BACKGROUND: In a large cropping area of northern Argentina, Sorghum halepense (Johnsongrass) has evolved towards glyphosate resistance. This study aimed to determine the molecular and biochemical basis conferring glyphosate resistance in this species. Experiments were conducted to assess target EPSPS gene sequences and 14C‐glyphosate leaf absorption and translocation to meristematic tissues. RESULTS: Individuals of all resistant (R) accessions exhibited significantly less glyphosate translocation to root (11% versus 29%) and stem (9% versus 26%) meristems when compared with susceptible (S) plants. A notably higher proportion of the applied glyphosate remained in the treated leaves of R plants (63%) than in the treated leaves of S plants (27%). In addition, individuals of S. halepense accession R2 consistently showed lower glyphosate absorption rates in both adaxial (10–20%) and abaxial (20–25%) leaf surfaces compared with S plants. No glyphosate resistance endowing mutations in the EPSPS gene at Pro‐101–106 residues were found in any of the evaluated R accessions. CONCLUSION: The results of the present investigation indicate that reduced glyphosate translocation to meristems is the primary mechanism endowing glyphosate resistance in S. halepense from cropping fields in Argentina. The temporomandibular joint is formed between the condyloid process of the mandible and the mandibular fossa of the temporal bone. The basic anatomy of this joint was assessed and described in a series of skulls including dolichocephalic, mesaticephalic and brachycephalic breeds. The facial index and rotational angles were measured with the facial index providing a useful method of classifying skull types but the rotational angle being of limited use in assessment of the temporomandibular joint until normal breed values are established. Equipment was designed to allow repeatable positioning of the temporomandibular joint for radiography at a variety of lateral and long axis rotational angles relative to the central x-ray beam. The regions of the joint and anatomic features visualized in each view are demonstrated. 10 degrees rotation was required in either axis to project the joints independently of each other. Lateral rotational angles of 10 to 30 degrees in mesaticephalic and dolichocephalic breeds and 20 to 30 degrees in brachycephalics and long axis rotational views of 10 to 30 degrees depending on the region of interest were considered to be the most useful. 相似文献

Sequence-related amplified polymorphism (SRAP) and morphological markers to estimate the genetic relations among forty pea varieties (Pisum sativum L.) were studied. Data on 15 morphological traits were collected and analyzed. A total of 162 polymorphic SRAP's bands were scored using seven combinations of primers. Cluster analysis and both principal component and principal coordinate analysis were carried out. The varieties were grouped in four clusters through procrustes generalized analysis. Relationships among varieties revealed by molecular markers were significantly correlated with those based on the agronomic traits, suggesting that the two systems give similar estimates of genetic relations among the varieties. Mycobacterium thermoresistibile was isolated in pure culture from ultrasound-guided pulmonary aspirates taken from a young cat with severe, chronic, pyogranulomatous pneumonia. Thoracic radiography and ultrasonography before therapy demonstrated severe diffuse alveolar disease. Twelve months combination therapy with doxycycline, rifampicin and clarithromycin resolved the infection. Thoracic radiographs taken at the completion of therapy showed multifocal pulmonary mineralisation. M thermoresistibile has been infrequently reported as a human or animal pathogen. This is the first reported pulmonary infection by M thermoresistibile in a cat and documents the successful treatment of the organism in a feline patient. 相似文献

Objective— Compare the biomechanical characteristics of screw and wire fixation with and without polymethylmethacrylate (PMMA) re-enforcement for acetabular osteotomy stabilization in dogs. Animals— Pelves removed from 8 adult mixed breed dogs weighing between 25 and 30 kg. Procedure— The pubic symphysis of each pelvis was split and a central transverse acetabular osteotomy was performed. One hemipelvis from each dog was stabilized with the composite fixation (interfragmentary Kirschner wire, two screws and a figure-of-eight orthopedic wire with PMMA). The contralateral hemipelves was stabilized with an interfragmentary Kirschner wire, two screws, and a figure-of-eight orthopedic wire without PMMA. All hemipelves were tested in bending by using a materials testing machine at a cross head speed of 5 mm/min. An extensometer was placed on the dorsomedial surface of the hemipelves centered over acetabular osteotomy to record distraction of the osteotomy during loading. A load/deformation curve and a load/distraction curve was produced for