Kansas and California wheat-growing regions differ dramatically in soils, climate, wheat cultivars, crop rotation patterns, and cultural practices, which could select for different fungal populations of Mycosphaerella graminicola. Our objective in this study was to use amplified fragment length polymorphism (AFLP) loci to assess the genetic diversity of M. graminicola populations within single fields in two widely separated, and geographically isolated sites in Kansas and California. Three primer-pair combinations were used to resolve polymorphism at 177 loci in 67 and 63 isolates from Kansas and California, respectively. Genotypic variability was high, which is consistent with a genetically diverse initial inoculum. There was no evidence of genetic disequilibrium in either population, with only 4.6% of the locus pairs in Kansas, and 5.4% of the locus pairs in California in detectable disequilibrium. The migration rate calculated between the two sites was as low as 1.8 individuals per generation, and significant differences in allele frequencies were observed. Therefore, these two populations do not represent mere subsamples of a larger, randomly mating population. This is a rare report of isolation by distance occurring between two North American populations of M. graminicola, indicating that at least some of these populations may be differentiating. Although genetic isolation by distance may occur, we cannot exclude movement of new gene combinations such as fungicide resistance or virulence between these two locations. 相似文献

OBJECTIVE: To determine applicability and size limits of an axial pattern flap based on the lateral caudal arteries in dogs to reconstruct caudodorsal trunk skin defects. STUDY DESIGN: Experimental study. ANIMALS: Ten mature, mixed breed dogs. METHODS: The lateral caudal vessels of the tail were incorporated in the flaps of the treatment group (n=5) and were ligated in the control group (n=5). Flaps were rotated and placed into experimentally created caudodorsal skin defects on the trunk. The length and area of tissue that remained viable in each flap were determined. RESULTS: Mean (+/-SD) survival area (222.8+/-32.9 cm2) and length (25.1+/-4.5 cm) of vascularized flaps were significantly greater (P<.05) compared with control flaps (94.9+/-13.4 cm2 and 14.61+/-4.7 cm). Necrosis occurred in all control flaps, resulting in lower percentage flap area (48.8%) and length (47.1%) survival compared with vascularized flaps (78.1%, 77.5%). CONCLUSIONS: Axial pattern flap based on lateral caudal arteries of the tail was successfully elevated and transferred in a single procedure, with 78% survival for closure of large experimentally created caudodorsal trunk defects in dogs. CLINICAL RELEVANCE: Large cutaneous defects of the caudodorsal trunk area in dog could be reconstructed with tail axial pattern flap. Limitations in terms of size and changes in animal appearance have to be considered before flap elevation. 相似文献

AbstractIn this research the effect of foliar application of selenium (Se) at four levels (Na2OSe4; 0, 5, 10 and 20?mg L?1) was evaluated on some phytochemical characteristics of Sultana grapevine under different salinity levels (NaCl; 0 or 75?mM). The vines were fed twice a week with Hoagland nutrient solution and Se was foliar applied twice with 24 intervals. During growing period, plant height, leaf number and leaf area were recorded. Moreover, at the end of experiment, mature leaves from middle nods of canes were used for measurement of some phytochemical indices. According to results, Se application had a positive effect on plant height, leaf numbers, leaf area and photosynthetic pigments content especially at 5?mg L?1 and to some extent 10?mg L?1 Se levels. Under salinity stress, foliar application of Se at 5?mg L?1 considerably decreased vines leaves electrolyte leakage and lipid peroxidation values compared to non se-treated plants under salinity stress condition. Selenium had an additive effect on salinity stress (75?mM NaCl) induced accumulation of total phenol, total flavonoid, soluble sugars and proline content in leave of vines. Moreover, the interaction of salinity and Se at 5 and 10?mg L?1 improved leaves antioxidant enzymes activities in Sultana grapevine. Likewise, foliar application of Se improved leaf mineral content in 75?mM NaCl -treated vines. Totally, foliar application of selenium (Se at 5 or 10?mg L?1) increased salt tolerance through improvement in nutritional balance and by enzymatic and non-enzymatic antioxidant capacity in grapevine leaves. 相似文献

