Uptake and elimination of aflatoxins (AFs) by rainbow trout ( Oncorhynchus mykiss ) during a long-term (21 days) dietary exposure were studied to assess contamination by AFs in aquaculture fish fed AF-containing feed. The uptake factor (UF) of aflatoxin B(1) (AFB(1)) in muscle ranged from 0.40 × 10(-3) to 1.30 × 10(-3). AFB(1) concentrations in liver were 165-342 times higher than in muscle. AFs from feed were more highly accumulated in liver than in muscle. Aflatoxicol (AFL) and aflatoxin M(1) (AFM(1)) were detected in muscle and liver and also in the rearing water. AFL concentrations were higher than AFM(1) by 2 orders of magnitude in muscle, and AFL was a major metabolite of AFB(1). The elimination rate constants (α) of AFB(1) and AFL in muscle (1.83 and 2.02 day(-1), respectively) and liver (1.38 and 2.41 day(-1), respectively) were very large. The elimination half-life (t(1/2)) of AFB(1) was 0.38 days (9.12 h) in muscle and 0.50 days (12.00 h) in liver. The elimination half-life of AFL in muscle and liver was 0.34 day (8.16 h) and 0.29 day (6.96 h), respectively. These data show that AFs are eliminated rapidly and are not biomagnified in fish. Thus, AFB(1) concentration in muscle of fish fed AFB(1)-containing feed (ca. 500 μg/kg) decreased to below the detection limit (20 ng/kg) of the most sensitive analytical method at 1.54 days (36.96 h) after the change to uncontaminated feed. 相似文献

Abstract The effect of sucrose on the growth of rice plants under gnotobiotic conditions was studied at test tube scale in vinyl isolators under weak or strong light. Under weak light (500 lux) the growth of rice plants on agar medium containing only inorganic nutrients during the 20 days after transplanting stopped at 3rd leaf stage, whereas plants on agar medium containing inorganic nutrient plus 2% sucrose grew to 5th or 6th leaf stage. Under strong light (1.5 × 104 lux) the stage of tillering was promoted by supplying sucrose. The growth of rice plants in pot-hydroculture under weak light had the same tendency as that obtained in test tube culture under weak light. Considering the concentration of inorganic elements in rice shoots, it seems most likely that the uptake of inorganic elements. especially phosphorus and potassium, was enhanced by the supplements of 2% sucrose under weak light. An increase of calcium and manganese uptake was observed in plants grown in medium containing 2% sucrose compared with that of plants grown in inorganic medium alone under strong light. The uptake of inorganic elements from medium containing inorganic nutrients plus 2% sucrose depended chiefly on light energy under strong light in the phytotron used. 相似文献

Eleven rice varieties differing in grain size were grown under controlled environmental conditions during the grain-filling period. The grain weight of upper grains in a panicle was examined at successive stages of growth during the grain-filling period. The effect of temperature on the rate and the duration of the period of grain-filling was determined using Khao Lo, a large-grain variety, and Bom Dia, a small-grain variety. Both the grain-filling rate and duration of the period of grain-filling differed among rice varieties and were positively and significantly correlated with the grain size. The duration of the grain-filling period from flowering to the time when almost maximum grain weight was attained ranged from 12 days at 32/24°C in Bom Dia to 36 days at 20/12°C in Khao Lo. The grain-filling rate was low in small-grain varieties, and generally increased with increasing grain size. By lowering the temperature, the grain-filling rate decreased, the duration of the grain-filling period increased but the grain weight was almost constant. Weight per grain was closely correlated with hull size. No relationship was found between weight per grain and nitrogen percentage of brown rice. 相似文献

We examined arsenic (As) accumulation and speciation in the major cultivars currently grown in Japan, because differences in grain As levels among cultivars may influence dietary As exposure in Japanese people. Ten major cultivars (Oryza sativa L.) were grown under flooded conditions in a paddy field with a background level of As (low-As soil) or in pots filled with soil containing a high level of As (high-As soil). In the low-As soil, the total grain As ranged from 0.11 to 0.17?mg?kg?1, with a mean concentration of 0.14?mg?kg?1, and inorganic As was the major species in all cultivars. There were few genotypic differences in the levels of either total As or inorganic As in the grain. In the high-As soil, total grain As increased to a mean level of 2.4?mg?kg?1 in the 10 cultivars, with markedly increased levels of dimethylarsinic acid. The genotypic variations among cultivars in the levels of both total As and dimethylarsinic acid were statistically significant. However, the genotypic variability of inorganic As levels was quite small, and these levels remained low (at about 0.2?mg?kg?1) even when total As levels increased markedly. These results suggest that differences in grain As levels among Japanese cultivars may not influence dietary As exposure, because there is little genotypic difference in the accumulation of inorganic As, which is considered more toxic than organic As to humans. We discuss the possible mechanism of As accumulation in Japanese paddy rice, in the context of the accumulation of As species in the developing grain and in other plant tissues. 相似文献

