The characterization of herbal materials is a significant challenge to analytical chemists. Goldenseal (Hydrastis canadensis L.), which has been chosen for toxicity evaluation by NIEHS, is among the top 15 herbal supplements currently on the market and contains a complex mixture of indigenous components ranging from carbohydrates and amino acids to isoquinoline alkaloids. One key component of herbal supplement production is botanical authentication, which is also recommended prior to initiation of efficacy or toxicological studies. To evaluate material available to consumers, goldenseal root powder was obtained from three commercial suppliers and a strategy was developed for characterization and comparison that included Soxhlet extraction, HPLC, GC-MS, and LC-MS analyses. HPLC was used to determine the weight percentages of the goldenseal alkaloids berberine, hydrastine, and canadine in the various extract residues. Palmatine, an isoquinoline alkaloid native to Coptis spp. and other common goldenseal adulterants, was also quantitated using HPLC. GC-MS was used to identify non-alkaloid constituents in goldenseal root powder, whereas LC-MS was used to identify alkaloid components. After review of the characterization data, it was determined that alkaloid content was the best biomarker for goldenseal. A 20-min ambient extraction method for the determination of alkaloid content was also developed and used to analyze the commercial material. All three lots of purchased material contained goldenseal alkaloids hydrastinine, berberastine, tetrahydroberberastine, canadaline, berberine, hydrastine, and canadine. Material from a single supplier also contained palmatine, coptisine, and jatrorrhizine, thus indicating that the material was not pure goldenseal. Comparative data for three commercial sources of goldenseal root powder are presented.  相似文献

A number of different beverage products claim to have antioxidant potency due to their perceived high content of polyphenols. Basic and applied research indicates that pomegranate juice (PJ), produced from the Wonderful variety of Punica granatum fruits, has strong antioxidant activity and related health benefits. Although consumers are familiar with the concept of free radicals and antioxidants, they are often misled by claims of superior antioxidant activity of different beverages, which are usually based only on testing of a limited spectrum of antioxidant activities. There is no available direct comparison of PJ's antioxidant activity to those of other widely available polyphenol-rich beverage products using a comprehensive variety of antioxidant tests. The present study applied (1) four tests of antioxidant potency [Trolox equivalent antioxidant capacity (TEAC), total oxygen radical absorbance capacity (ORAC), free radical scavenging capacity by 2,2-diphenyl-1-picrylhydrazyl (DPPH), and ferric reducing antioxidant power (FRAP)]; (2) a test of antioxidant functionality, that is, inhibition of low-density lipoprotein (LDL) oxidation by peroxides and malondialdehyde methods; and (3) evaluation of the total polyphenol content [by gallic acid equivalents (GAEs)] of polyphenol-rich beverages in the marketplace. The beverages included several different brands as follows: apple juice (3), a?aí juice (3), black cherry juice (3), blueberry juice (3), cranberry juice (3), Concord grape juice (3), orange juice (3), red wines (3), iced tea beverages (10) [black tea (3), green tea (4), white tea (3)], and a major PJ available in the U.S. market. An overall antioxidant potency composite index was calculated by assigning each test equal weight. PJ had the greatest antioxidant potency composite index among the beverages tested and was at least 20% greater than any of the other beverages tested. Antioxidant potency, ability to inhibit LDL oxidation, and total polyphenol content were consistent in classifying the antioxidant capacity of the polyphenol-rich beverages in the following order: PJ>red wine>Concord grape juice>blueberry juice>black cherry juice, a?aí juice, cranberry juice>orange juice, iced tea beverages, apple juice. Although in vitro antioxidant potency does not prove in vivo biological activity, there is also consistent clinical evidence of antioxidant potency for the most potent beverages including both PJ and red wine.  相似文献

Development of robust analytical procedures is critical when using hyperspectral imaging technology in food technology and agriculture. This study used near-isogenic inbred corn lines to address two basic questions: (1) To what extent is classification accuracy increased by grinding maize kernels? (2) Can the classification accuracy of two near-isogenic inbred lines be increased by using a spectral filter to classify only certain hyperspectral profiles from each image cube? Whole kernels and ground kernels in two particle intervals, 0.250-0.354 mm (size 1) and 0.354-0.841 mm (size 2), were examined. Spectral profiles acquired from ground kernels had higher spectral repeatability than data collected from whole kernels. The classification error of discriminant functions from whole kernels was >3 times lower than that of size 1 ground particles. Applying a spectral filter to input data had negligible effect on classifications of hyperspectral profiles from whole kernels and size 2 ground particles, but for size 1 ground particles a considerable increase in accuracy was observed. Independent validation confirmed that distinction between wild type and mutant inbred maize lines could be conducted with >80% accuracy after the proposed spectral filter had been applied to hyperspectral profiles of size 1 ground particles. A combination of discriminant analysis and regression analysis could be used to accurately predict mixture ratios of the two inbred lines. The use of spectral filtering to increase the level of spectral repeatability and the use of hyperspectral imaging technology in large-scale commercial operations are discussed.  相似文献

