The development and distribution of lymphocyte subsets in calf palatine and pharyngeal tonsil were examined. During prenatal development, B cells were distributed in the subepithelial area, and T cells and MHC class II+ cells were found in the deep layer of B-cell area, respectively, in both tonsils. At neonatal stage, lymphoid follicle containing a few CD4+ cells have been formed in both tonsils. IgG+ and IgA+ cells were found in the parafollicular and epithelial area. At 3 months old, many germinal centers were recognized in both tonsils. CD4+ cells and IgG mRNA expression were detected in light zone of germinal centers. Many IgG, and IgA mRNA expressions also could be detected in the parafollicular and subepithelial area of both tonsils. The data suggest that both tonsils have an important role of local immune defense against invading antigen after birth. The comparison of the histological characteristics of tonsil and Peyer's patch during ontogeny is also discussed. 相似文献

The duration of maternally-derived antibodies against three arboviruses was investigated in calves, using the results of arbovirus serosurveillance performed in Kagoshima Prefecture during 2002–2016. The duration of maternally-derived antibodies against Akabane virus (AKAV), Aino virus (AINOV), and Chuzan virus (CHUV) was estimated to be 178 (sensitivity: 0.769, specificity: 0.730), 156 (sensitivity: 0.806, specificity: 0.791), and 156 days of age (sensitivity: 0.845, specificity: 0.814), by receiver operating characteristic analysis. The duration of maternally-derived antibodies against AKAV, AINOV, and CHUV differed 7–14, 22–28, and 20–31 days in the same calf types between the regions far from each other although it was similar between the adjacent regions. The dairy calves showed 6–29 days longer duration than the beef calves rearing in a similar region. 相似文献

The present study evaluated the advantage of mixed‐model techniques over a selection index under different magnitudes of an additional systematic environmental effect (ASEE) in terms of accuracy of prediction and expected genetic gain. The data attempted to simulate a closed herd in a pig breeding program. The base population (G0) consisted of 10 males and 50 females. Six generations (G0 to G5) were selected by using a selection index of three traits without overlapping. Additional systematic environmental constants with four levels in a generation were assigned from a uniform distribution at different ranges. Breeding values of animals in the last generation (G5) were estimated on the basis of an index of individual phenotype (SI‐U), SI‐U adjusted for ASEE using a least‐squares mean (SI‐A), best linear unbiased prediction using an animal model excluding ASEE (AM‐E), and an animal model including ASEE (AM‐I). Accuracy of prediction and expected genetic gain were larger by the animal model than by the selection index, even if heritability of the traits selected was high and ASEE was set to zero. When ASEE was zero, the accuracy of prediction and expected genetic gain given by SI‐U and AM‐I were similar to those given by SI‐A and AM‐E, respectively. However, the differences in accuracy and expected gain between SI‐U and AI‐A and between AM‐I and AM‐E increased as the range of ASEE increased. It was concluded that selection based on an animal model was more effective than index selection, even if the herd environment was uniform and traits with high heritability were selected, and that it should be always included in an evaluation model, however slight any systematic environmental effect may be in a closed herd. 相似文献

Experiments were designed to compare the adipocyte cellularity of subcutaneous adipose tissue between growing Landrace (low backfat) and Meishan (high backfat) pigs at 1 week, 3 weeks, 6 weeks, 3 months and 5 months of age. As pigs aged, body weight and backfat thickness of both breeds significantly increased. When compared at equal ages, backfat thickness adjusted to equal body weight was greater for Meishan pigs. The mean diameter of fat cell size also increased with age, and by 6 weeks adipocytes from both outer and inner layers of subcutaneous adipose tissue were larger in Meishan pigs. At 5 months, approximately 80% of the adipose tissue mass in Meishan pigs was attributable to adipocytes measuring 95–165 µm in diameter, whereas adipocytes of 75–145 µm comprised most of the tissue mass in the Landrace. Although the contribution of smaller adipocytes (25–45 µm) to the tissue volume was negligible, both breeds showed a biphasic diameter distribution at all ages, suggesting that adipocyte hyperplasia is still active. Our results demonstrate that cellularity differences exist between the subcutaneous adipose tissues of Landrace and Meishan pigs, and adipocyte hypertrophy is the most overwhelming contributor to the greater backfat deposition for Meishan pigs. 相似文献

