Morphological changes to and collagen loss from the rat uterus during postpartum involution were investigated. The expression patterns of collagen type III and myeloperoxidase (MPO) were also determined. Morphological changes were studied on days 1, 3, 5, 10, 15, 20, 22 and 25 postpartum. As a control, diestrus rats’ uterine were used. Specimens from the uterine horn were embedded in paraffin, cut into 8 µm coronal sections, and stained with hematoxylin‐eosin. The thickness of the longitudinal and circular smooth muscle layers and of the endometrium were measured. The collagen content was determined using hydroxyproline analysis. Immunostaining was used to examine the expression of collagen type III on days 1, 3, 5, 10, 15 and 20 postpartum; and MPO on days 1, 3, 5, 10 and 22 postpartum. The thickness of the smooth muscle layers was found to decrease rapidly postpartum: the circular smooth muscle layer returned to that of a non‐pregnant, control uterus by day 5 postpartum and the longitudinal smooth muscle layer by day 15 postpartum. Eosinophilic cells were observed in the endometrial stroma adjacent to the myometrium on days 10, 15 and 20 postpartum, and were confirmed as collagenous cells. Immunostaining identified collagen type III positive cells in the vessel‐rich layer adjacent to the placental site on days 1, 3, 5 and 10 postpartum, and these cells were confirmed to be phagocytic. Postpartum reduction in the weight of the uterus was accompanied by decreases in both the collagen content and the thickness of the longitudinal and circular smooth muscle layers. Furthermore, the phagocytic cells were shown to express MPO during postpartum involution of the uterus.  相似文献

Three evolutionary lineages of the tomato wilt pathogen Fusarium oxysporum f. sp. lycopersici were found among a worldwide sample of isolates based on phylogenetic analysis of the ribosomal DNA intergenic spacer region. Each lineage consisted of isolates mainly belonging to a single or closely related vegetative compatibility group (VCG) and a single mating type (MAT). The first lineage (A1) was composed of isolates VCG 0031 and MAT1-1; the second (A2) included VCG 0030 and/or 0032 and MAT1-1; and the third (A3) included VCG 0033 and MAT1-2. Race 1 and race 2 isolates belonged to the A1 or A2 lineages, and race 3 belonged to A2 or A3 lineages, suggesting that there is no correlation between race and lineage. However, for the isolates from Japan, race 1 (with one exception), race 2, and race 3 isolates belonged to A2, A1, and A3 lineages, respectively. These results suggest that the races could have evolved independently in each lineage; and in Japan the present races were likely to have been introduced independently after they had evolved in other locations.  相似文献

The aim of this study was to determine whether 2,4-thiazolidinedione (2,4-TZD) influences the effects of peripheral insulin action in steers given recombinant bovine tumor necrosis factor (TNF) alpha (rbTNF). Steers were treated once daily for 9 d (d0 - d8) with either s.c. injection of rbTNF (2.5 microg/kg), rbTNF + i.v. injection of 2,4-TZD (2.0 mg/kg), or s.c. injection of saline (control). The plasma glucose, NEFA, and insulin concentrations in the rbTNF-treated group increased compared to those in the control and rbTNF + 2,4-TZD groups, whereas glucagon concentration decreased. A single i.v. injection of insulin (0.2 U/kg), glucose (112.5 mg/kg), or growth hormone (GH)-releasing hormone (GHRH) (0.25 microg/kg) was performed on d4, d6, and d8, respectively. In the insulin challenge, the net area under the glucose curve (AUC) was smaller in the rbTNF group than in the control and rbTNF + 2,4-TZD groups. In the glucose challenge, the net insulin AUC was smaller in rbTNF + 2,4-TZD group than in rbTNF group. In the GHRH challenge, there was no difference in GH responses to GHRH between the rbTNF and rbTNF + 2,4-TZD groups, respectively. We conclude that 2,4-TZD treatment partially reverses the impairment of peripheral insulin action caused by rbTNF injection in steers.  相似文献

Forest management has significant influences on the preferences of people for forest landscapes. This study attempted to evaluate the effects of thinning on the scenic value of forests. Five typical stands in Nanjing Wuxiangsi National Forest Park were subject to a designed thinning experiment from February to May 2007 with four intensities: unthinned, light-thinning, middle-thinning, and high-thinning. Next, people’s preferences for landscape photographs taken in plots with different thinning intensities were assessed by a scenic beauty estimation (SBE) developed by using normal function theory (N-SBE) 2 years after thinning. In order to resolve shortcomings of N-SBE, another SBE calculation method based on weighted sum theory (W-SBE) was developed in the study, and it was demonstrated to have the same reliability as N-SBE by statistical methods. Then, the difference of W-SBE between different thinning intensities was analyzed by paired samples T test. The results suggested that it was significant, at the 0.01 level, between unthinned plots and all thinned plots, and significant, at the 0.05 level, between high-thinning and light-thinning, and between high-thinning and middle-thinning, while there was no significant difference between middle-thinning and light-thinning. Finally, a stepwise regression model was established between W-SBE as the dependent variable and some possible impact preference factors as independent variables. This indicated that landscape value was diminished with increase of tree density, canopy density, and opening of understory, while the value improved with the increment of average diameter at breast height and tree height, species diversity or uniformity, and also the improvement of accessibility and environment neatness in the forest.  相似文献

