Two dogs, one from California and one from Arizona, were found to have aberrant infections caused by filarial nematodes of the genus Onchocerca. In both cases, the parasites are localized in or near the eye. In one case the worm was located in the cornea and was surgically removed. In the second case, a very marked granulomatous reaction was induced in the retrobulbar space, mimicking an abscess. This eye was enucleated. The worms in both instances were female, and were gravid, i.e. contained microfilariae in utero, indicating that a male worm(s) had been present and mating had occurred. The exact identity of the species of Onchocerca responsible cannot be determined, although the features observed are most like Onchocerca lienalis of cattle. These cases represent the fourth and fifth such cases reported from the US, and are especially interesting because of the unusual location of the worms, the small number of recognized cases, and the similarity to a recent zoonotic human infection.  相似文献

We report efficient sample extraction and assay methods allowing quantitative determinations of proteinase activities from barley malt. The improved methods are used to assay >2,200 developmental lines of malting barley for two subsets of proteinase activity. The distributions of the resulting activities suggest differences in population structures between the two types of proteinases. Comparison of the activities of the green malt proteinases with standard malting quality measurements show highly significant correlations that differ between the proteinase subsets. The pH 4.5 hydrolysis of the artificial substrate Z-Phe-Arg-AMC correlates well with the traditional malting quality measurements, supporting the role of cysteine-class proteinases in mobilization of grain reserves during malting and mashing. Results from assays of gelatin hydrolysis at pH 6.0 suggest that these proteolytic activities may be involved in other aspects of seed C and N dynamics also linked to malting quality measurements. The differences between the pH 4.5 and 6.0 activities assayed here and their association with malting quality measurements suggest different physiological roles for the two proteinase activities in several aspects of seed germination. Either assay could be useful for population surveys, depending on the particular facet of seed metabolism under study.  相似文献

Rice endosperm often develop a yellow discoloration during commercial storage in conditions of high temperature and moisture, thereby reducing the value of the grain. This postharvest yellowing (PHY) appears to be coincidental with fungal presence. To study the yellowing process in a controlled manner, we developed a technique to induce PHY on a small, laboratory scale. Milled rice kernels were rinsed with water and incubated in clear test tubes or microfuge tubes at 65–80°C. This allowed direct observation of the color change and measurement using a colorimeter. Every rice cultivar tested (long and medium grain japonicas and indicas) showed some level of PHY, which increased with temperature yielding a maximum color change at 79°C. Most color change occurred within one day. The moisture parameters required for yellowing to occur were measured. Using sterilization and culture techniques, we found no indications of direct fungal involvement in the yellowing process.  相似文献

A significant and well-supported hypothesis is that increased growth following nitrogen (N) fertilization is a function of the relationships among photosynthesis, tissue N content and the light environment-specifically, the within-canopy allocation of N among leaves and the within-leaf allocation of N between Rubisco and chlorophyll. We tested this hypothesis in a field trial that included annual applications of N,P,K fertilizer (from planting) to a Eucalyptus globulus Labill. plantation growing on uniform leached sands. Growth of 4-year-old E. globulus increased significantly in response to fertilization. Leaf N and phosphorus concentrations were 0.1-0.5 g m(-2) and 0.4-0.5 g m(-2) higher in fertilized trees compared to unfertilized trees, respectively. Stomatal conductance (g(s)) at the maximum photosynthetic rate (A(max)) was between 0.2 and 0.4 mol m(-2) s(-1) higher in fertilized trees, but A(max) and the concentration of Rubisco (Rub(a)) were unaffected by fertilization. This seeming paradox, where there was no response of A(max) to fertilization despite increases in g(s) and leaf N concentration, was explained by reduced in vivo specific activity of Rubisco in fertilized trees. Acclimation to light, measured by redistribution of N between Rubisco and chlorophyll, was unaffected by fertilization. Distribution of leaf N followed irradiance gradients, but A(max) did not. Maximum photosynthetic rate was correlated with leaf N concentration only in unfertilized trees. These findings indicate that the relationships among photosynthesis, N and the light environment in E. globulus are affected by N,P,K fertilization.  相似文献

