A high incidence (about 70%) of alteration in endometrial epidermal growth factor
(EGF) profile, i.e., loss of 2 peaks on days 2–4 and 13–14, has been linked to a
reduced fertility in multiparous repeat breeder Holstein cows. However, the EGF
profile in Holstein heifers and other breeds (types) of cattle has not been
investigated. In study 1, EGF concentrations were determined using endometrial
tissues obtained by biopsy on days 3, 7 and 14 from 84 fertile Holstein heifers to
obtain a normal range and 53 repeat breeder Holstein heifers to estimate incidence of
alterations in the EGF profile. In repeat breeder heifers, EGF concentrations were
similar to fertile controls on 3 days and five animals (9.4%) had an altered EGF
profile with EGF concentrations below the normal range on days 3 and 14. In study 2,
EGF concentrations on day 3 were repeatedly examined from the nulliparous period to
the third postpartum period in 28 Holstein (dairy) and 47 Japanese Black (beef)
cattle. The effect of parity on EGF concentrations on day 3 was different between
Holstein and Japanese Black cattle. In Japanese Black cows, the EGF concentrations
were consistently high throughout the study period, while in Holstein cows, the EGF
concentrations decreased after the second calving. In conclusion, unlike multiparous
repeat breeder Holstein cows, an altered EGF profile may not be a major cause of
repeat breeding in Holstein heifers, and the peak EGF concentrations around day 3 may
decrease even in fertile populations of multiparous dairy cows, but not in beef

This study investigated the effects of skim milk on the quality and fertility of boar spermatozoa under long‐term chilled preservation. Semen samples were stored in Modena solution supplemented with 0 (control) to 50 mg/mL skim milk at 5°C for 4 weeks; spermatozoa stored with 7.5 and 15 mg/mL of skim milk (7.5‐SM and 15‐SM groups, respectively) exhibited significantly higher motility indices than those of the control group up to 3 weeks (P < 0.05), and the 7.5‐SM group showed improved motility indices even after 4 weeks (P < 0.05). In vitro fertilization using spermatozoa in the 7.5‐SM and 15‐SM groups stored at 5°C for 2 weeks showed significantly higher fertilization rates of spermatozoa and the development rates to blastocyst than the control group (P < 0.05), and the 7.5‐SM group showed similar rates of fertilization and blastocyst formation in the fresh non‐stored spermatozoa group. After artificial insemination using spermatozoa stored for 2 weeks in the 7.5‐SM group, healthy piglets were obtained. Boar spermatozoa can be stored at 5°C in a Modena solution containing skim milk. Supplementation of 7.5 mg/mL skim milk improves boar spermatozoa motility and fertility even after liquid preservation at 5°C for 2 weeks. 相似文献

To improve embryo development in bovine separated blastomeres, we evaluated applicability of co‐culture with intact embryos. The morphological quality of blastocysts derived from separated blastomeres and rate of blastocyst formation were only slightly increased when the cells were co‐cultured with intact embryos, which did not provide significant differences when statistically analyzed. However, the cell count of inner cell mass (ICM), trophectoderm (TE) and total number of cells in Day 8 blastocysts were significantly higher when the cells were co‐cultured with the intact embryos than those with the cells cultured individually (P < 0.05). Transfer of four monozygotic pairs of blastocysts derived from the cells co‐cultured with intact embryos led to three pregnancies even when the blastomeres were produced by in vitro maturation and in vitro fertilization of oocytes collected by ovum pick‐up from elite cows. These results suggest that co‐culturing with intact embryos may enhance development of bovine separated blastomere. 相似文献

Bordetella bronchiseptica (B. bronchiseptica) is associated with respiratory tract infections in laboratory animals. In our laboratory animal facility, B. bronchiseptica was isolated from 21 of 27 apparently healthy rabbits obtained from a breeding farm contaminated with B. bronchiseptica. Restriction fragment length polymorphism (RFLP) analysis showed that the flagellin genotype of isolates from the laboratory animal facility and breeding farm was type A, which is seen relatively frequently in rabbits in Europe. To examine its pathogenicity, guinea pigs, rats, and mice were inoculated intranasally with a representative strain isolated in the laboratory animal facility. Following inoculation of 107 colony forming unit (cfu), severe inflammation was observed in the lungs of guinea pig and mice, although the inflammation was less severe in rats. The strain was recovered from the trachea and lungs of these species after inoculation with lower dose such as 103 or 104 cfu. These results suggest that the isolated strain causes respiratory tract infection in guinea pigs, rats, and mice, and that its pathogenicity higher in mice than in rats. This study extends our knowledge of interpreting the microbiologic status of laboratory animals, which will contribute to the development of reliable and reproducible animal experiments. 相似文献

The fluctuations in the plasma levels of several bone markers were investigated in newborn calves. Experiment
1 monitored the postnatal changes in the plasma levels of tartrate-resistant acid phosphatase isoform 5b
(TRAP5b), total alkaline phosphatase (t-ALP) and bone-specific alkaline phosphatase (BAP) in four calves.
These markers increased significantly from 9−20 hr after the first colostrum-suckling compared with the values
immediately after birth. Experiment 2 evaluated changes in the plasma TRAP5b, t-ALP, BAP and type I collagen
cross-linked N-telopeptide (NTx) levels within 2 days post-birth in five calves with successful passive
immunization via colostrum (non-deficient group) and five others with poor colostrum intake (deficient group).
The non-deficient group had significantly higher plasma levels of the four parameters around 12 hr of life
compared with the deficient group. The results suggest that the increase in plasma bone markers in calves in

通过实验检验了克隆植物结缕草在异质生境条件下的生理整合作用。结缕草被栽植于4种类型的“生境”内(它们的总体土壤氮素资源水平呈现梯度变化),并同时对匍匐茎的节间实施连接与切断2种处理。实验结果表明,呈连接状态的结缕草克隆的平均复合节数量、平均匍匐茎长度和生物量在各个生境类型间并未表现出明显的差异。但主匍匐茎实施切断处理后的结缕草克隆的上述各项指标均显著减小。生长于贫瘠土壤斑块上的复合节平均根系生物量和根/茎比均高于肥沃土壤斑块。这一特征与其它典型的克隆植物明显不同。结缕草克隆在保持连接的情况下,土壤氮素对节间长度和比节间长度的伸长生长以及分蘖的形成具有促进作用。实验过程中的节间切断处理抑制了节间的进一步伸长但提高了比节间长度。保持结缕草克隆整合并尽量避免严重干扰显得十分重要,特别是在结缕草早期生长阶段。 相似文献

Recently, we reported a method for discriminating a Japanese brand of chicken, the Hinai-jidori. As an application of this method for discriminating Hinai-jidori eggs, we here report an efficient method for extracting maternal DNA from eggshells. Eggshell powder was completely decalcified with EDTA solution, and then DNA was isolated by conventional phenol-chloroform extraction and ethanol precipitation. The efficiency of DNA recovery from eggshells was 50-fold higher than that of a previously reported method. The recovered DNA could be used for PCR, and 10 markers for identifying the Hinai-jidori chicken were detected. The genotypes of the Hinai-jidori exactly matched those of the Hinai-dori breed. Using this method, Hinai-jidori and Hinai-dori eggs could be distinguished from the eggs of Rhode Island Reds. This is the first report of a technique that can be used to discriminate the eggs of Hinai-jidori from those of other chickens, and it can also be utilized to validate the labeling of Hinai-jidori eggs in the market. 相似文献