The disease known as pitch canker results from infection of Pinus species by the fungus Fusarium circinatum. This fungus also causes a serious root disease of Pinus seedlings and cuttings in forestry nurseries. Pinus radiata and P. patula are especially susceptible to the pathogen, but there are no records of pitch canker on P. patula in established plantations. To date, only planting material of this tree species in nurseries or in plantations at the time of establishment have been infected by F. circinatum. Symptoms of pitch canker have recently emerged in an established P. patula plantation in South Africa and this study sought to determine whether the symptoms were caused by F. circinatum. Isolates from cankers were identified as F. circinatum using morphology and DNA-based diagnostic markers. Microsatellite markers were then used to determine the genetic diversity of a collection of 52 isolates. The entire population included 17 genotypes representing 30 alleles, with a greater number of genotypes collected from younger (three- to six-year-old) than older (12- to 19-year-old) trees. Both mating types of F. circinatum were present, but no evidence of sexual recombination was inferred from population genetic analyses. This is the first record globally of pitch canker on P. patula trees in managed plantations. It is of significant concern to South Africa, where P. patula is the most important Pinus species utilised for plantation forestry.  相似文献

The long-term nature of forest crop rotations makes it difficult to determine impacts of forestry on soil nutrients that might be depleted by forest growth. We used small scale, highly stocked plots to compress the length of the rotation and rapidly induce nutrient depletion. In the study, two species (Pinus radiata D. Don and Cupressus lusitanica Miller) are compared under two disturbance regimes (soil undisturbed and compacted), and two fertiliser treatments (nil and plus fertiliser), applied in factorial combination at 33 sites, covering the range of climatic and edaphic variation found in plantation forests across New Zealand. To assess our ability to rapidly highlight important soil properties, foliar nutrient concentrations were determined 20 months after planting. It was hypothesised that the densely planted plots, even at a young age, would create sufficient pressure on nutrient resources to allow development of relationships between properties used as indicies of soil nutrient availability and foliar nutrient concentrations. For both species significant relationships between foliar nutrients and 0–10 cm layer soil properties from unfertilised plots were evident for N (total and mineralisable N) and P (total, acid extractable, organic, Bray-2 and Olsen P). With the exception of Ca in C. lusitanica, foliar K, Ca and Mg were correlated with their respective soil exchangeable cation measures. The results thus confirm the utility of the experimental approach and the relevance of the measured soil properties for forest productivity.

In unfertilised plots foliar N and P concentrations in P. radiata exceeded those in C. lusitanica, the differences being eliminated by fertiliser application. Foliar N/P ratios in P. radiata also exceeded those in C. lusitanica. In contrast to N and P, foliar K, Ca and Mg concentrations were all higher in C. lusitanica, the difference being particularly marked for Ca and Mg. P. radiata contained substantially higher concentrations of the metals Zn, Mn and Al than C. lusitanica, whereas the latter contained higher B concentrations. Possible reasons for differences between species in foliar nutrient concentrations are discussed.  相似文献

Earthworm activity is beneficial in most natural and agricultural systems, but excessive earthworm casting is a problem on sports fields worldwide. Expulsion of soil‐rich earthworm fecal matter, or casts, as mounds of soil on the turf canopy can muddy the surface, reduce photosynthesis, and lead to thinning, weed invasion and surface softening. Casts affect ball roll on sports fields, cause fouling of maintenance equipment, and dull mower blades. Build‐up of cast material on reel mower units can affect height and quality of cut. Casting activity is dependent on environmental conditions such as soil texture, temperature, moisture, pH, and organic matter. Response to environmental conditions varies by species. Management options are limited, because no pesticides are specifically labeled for earthworm control at this time, and cultural control methods such as soil modification, turfgrass clipping removal, and sand topdressing have limited and inconsistent efficacy. Products containing plant‐derived saponins and irritants show promise for earthworm management. Pest management practices to mitigate excessive earthworm casting will likely need to be species‐specific, but limited knowledge of earthworm identification by end‐users further inhibits the efficacy of control measures. © 2019 Society of Chemical Industry  相似文献
