Blooms of toxic cyanobacteria are becoming increasingly frequent, mainly due to water quality degradation. This work applied qPCR as a tool for early warning of microcystin(MC)-producer cyanobacteria and risk assessment of water supplies. Specific marker genes for cyanobacteria, Microcystis and MC-producing Microcystis, were quantified to determine the genotypic composition of the natural Microcystis population. Correlations between limnological parameters, pH, water temperature, dissolved oxygen and conductivity and MC concentrations as well as Microcystis abundance were assessed. A negative significant correlation was observed between toxic (with mcy genes) to non-toxic (without mcy genes) genotypes ratio and the overall Microcystis density. The highest proportions of toxic Microcystis genotypes were found 4-6 weeks before and 8-10 weeks after the peak of the bloom, with the lowest being observed at its peak. These results suggest positive selection of non-toxic genotypes under favorable environmental growth conditions. Significant positive correlations could be found between quantity of toxic genotypes and MC concentration, suggesting that the method applied can be useful to predict potential MC toxicity risk. No significant correlation was found between the limnological parameters measured and MC concentrations or toxic genotypes proportions indicating that other abiotic and biotic factors should be governing MC production and toxic genotypes dynamics. The qPCR method here applied is useful to rapidly estimate the potential toxicity of environmental samples and so, it may contribute to the more efficient management of water use in eutrophic systems. 相似文献

Computed tomography (CT) is commonly used in veterinary practice to evaluate dogs with suspected brain disease, however contrast resolution limitations and artifacts may reduce visualization of clinically important anatomic features. The purpose of this study was to develop an optimized CT protocol for evaluating the canine brain. The head of a 5‐year‐old Springer Spaniel with no neurological signs was imaged immediately following euthanasia using a 4‐slice CT scanner and 282 protocols. Each protocol used a fixed tube voltage of 120 kVp and 10 cm display field of view. Other acquisition and reconstruction parameters were varied. For each protocol, four selected images of the brain were reconstructed, anonymized and saved in DICOM format. Three board‐certified veterinary radiologists independently reviewed each of the four images for each protocol and recorded a numerical quality score for each image. The protocol yielding the lowest total numerical score was defined as the optimal protocol. There was overall agreement that the optimal protocol was the one with the following parameters: sequential mode, 300 mAs, 1 mm slice thickness, 1 s tube rotation time, medium image reconstruction algorithm and applied beam hardening correction. Sequential imaging provided optimal image resolution. The thin‐sliced images provided a small blur due to partial volume artifacts. A high tube current resulted in a relatively low noise level. Use of a medium frequency image reconstruction algorithm provided optimal contrast resolution for brain tissue. Use of a proprietary beam hardening correction filter (Posterior Fossa Optimization) markedly reduced beam‐hardening artifact. 相似文献

Cooking as a domestic processing method has a great impact on food nutrients. Most Brassica (Brassicaceae, Cruciferae) vegetables are mainly consumed after being cooked, and cooking considerably affects their health-promoting compounds (specifically, glucosinolates, phenolic compunds, minerals, and vitamin C studied here). The microwave cooking process presents controversial results in the literature due to the different conditions that are employed (time, power, and added water). Therefore, the aim of this work was to study the influence of these conditions during microwave cooking on the human bioactive compounds of broccoli. The results show a general decrease in the levels of all the studied compounds except for mineral nutrients which were stable under all cooking conditions. Vitamin C showed the greatest losses mainly because of degradation and leaching, whereas losses for phenolic compounds and glucosinolates were mainly due to leaching into water. In general, the longest microwave cooking time and the higher volume of cooking water should be avoided to minimize losses of nutrients. 相似文献

A fast method to estimate the loss of corn quality for wet milling as a result of excessive thermal treatment during drying is proposed. Proteins that remain soluble after drying were quantitatively extracted with a solution of phosphoric acid and ethanol. The soluble proteins were colorimetrically quantified using Coomasie Brillant Blue G dye and measuring at 595 nm. A decrease in absorbance values indicates a reduction in commercial quality of the corn. The application time of the proposed technique is ≈30 min. 相似文献
