This study evaluated immune cell populations in pigs following weaning and vaccination for Mycoplasma hyopneumoniae. Piglets (n = 24) were weaned (day 0) at 16 (±1) days of age, and randomly assigned to the vaccination group (n = 16) or control group (n = 8). Complete blood cell counts, flow cytometry and serology were completed for blood samples collected on days 0 (within hours of weaning), 3, 7, 14, 30 and 60. The M. hyopneumoniae S:P ratios (sample optical density: positive control optical density) were negative in the vaccination group until days 30 and 60, when the S:P ratios were 1.3 and 1.0, respectively. Control animals remained serologically negative. The percentage of CD4+ T cells was less (P < 0.01) in control pigs than vaccinated pigs at day 3. In contrast, numbers of CD8+ and CD4+CD8+ T cells were greater (P < 0.01) in control pigs than in vaccinated pigs at days 3 and 7. After day 7, few differences in immune cell types were evident between the groups. Differences in lymphocyte populations could not be solely attributed to vaccination, due at least in part, to the confounding influence of weaning. It was difficult to distinguish the influence of vaccination from the impact of weaning on peripheral immune cell populations.  相似文献

Driven by population pressures on natural resources, peri-urban pastoralists in the Far North Province of Cameroon have recently intensified livestock production in their traditional pastoral system by feeding their cattle cottonseed cakes and other agricultural byproducts to cope with the disappearance of rangelands typically available through the dry season. Although the crop–livestock interactions in this altered intensive pastoral system seem similar to alterations recently named in mixed-farming systems in West Africa, they are distinctly different and would require a different type of agricultural development support. I use Bourdieu’s theoretical constructs of “habitus” and “capital” to explain those differences.  相似文献

Structure determination of proteins and other macromolecules has historically required the growth of high-quality crystals sufficiently large to diffract x-rays efficiently while withstanding radiation damage. We applied serial femtosecond crystallography (SFX) using an x-ray free-electron laser (XFEL) to obtain high-resolution structural information from microcrystals (less than 1 micrometer by 1 micrometer by 3 micrometers) of the well-characterized model protein lysozyme. The agreement with synchrotron data demonstrates the immediate relevance of SFX for analyzing the structure of the large group of difficult-to-crystallize molecules.  相似文献

Variation in nutrition is a key determinant of growth, body composition, and the ability of animals to perform to their genetic potential. Depending on the quality of feed available, animals may be able to overcome negative effects of prior nutritional restriction, increasing intake and rates of tissue gain, but full compensation may not occur. A 2 × 3 × 4 factorial serial slaughter study was conducted to examine the effects of prior nutritional restriction, dietary energy density, and supplemental rumen undegradable protein (RUP) on intake, growth, and body composition of lambs. After an initial slaughter (n = 8), 124 4-mo-old Merino cross wethers (28.4 ± 1.8 kg) were assigned to either restricted (LO, 500 g/d) or unrestricted (HI, 1500 g/d) intake of lucerne and oat pellets. After 8 wk, eight lambs/group were slaughtered and tissue weights and chemical composition were measured. Remaining lambs were randomly assigned to a factorial combination of dietary energy density (7.8, 9.2, and 10.7 MJ/kg DM) and supplemental RUP (0, 30, 60, and 90 g/d) and fed ad libitum for a 12- to 13-wk experimental period before slaughter and analysis. By week 3 of the experimental period, lambs fed the same level of energy had similar DMI (g/d) and MEI (MJ/d) (P > 0.05), regardless of prior level of nutrition. Restricted-refed (LO) lambs had higher rates of fat and protein gain than HI lambs (P < 0.05) but had similar visceral masses (P > 0.05). However, LO lambs were lighter and leaner at slaughter, with proportionally larger rumens and livers (P < 0.05). Tissue masses increased with increasing dietary energy density, as did DMI, energy and nitrogen (N) retention (% intake), and rates of protein and fat gain (P < 0.05). The liver increased proportionally with increasing dietary energy density and RUP (P < 0.05), but rumen size decreased relative to the empty body as dietary energy density increased (P < 0.05) and did not respond to RUP (P > 0.05). Fat deposition was greatest in lambs fed 60 g/d supplemental RUP (P < 0.05). However, lambs fed 90 g/d were as lean as lambs that did not receive supplement (P0, P > 0.05), with poorer nitrogen retention and proportionally heavier livers than P0 lambs (P < 0.05). In general, visceral protein was the first tissue to respond to increased intake during refeeding, followed by non-visceral protein and fat, highlighting the influence of differences in tissue response over time on animal performance and body composition.  相似文献