Ferroptosis is a form of cell death associated with iron-dependent lipid peroxidation. We used a chicken embryo model to investigate if ferroptosis was implicated in the molecular mechanism underlying the potential effects of maternal exposure to iron oxide nanoparticles (IONPs) on the developing brain. One hundred and eighty fertilized eggs were randomly divided into six groups (30 eggs/group; 10 eggs/replicate). Groups I and II received maghemite (γ-Fe2O3) NPs (MGMNPs), while groups III and IV received magnetite (Fe3O4) NPs (MGTNPs). Both MGMNP and MGTNP were administrated at the concentrations of 100 and 250 ppm. One group (placebo) received saline, and the other remained untreated (control). The compounds were given by in ovo method (0.3 ml/egg) only once on the first day of the embryonic period. Samples from cerebral tissue were collected on day 20 for histopathological, biochemical and gene expression analyses. Total antioxidant capacity (TAC) and malondialdehyde (MDA) increased; glutathione peroxidase (GPX) expression and activity decreased in IONPs-treated groups. Ferroptotic cells appeared in the cerebral tissue following exposure to the low dose of MGMNP and MGTNP. Oxidative stress and ferroptotic cells were more evident for MGMNP compared to MGTNP. The low dose of MGMNP and MGTNP induced more severe oxidative stress in the cerebral tissue. According to the results, maternal exposure to IONPs is associated with ferroptosis in the brain. This work could encourage future researches to investigate inhibitors of ferroptosis as a protective strategy against iron-induced cell injuries and cell death. 相似文献

This investigation was conducted to determine the feasibility of using a two-component polyurethane (PUR) adhesive, with special waterproof properties, in constructing wooden structures. We designed and conducted tests to compare the shear strength and adhesion performance of PUR with polyvinyl acetate (PVAc) adhesive on block-shear specimens constructed of oriental beech (Fagus orientalis L.), fir (Abies alba Mill.), poplar (Populus deltoides Bartr.), white oak (Quercus alba L.), sycamore (Platanus orientalis L.) and white walnut (Juglans cinerea L.). The values of the percentage of wood failure were also determined in specimens constructed with each adhesive. The highest shear strength values of both adhesives were obtained in specimens constructed of beech, while the lowest shear strength values were obtained in fir and poplar specimens. Average shear strength of the PUR adhesive was 16.5% higher than that of the PVAc adhesive. Specimens constructed of fir, poplar and sycamore were characterised by the highest percentages of wood failure, whereas the lowest average percentages of wood failure were obtained in beech and oak specimens. With the exception of oak specimens, there was no statistically significant difference between percentage of wood failure among the PUR and PVAc adhesives. Generally, the PUR adhesive showed an acceptable adhesion performance on wood materials used in our study. 相似文献

Rat Everted Gut Sac (EGS) model was employed to study the intestinal uptake of titanium and iron. Incubation of freshly prepared rat EGS in Earle's medium pH = 7.4 containing titanium showed that the absorption of titanium as well as iron was a dose dependent process. Ascorbic acid enhanced the absorption of both metal ions, while NaF (1 mM) as an inhibitor of glycolytic energy supply, decreased their absorption. The Na+-K+ ATPase inhibitor, ouabain (1 mM) reduced intestinal absorption of Titanium. This suggests that titanium uptake is an active transport process as is iron uptake. Iron absorption was reduced approximate by 17% when titanium was presented to incubation medium EGS whereas, the absorption of titanium was decreased by 35% when iron was added to the reaction mixture. 相似文献

Background: Stroke is the third leading cause of death. Hypothermia has been recognized as an effective method in reducing brain injury. In this study, we assessed the effects of granulocyte colony-stimulating factor (G-CSF) as a neuroprotective agent and mild hypothermia on mortality, behavioral function, infarct volume, and brain edema in Wistar rats. Methods: Forty male rats were used in five groups (eight rats in each group): control, hypothermy, G-CSF, combination hypothermy + CSF, and sham. Rats were anesthetized by injection of chloral hydrate (400 mg/kg) intraperitoneally. Transient cerebral ischemia was induced by 60-min intraluminal occlusion of left middle cerebral artery. Hypothermia, initiated at the time of reperfusion and G-CSF was started one hour after reperfusion at a dose of 15 mg/kg subcutaneously. The motor behavior was measured using Garcia’s index and animals were assigned for the assessments of infarction, brain swelling, and mortality rate. Results: The mortality was 38.46% (control group) and reduced in other groups. Neurological deficit score of control group (40.31 ± 1.56) was significantly lower than in treatment groups. The total cerebral infarct volume of treatment group was significantly lower than control group (43.96 ± 44.05 mm3). Treatment with hypothermy plus G-CSF (2.69 ± 0.24%) could significantly reduce brain swelling volume than other treatment groups. Conclusion: Our major finding is that mild hypothermic treatment plus G-CSF significantly reduced mortality rate and edema and improved neurological function. The results suggest that the combination of hypothermia and G-CSF is more effectively than other treatment groups being used alone.