each hemipelvis. The slope for the initial linear portion of the load/deformation curve and the load/distraction curve, yield load and maximum load sustained were compared between repair groups using a paired t-test with P < .05 considered significant. Results— The slope of the load/deformation curve was significantly greater (P= .001 ) for hemipelves stabilized with the composite fixation (mean ± SD: 69 ± 18 N/mm) compared with hemipelves stabilized without PMMA (mean ± SD: 39 ± 8 N/mm). There was no significant difference (P= .593 ) between repair groups in the slope of the load/distraction curves as measured on the extensometer. Yield load was significantly greater (P= .0002 ) for hemipelves stabilized with the composite fixation (mean ± SD: 184 ± 25 N) compared to hemipelves stabilized without PMMA (mean ± SD: 74 ± 12 N). Maximum load sustained was also significantly greater (P= .013 ) for hemipelves stabilized with the composite fixation (mean ± SD: 396 ± 71 N) compared to hemipelves stabilized without PMMA (mean ± SD: 265 ± 94 N). Failure of hemipelves stabilized with the composite fixation occurred primarily by ventrolateral bending of the cranial and caudal pelvic segments at the osteotomy site. Failure of hemipelves stabilized without PMMA occurred by ventrolateral bending of the cranial and caudal pelvic segments at the osteotomy site with pronounced concurrent ventrolateral rotation of the cranial pelvic segment. Conclusion— PMMA improves the mechanical characteristics of acetabular fracture fixation, at least in part by neutralization of rotational forces. - 1. Freshwater pearl mussel (Margaritifera margaritifera L.) populations are under serious threat of extinction throughout their geographical range and only a few remnant populations are recruiting to adulthood. Consequently, M. margaritifera is classified as endangered on the International Union for Conservation of Nature Red List.
- 2. Several institutions across many countries have set up Ark sites at hatcheries to culture and rear young M. margaritifera from population remnants, with the intention of stocking these juveniles into rivers. The release location must fulfill the habitat requirements for the full life‐cycle of the species, so they can contribute to the next generation and thus the long‐term recovery of the species. However, little research or advice exists about how to decide if river environments are suitable for stocking.
- 3. A protocol is presented for determining whether a M. margaritifera population will benefit from stocking hatchery reared juveniles and how to identify suitable areas. Stocking locations are considered from catchment scale to microscale using water quality (reach), macrohabitat (site) and microhabitat, including physicochemical properties of the substratum (spots).
- 4. A case study of the River Esk in north‐east England, is incorporated to exemplify the myriad of considerations surrounding attempts to conserve M. margaritifera, and describes how implementation of the protocol can structure and assist stocking programmes. Trees alter microclimate and radiation budget in alley cropping systems, which in turn change physiological processes of the grassland component. The influence of the trees on grassland growth depends on the climate zone as well as possibly on weather conditions during the respective growth interval. The proportion of shade-tolerant species in the grassland further modifies the tree effect on grassland growth, as well as the number of annual harvests. We studied the effect of distance and orientation relative to the tree line (tree line in the South-West vs. North-East) on growth rates of the adjacent grassland with two cutting frequencies (frequent vs. infrequent) in an alley cropping system with grassland of two different vegetation compositions (diverse vs. grass-clover) and willow clones. Microclimate parameters and radiation were measured at the points where growth rates were determined to better characterize the effect of position relative to the tree line. Growth rates differed significantly (p < 0.05) depending on the distance to the trees and annual biomass accumulation amounted to 389 ± 13.5 g m−2 in the center vs. 321 ± 10.0 g m−2 with trees in the North-East vs. 292 ± 9.2 g m−2 with trees in the South-West of the transect position. Lower growth rates occurred at positions with lower light quantities close to the trees, namely the position with the trees adjacent in the South-West. Temperature and relative humidity differences between the distances were significantly (p < 0.05) affected by distance to the tree line, but differences in neither soil nor air temperatures between the distances affected growth rates. Vegetation composition mediates the influence of the trees: the difference between positions was more pronounced in the diverse vegetation composition with higher productivities in the center between the alleys, but also with higher overall sward productivity. 相似文献
