of a case of oligodendroglial GC in the canid. 相似文献