We investigated the influences of chemical pretreatments and subsequent repeated dry-and-wet (RDW) treatments on the mechanical linkage between cellulose microfibrils (CMFs) and matrix substances (MT) in wood cell wall. Sugi (Cryptomeria japonica D. Don) quarter-sawn specimens were subjected to various types of chemical pretreatments, including ethanol and benzene extraction, delignification, alkali extraction, and hygrothermal treatment, to give substantial damages to principal constituents of wood cell wall, followed by 5 times of RDW treatment. After giving chemical pretreatment, the d-spacing of (200) lattice plane of cellulose Iβ (d 200), the crystallinity of wood cell wall, and the crystal size of the cellulose were measured at the fiber saturated point, using X-ray diffraction techniques. Thereafter, these were measured again at the absolutely dried condition in the process of subsequent RDW treatments. The d 200 in specimens, which were given to light pretreatments, largely expanded by drying at the early stages of RDW treatments, then it decreased and became constant after 5 times of RDW treatments. On the other hand, d 200 in specimens, which were given to intensive pretreatments, remained constant at a relatively small level throughout the whole process of RDW treatments. After 5 times of RDW treatments, d 200 became similar values between the above two groups. This suggests that RDW treatments have similar effects as intensive pretreatments, which induce substantial damages to the microscopic region in the wood cell wall such as interfacial separation between CMF, MT, and so forth. These defects would weaken mechanical interaction between CMF and MT in the wood cell wall during drying.  相似文献

To advance the identification of quantitative trait loci (QTLs) to reduce Cd content in rice (Oryza sativa L.) grains and breed low-Cd cultivars, we developed a novel population consisting of 46 chromosome segment substitution lines (CSSLs) in which donor segments of LAC23, a cultivar reported to have a low grain Cd content, were substituted into the Koshihikari genetic background. The parental cultivars and 32 CSSLs (the minimum set required for whole-genome coverage) were grown in two fields with different natural levels of soil Cd. QTL mapping by single-marker analysis using ANOVA indicated that eight chromosomal regions were associated with grain Cd content and detected a major QTL (qlGCd3) with a high F-test value in both fields (F = 9.19 and 5.60) on the long arm of chromosome 3. The LAC23 allele at qlGCd3 was associated with reduced grain Cd levels and appeared to reduce Cd transport from the shoots to the grains. Fine substitution mapping delimited qlGCd3 to a 3.5-Mbp region. Our results suggest that the low-Cd trait of LAC23 is controlled by multiple QTLs, and qlGCd3 is a promising candidate QTL to reduce the Cd level of rice grain.  相似文献

A goal in visual neuroscience is to reveal how the visual system reconstructs the three-dimensional (3D) representation of the world from two-dimensional retinal images. Although the importance of texture gradient cues in the process of 3D vision has been pointed out, most studies concentrate on the neural process based on binocular disparity. We report the neural correlates of depth perception from texture gradient in the cortex. In the caudal part of the lateral bank of intraparietal sulcus, many neurons were selective to 3D surface orientation defined by texture gradient, and their response was invariant over different types of texture pattern. Most of these neurons were also sensitive to a disparity gradient, suggesting that they integrate texture and disparity gradient signals to construct a generalized representation of 3D surface orientation.  相似文献

Cloning and characterization of cDNA for carp matrix metalloproteinase 9   总被引:1,自引:0,他引:1  
ABSTRACT: We have cloned a cDNA encoding the MMP-9 from a carp epidermal cell (EPC) cDNA library. The clone contains a 2025-base pair (bp) open reading frame encoding a protein of 674 amino acids. The deduced amino acid sequence shares 68% and 69% identity with medaka and Japanese flounder MMP-9. The hinge domain of the carp MMP-9, like those of the other non-mammalian species, lacks a type V collagen-like region that is typical of mammalian MMP-9. Gelatin zymography and immunoblot analysis of conditioned media of EPC cells and cDNA-transfected COS-7 cells detected a 76-kDa gelatinase. The apparent molecular mass of the carp zymogen is much smaller than those of its mammalian counterparts while almost identical with that of chicken 75-kDa gelatinase B-like enzyme. Although hypo-osmotic stress induced the elevation of MMP-9 mRNA level in EPC cells, no significant change in the protein in conditioned medium was detected during hypo-osmotic stress. Northern blot analysis detected a large amount of MMP-9 mRNA in carp kidney and spleen, suggesting the high expression of MMP-9 in blood cells, neutrophils, and macrophages. The smaller amount of MMP-9 mRNA was detected in gill, heart, fin, and eye, whereas none of the mRNA was detected in the hepatopancreas, intestine, brain, muscle, and skin.  相似文献