Microscopic sulfides with low 34S/32S ratios in marine sulfate deposits from the 3490-million-year old Dresser Formation, Australia, have been interpreted as evidence for the presence of early sulfate-reducing organisms on Earth. We show that these microscopic sulfides have a mass-independently fractionated sulfur isotopic anomaly (Delta33S) that differs from that of their host sulfate (barite). These microscopic sulfides could not have been produced by sulfate-reducing microbes, nor by abiologic processes that involve reduction of sulfate. Instead, we interpret the combined negative delta34S and positive Delta33S signature of these microscopic sulfides as evidence for the early existence of organisms that disproportionate elemental sulfur.  相似文献

We review aspects of climate change likely to impact upon the Collembola and mites (microarthropods) of Victoria Land and the Queen Maud Mountains (VLQMM) in the Ross Sea Region of Antarctica. Five important aspects of biological and biological-environmental interactions are identified as key for understanding the impact of climate change on VLQMM microarthropods: (1) Water availability and utilization; (2) mean temperature (which will affect development and population processes) and extreme temperatures (which affect persistence); (3) ultraviolet radiation, although we note that the periods of peak UV irradiance and microarthropod activity do not coincide; (4) dispersal within and between habitats; and (5) potential establishment of invasive species from within and without Antarctica. The current evidence for effects of climate change on VLQMM microarthropods is equivocal, and we advocate targeted experimental and monitoring studies. Finally, we highlight several areas of high priority for future research, particularly on the mite fauna for which detailed information is currently lacking. These are: (1) functional ecology (including thermal biology, feeding and nutrition and water relations); (2) distribution, dispersal and colonization processes and (3) population and community ecology.  相似文献

As a first step toward understanding how rare variants contribute to risk for complex diseases, we sequenced 15,585 human protein-coding genes to an average median depth of 111× in 2440 individuals of European (n = 1351) and African (n = 1088) ancestry. We identified over 500,000 single-nucleotide variants (SNVs), the majority of which were rare (86% with a minor allele frequency less than 0.5%), previously unknown (82%), and population-specific (82%). On average, 2.3% of the 13,595 SNVs each person carried were predicted to affect protein function of ~313 genes per genome, and ~95.7% of SNVs predicted to be functionally important were rare. This excess of rare functional variants is due to the combined effects of explosive, recent accelerated population growth and weak purifying selection. Furthermore, we show that large sample sizes will be required to associate rare variants with complex traits.  相似文献

The Caloris basin, the youngest known large impact basin on Mercury, is revealed in MESSENGER images to be modified by volcanism and deformation in a manner distinct from that of lunar impact basins. The morphology and spatial distribution of basin materials themselves closely match lunar counterparts. Evidence for a volcanic origin of the basin's interior plains includes embayed craters on the basin floor and diffuse deposits surrounding rimless depressions interpreted to be of pyroclastic origin. Unlike lunar maria, the volcanic plains in Caloris are higher in albedo than surrounding basin materials and lack spectral evidence for ferrous iron-bearing silicates. Tectonic landforms, contractional wrinkle ridges and extensional troughs, have distributions and age relations different from their counterparts in and around lunar basins, indicating a different stress history.  相似文献

Parasitic filarial nematodes infect more than 200 million individuals worldwide, causing debilitating inflammatory diseases such as river blindness and lymphatic filariasis. Using a murine model for river blindness in which soluble extracts of filarial nematodes were injected into the corneal stroma, we demonstrated that the predominant inflammatory response in the cornea was due to species of endosymbiotic Wolbachia bacteria. In addition, the inflammatory response induced by these bacteria was dependent on expression of functional Toll-like receptor 4 (TLR4) on host cells.  相似文献

Operational airborne and satellite remote sensing in agriculture remains constrained by matching platform availability to suitable daytime weather and illumination conditions, crop development, and availability of ground staff. An ultra low-level aircraft carrying an active NIR/Red CropCircle™ sensor was successfully deployed to record and subsequently map crop vigour via the simple ratio (SR) index over a field of sorghum. Given the logging frequency of ≈20 Hz and the presence of alternate rows of bare soil, the Moiré effect reduced the contrast between crop and bare soil skip-rows. Such effects would not be expected to occur in non-skip-row crops. The ultra low-level airborne (ULLA)-SR map derived from the 20 m transect records compared favorably with the SR map derived from a meter-resolution airborne digital multispectral image that was re-sampled to a similar spatial resolution. This case study, involving a CropCircle™ sensor mounted in a low-level aircraft demonstrates another deployment option for users of this class of sensor. Moreover, an ULLA configuration offers the potential for greater flexibility in scheduling compared to airborne imaging, given it can be flown at any sun-angle, under cloud, at night, and may easily be incorporated into aircraft already conducting low-level operations, for example crop dusting and reconnaissance, over agricultural fields.  相似文献