Land-use type and nitrogen (N) addition strongly affect nitrous oxide (N2O) and carbon dioxide (CO2) production, but the impacts of their interaction and the controlling factors remain unclear. The aim of this study was to evaluate the effect of both factors simultaneously on N2O and CO2 production and associated soil chemical and biological properties. Surface soils (0–10 cm) from three adjacent lands (apple orchard, grassland and deciduous forest) in central Japan were selected and incubated aerobically for 12 weeks with addition of 0, 30 or 150 kg N ha–1 yr–1. Land-use type had a significant (p < 0.001) impact on the cumulative N2O and CO2 production. Soils from the apple orchard had higher N2O and CO2 production potentials than those from the grassland and forest soils. Soil net N mineralization rate had a positive correlation with both soil N2O and CO2 production rates. Furthermore, the N2O production rate was positively correlated with the CO2 production rate. In the soils with no N addition, the dominant soil properties influencing N2O production were found to be the ammonium-N content and the ratio of soil microbial biomass carbon to nitrogen (MBC/MBN), while those for CO2 production were the content of nitrate-N and soluble organic carbon. N2O production increased with the increase in added N doses for the three land-use types and depended on the status of the initial soil available N. The effect of N addition on CO2 production varied with land use type; with the increase of N addition doses, it decreased for the apple orchard and forest soils but increased for the grassland soils. This difference might be due to the differences in microbial flora as indicated by the MBC/MBN ratio. ABSTRACT Two begomoviruses (Java virus-1 and Java virus-2), two satellite DNAs (DNAbeta01 and DNAbeta02), and a recombinant DNA (recDNA) were cloned from a single tomato plant from Indonesia with leaf curl symptoms, and the role of these satellite DNAs in the etiology of begomovirus disease was investigated. The genome organizations of the two viruses were similar to those of other Old World monopartite begomoviruses. Comparison of the sequences with other begomoviruses revealed that Java virus-1 was a newly described virus for which the name Tomato leaf curl Java virus (ToLCJAV) is proposed. Java virus-2 was a strain of Ageratum yellow vein virus (AYVV) (AYVV-[Java]). ToLCJAV or AYVV-[Java] alone did not induce leaf curl symptoms in tomato plants. However, in the presence of DNAbeta02, both ToLCJAV and AYVV-[Java] induced leaf curl symptoms in tomato plants. In the presence of DNAbeta01, these viruses induced mild leaf curl symptoms in tomato plants. The recDNA had a chimeric sequence, which arose from recombination among ToLCJAV, AYVV-[Java], DNAbeta01, and DNAbeta02; it was replicated only in the presence of AYVV-[Java] in tomato plants. 相似文献

Myostatin (MSTN) is a negative regulator during muscle differentiation, whereas insulin‐like growth factors (IGFs) are essential for muscle development. MSTN and IGFs act oppositely during myogenesis, but there is little information on the mutual relationship of MSTN and IGFs. The present study was conducted to examine whether MSTN affects IGF expression during early myogenesis in cattle. IGF‐1 mRNA was similarly expressed in M. longissimus thoracis of double‐muscled (DM) and normal (NM) Japanese shorthorn cattle. IGF‐2 mRNA expression was consistently higher in the normal and regenerating muscle of DM cattle than those of NM cattle. When myoblasts were isolated from regenerating M. longissimus thoracis, IGF‐2 mRNA expression showed a significant increase in differentiating DM derived myoblasts (DM‐myoblasts) as compared with differentiating NM derived myoblasts (NM‐myoblasts). An addition of recombinant mouse myostatin (rMSTN) to myoblast cultures attenuated IGF‐2 mRNA expression and decreased myotube formation, but did not effect IGF‐1 mRNA expression. An activin‐like kinase (ALK) inhibitor, SB431542, mediates MSTN action, suppressed the translocation of Smad2/3 into the nucleus in DM‐myoblasts, and restored the attenuated IGF‐2 mRNA expression and the decreased myotube formation induced by rMSTN in myoblast cultures. The findings indicate that MSTN may negatively regulate myoblast differentiation by suppressing IGF‐2 expression via ALK‐Smad signaling. 相似文献