Two new, one known and three unidentified species of the nematode family Camallanidae are reported and described from the intestines of marine perciform fishes off the southwestern coast of New Caledonia, South Pacific: Camallanus carangis Olsen, 1952 from the forked-tailed threadfin bream Nemipterus furcosus (Nemipteridae), the yellowstriped goatfish Upeneus vittatus and the whitesaddle goatfish Parupeneus ciliatus (both Mullidae) (new host records); Procamallanus (Spirocamallanus) variolae sp. n. from the white-edged lyretail Variola albimarginata (type host) and the blacktip grouper Epinephelus fasciatus (both Serranidae); Procamallanus (Spirocamallanus) longus sp. n. from the twotone tang Zebrasoma scopas (Acanthuridae); Procamallanus (Spirocamallanus) sp. 1 (female tail with 2 terminal spikes on a digit-like projection) from the speckled sand-perch Parapercis hexophtalma (Pinguipedidae); Procamallanus (Spirocamallanus) sp. 2 (female tail with 1 spike on a digit-like projection) from the drab emperor Lethrinus ravus (Lethrinidae) and Procamallanus (Spirocamallanus) sp. 3 (female tail with a smooth digit-like protrusion) from the two-lined monocle bream Scolopsis bilineata (Nemipteridae). Camallanus paracarangis Velasquez, 1980 is synonymized with C. carangis. Several additional species of Camallanus from marine fish of the Indo-Pacific region may be synonymous with C. carangis as it has a poorly sclerotized left spicule and 3 small caudal projections on the tail of young (i.e., non-gravid) females. The fourth-stage larva of C. carangis is described for the first time. P. (S.) variolae differs from most similar species of this region mainly in the position (i.e., at level or posterior to the nerve ring) and shape of deirids. P. (S.) longus differs from the similar P. (S.) chaimha mainly in a different arrangement of postanal papillae, shape of the female tail, much longer right spicule (429 microm) and longer body of gravid females (38-55 mm). All Camallanus and Procamallanus (Spirocamallanus) spp. reported here represent the first records of camallanids from marine fishes in New Caledonian waters.  相似文献

Climate change and rising carbon dioxide (CO2) levels are expected to reduce the mineral nutrient content of soybean seeds. The main objective of this study was to survey diverse soybean germplasm for variation in seed elemental concentrations and their relationships between elements, protein content, and individual seed weight. Seeds from 90 soybean genotypes were weighed and subjected to inductively coupled plasma-mass spectrometry (ICP-MS) ionomics analysis and Carbon/Nitrogen (C/N) analysis to determine protein. The results demonstrated substantial variation with the possibility of significantly improving most mineral nutrients, especially selenium (Se), copper (Cu), iron (Fe), and manganese (Mn). This diverse survey identifies genotypes that can complement existing soybean breeding programs for improving seed nutritional quality. Correlation analysis identified two clusters of co-variant elements: zinc (Zn), phosphorus (P), and sulfur (S) as well as Zn, Cu, Se, and rubidium (Rb) were positively correlated with each other. Tolerable upper limits of Rb intake are not defined for humans illustrating the need to monitor trace elements along with desirable nutrients.  相似文献

Flooding river valleys following construction of dams restrict fluvial environments to reaches that were formerly headwaters. Whether remaining habitat is suitable for all life stages of fluvial species is poorly understood. A fluvial species, Arctic grayling Thymallus arcticus, showed a dramatic decline following flooding of the Upper Peace River and the formation of the Williston Reservoir. We related landscape and field site‐specific features with occurrence of juvenile Arctic grayling using an information theoretic approach. For the landscape model, an association was identified between stream order and Arctic grayling occurrence although stream order alone was a poor predictor. A positive association between juvenile Arctic grayling occurrence and distance from the Williston reservoir and stream order, as well as a negative association with water temperature and temperature variance, was deemed important for the field site model. Both modelling approaches indicated size of stream system to be an important influence on occurrence of juvenile grayling in the Williston watershed. River length required for suitable river habitat for salmonids has not previously been identified, but should be factored into future management plans when evaluating the impact of proposed hydroelectric dams and subsequent flooding of river systems.  相似文献
