BACKGROUND: Bait-formulated spinosad is currently being introduced for housefly (Musca domestica L.) control around the world. Spinosad resistance was evaluated in a multiresistant field population and strains derived from this by selection with insecticides. Constitutive and spinosad-induced expression levels of three cytochrome P450 genes, CYP6A1, CYP6D1 and CYP6D3, previously reported to be involved in insecticide resistance, were examined. RESULTS: In 2004 a baseline for spinosad toxicity of Danish houseflies where all field populations were considered to be susceptible was established. In the present study, females of a multiresistant field population 791a were, however, 27-fold spinosad resistant at LC50, whereas 791a male houseflies were susceptible. Strain 791a was selected with spinosad, thiamethoxam, fipronil and imidacloprid, resulting in four strains with individual characteristics. Selection of 791a with spinosad did not alter spinosad resistance in either males or females, but counterselected against resistance to the insecticides thiamethoxam and imidacloprid targeting nicotinic acetylcholine receptors. A synergist study with piperonyl butoxide, as well as gene expression studies of CYP6A1, CYP6D1 and CYP6D3, indicated a partial involvement of cytochrome P450 genes in spinosad resistance. CONCLUSION: This study reports female-linked spinosad resistance in Danish houseflies. Negative cross-resistance was observed between spinosad and neonicotinoids in one multiresistant housefly strain. Spinosad resistance involved alterations of cytochrome P450 gene expression. A control diet without supplemental fat and four diets containing 8% of coconut oil, rapeseed oil, fish oil or sunflower oil were fed to lactating sows in order to investigate the lipid absorption capacity of their progeny in terms of pancreatic enzyme activity, hormonal regulation, and bile salt concentration. In addition, the effect of age was studied during the four week suckling period, and during a three week postweaning period, in which a standard weaner diet was fed to all piglets. It is concluded that fat source and level in the maternal diet plays a minor role in comparison to piglet age with regard to lipid absorption capacity of the progeny postweaning. 相似文献

Comparisons were made of Cortisol concentration and behavioural activity between piglets weaned at 3 weeks of age (early weaned piglets), and piglets weaned at 8 weeks of age (control piglets). Early weaned littermates were placed in cages with a floor area of either 0.20 m2 per piglet or 0.15 m2 per piglet. At 8 weeks of age, the piglets in the control litters were moved from the sow and early weaned piglets were moved from the cages. All litters were placed in pens, 1 pen for each litter. Radioimmunoassay was used for plasma Cortisol determination. Behavioural activity included determination of play, aggressive behaviour and non-nutritive oral activity. Statistical analysis for possible association between plasma Cortisol concentration and age, housing conditions, litter and sequence of capture of littermates for blood collection revealed a complex pattern. Generally, higher plasma Cortisol concentrations and more aggressive and non-nutritive oral activity, but less playing activity, were associated with early weaning. Plasma Cortisol concentration in individual piglets was positively correlated to both aggressive and non-nutritive oral behaviour, but was negatively correlated to playing behaviour. Pythium oligandrum Drechsler is a mycoparasite which parasitizes hyphae of many fungal species. A detailed study of the interactions between P. oligandrum and the sclerotia of the plant pathogen Sclerotinia sclerotiorum (Lib.) de Bary is presented. Pythium oligandrum was present in Danish soils at concentrations between 4 and 26 cfu g–1 soil. An increase in the natural population of P. oligandrum by addition of P. oligandrum zoospores to a soil reduced the ability of sclerotia of S. sclerotiorum to germinate myceliogenically and the sclerotia were colonized internally by P. oligandrum. This colonization and reduction of germination of sclerotia were also seen when sclerotia and P. oligandrum were incubated together in water. Small sclerotia were significantly more susceptible to parasitism by P. oligandrum than large sclerotia, and increasing the incubation time caused a further reduction in the germination ability of the sclerotia. P. oligandrum was able to pass through its entire life-cycle from zoospores to oogonia both with sclerotia as sole nutrient-source and in water containing exudates from the sclerotia. The cell wall degrading enzymes N-acetyl--D-glucosaminidase (NAGase), endo-chitinase, protease, -glucanase, -glucosidase and cellobiohydrolase were detected in culture filtrates of P. oligandrum cultivated with S. sclerotiorum. These findings suggest that P. oligandrum has a potential to reduce the survival of S. sclerotiorum sclerotia present naturally in soils, through mycoparasitic activity. 相似文献