Akabane virus (AKAV), belonging to the genus Orthobunyavirus and family Peribunyaviridae, causes reproductive and congenital abnormalities in ruminants. Its envelope glycoprotein Gc is a neutralizing antigen, on which at least five distinct antigenic regions have been identified. We attempted to identify the domains using truncated recombinant AKAV Gc proteins expressed in Escherichia coli and monoclonal antibodies (mAbs) with AKAV-neutralizing activity. Dot blot analysis revealed that amino acid positions 1–97 and 189–397 (Gc1–97 and Gc189–397) in the truncated recombinant proteins reacted with the mAbs. Additionally, AKAV was neutralized by sera from mice immunized with these recombinant proteins. The results suggested that the two domains contain neutralizing epitopes and could be potential subunit vaccines against AKAV. 相似文献

Serum samples from 2420 clinically healthy dairy cattle, randomly selected from stored sera in 18 districts of Japan, were tested for the presence of Neospora caninum antibodies using an indirect fluorescent antibody test (titer > or =1:200). Nationwide seroprevalence is estimated at 5.7% (139/2420). Seropositive cattle were detected in all surveyed districts despite the evidence of confirmed case reports of bovine neosporosis, showing that N. caninum is widely distributed throughout Japan. Age-specific seroprevalence did not increase with cattle age, suggesting that Neospora infection is likely to be transmitted vertically rather than horizontally in Japan. Considering that N. caninum seropositive cows are thought to be more likely to abort, substantial fetal losses may be induced by N. caninum infection in Japan. Devising strategies are needed to reduce the economic impact on the Japanese dairy industry. This is the first study to investigate the nationwide seroprevalence of N. caninum in cattle in Asia. 相似文献

通过测定盐酸、己酸处理的玉米秸秆青贮饲料的pH值、含水率、有机酸含量及组成、干物质消失率、NDF瘤胃降解率、干物质回收率和青贮饲料化学成分,研究了盐酸、己酸的处理对玉米秸秆青贮饲料质量的影响。试验结果表明:在玉米秸秆青贮饲料中添加盐酸,降低了青贮饲料中乳酸占总酸的比例和干物质回收率,并使青贮饲料的细胞壁物质含量显著增加,明显降低了玉米秸秆青贮饲料的质量;己酸的添加对青贮饲料质量无显著影响(P>0.05);盐酸和己酸的共同处理除了明显改善青贮饲料的色泽、气味和质地外,还可明显降低青贮饲料中OB,ADF和ADL的含量(P>0.05),显著提高了青贮饲料的营养价值。 相似文献

Using RT-PCR and semi-quantitative PCR, mRNA expression for canine interferon (IFN)-gamma, interleukin (IL)-2, IL-4, IL-5, IL-10, tumor necrosis factor (TNF)-alpha and transforming growth factor (TGF)-beta in peripheral blood mononuclear cells (PBMCs) was examined in dogs with or without demodicosis. mRNA expression for IFN-gamma as well as TNF-alpha in dogs with demodicosis (localized (LD) and generalized (GD)) was slightly lower than those in dogs without demodicosis (healthy controls). Expression of IL-5 mRNA in dogs with demodicosis was higher than that in control dogs, but there were no significant differences in IL-4 and IL-10 mRNA expression levels among the three groups. On the other hand, expression levels of TGF-beta mRNA in dogs with GD were higher than those in control dogs and dogs with LD. The expression levels of IL-5 and TGF-beta mRNA decreased in all three dogs with GD which showed resolution of the clinical signs. Taken together, these results suggest that the Th2-like response in PBMCs from dogs with demodicosis is up-regulated, and that subsequent increased expression of IL-5 and TGF-beta mRNA in dogs with GD is reversible after treatment. Therefore, these cytokines, particularly IL-5, might be a useful clinical index of the clinical course in demodicosis. Also, increased TGF-beta mRNA expression might be a key factor for revealing the difference in the mechanism of onset between LD and GD. 相似文献