The ability of rapeseed oil and/or different antioxidants (alpha- and gamma-tocopherol mixture, ascorbyl palmitate, and EDTA) to protect fish-oil-enriched milk emulsions against oxidation was investigated. Tocopherol isomers in concentrations similar to those found in natural rapeseed oil were added to rapeseed oil stripped of natural tocopherols. The rapeseed oil with added tocopherols significantly inhibited oxidation in the fish-oil-enriched milk emulsions. In contrast, the emulsions with only fish oil and added alpha- and gamma-tocopherol were less stable than the emulsions with fish oil alone. When added individually, the gamma-tocopherol seemed to inhibit oxidation more efficiently than alpha-tocopherol. Ascorbyl palmitate (AP) almost completely retarded oxidation in the fish-oil-enriched milk emulsions, as determined by PV, volatile oxidation products, and sensory evaluation. AP also prevented the otherwise prooxidant effect of tocopherols added to fish oil before emulsification. No interactions between AP, tocopherols, and EDTA were observed, and EDTA added alone to fish oil did not show antioxidant properties in the milk emulsions. Overall, the results showed that addition of AP or rapeseed oil containing natural tocopherols to fish oil was equally efficient in inhibiting oxidation in the fish-oil-enriched milk emulsions. 相似文献

More efficient use of green manure-derived nitrogen (N) may improve crop yields and reduce environmental impacts in stockless organic arable farming. In this 3-month incubation study, we tested a new strategy where green manure leys are harvested and preserved until the following spring either as compost mixed with straw or as silage of harvested ley biomass. Grass-clover compost or silage was soil-incorporated by either simulated ploughing (green manure placed at 15 cm depth) or harrowing (green manure mixed into the upper 5-cm soil horizon) in order to assess treatment effects on net release of plant-available N, nitrous oxide (N2O) fluxes and soil respiration. Grass-clover silage provided the highest net N release with similar results for the two incorporation methods. Up to one third of the total N content in silage became plant-available during the 3 months. In contrast, no net N release was observed for the composted grass-clover and straw mixture. In fact, soil incorporation of compost by harrowing caused temporal immobilization of soil mineral N. Silage incorporated by ploughing gave rise to the largest N2O effluxes with silage-induced emissions corresponding to 0.3 % of applied total N. Possibly N2O production via denitrification was stimulated by oxygen-limited conditions near the decomposing silage. In contrast, compost incorporated by harrowing caused net N2O uptake, presumably an effect of reduced mineral N availability in this treatment. Overall, our study revealed that ensiled grass-clover was the best fertilizer product and that the method chosen for incorporation of green manure is likely to influence N2O emissions. 相似文献

The toxicity of fipronil to insecticide-susceptible houseflies and the cross-resistance potential of fipronil were determined for six insecticide-resistant laboratory housefly strains by topical application and feeding bioassay. The insecticide-resistant strains represented different levels and patterns of resistance to pyrethroids, organophosphates, carbamates and organochlorines. Five strains were almost susceptible to fipronil in feeding bioassay with resistance factors at LC50 between 0.36 and 3.0. Four of these strains were almost susceptible to topically applied fipronil (resistance factors at LD50 were 0.55, 0.83, 3.3 and 2.5, respectively), whereas one strain was 13-fold resistant to topically applied fipronil. A highly gamma-HCH-resistant strain, 17e, was 430-fold resistant to fipronil in topical application bioassay and 23-fold resistant in feeding bioassay at LD50/LC50. We also tested the toxicity of fipronil in a feeding bioassay and gamma-HCH in topical application bioassay on thirteen housefly field populations. Eleven of the field populations had resistance factors for fipronil ranging from 0.98 to 2.4 at LC50, whereas two populations were 4.0- and 4.6-fold resistant to fipronil. The resistance level to gamma-HCH at LD50 in the field populations ranged from 1.8- to 8.1-fold. The two strains showing fipronil resistance were 3.4- and 8.1-fold resistant to gamma-HCH. Fipronil and gamma-HCH toxicities were positively correlated in the field populations. Biochemical assays of esterase, glutathione S-transferase and cytochrome P450 monooxygenase indicated that the low fipronil resistance observed in laboratory and field strains could be caused by elevated detoxification or be due to a target-site resistance mechanism with cross-resistance to gamma-HCH. 相似文献

Light-induced oxidation of semihard cheese has been evaluated by fluorescence spectroscopy. The cheese was packaged in two packaging materials and exposed to different storage conditions, which included light/dark storage, oxygen availability, and storage time (0, 4, 7, 14, 21, 42, 70, or 84 days). Fluorescence excitation-emission matrices (EEM) were analyzed by PARAFAC, which gave an estimation of the pure excitation and emission spectra of the fluorophores and the concentrations of these. This analysis showed the presence of components such as tryptophan, tyrosine, vitamin A, fluorescent oxidation products, and riboflavin. Effects of packaging material, light or dark storage, and storage time were seen. However, there was no effect of the oxygen availability on the fluorescence measurements. The score values obtained by the PARAFAC models and chemical and physical measurements were analyzed together by principal component analysis (PCA). The loadings showed a separation of the variables into three groups; the first group was related to oxidation, the second group was related to the degradation of both riboflavin and vitamin A, and the third group was linked to the protein structure. 相似文献
