Abstract Tropical forests are very rich in biological diversity and form an important economic and ecological resource. This biodiversity is of great value for communities living in or near these forests as a ready source of subsistence and cash income, and for the world at large as a source of tropical timber and non-timber products and a repository of genetic and chemical information. However, this biological complexity is diminishing rapidly. We analyze the main human (anthropogenic) actions causing loss of tropical forest biodiversity along with the strategies of management for forest biodiversity. Not only is the biophysical component important in management for biodiversity, but the active participation and support of local people, national governments, and the international community as a whole are essential to effective and sustainable development of tropical forests. Campylobacter is the most commonly reported bacterial cause of enteritis in humans in the EU Member States and other industrialized countries. One significant source of infection is broilers and consumption of undercooked broiler meat. Campylobacter jejuni is the Campylobacter sp. predominantly found in infected humans and colonized broilers. Sequence analysis of the 16S rRNA gene is very useful for identification of bacteria to genus and species level. The objectives in this study were to determine the degree of intraspecific variation in the 16S rRNA genes of C. jejuni and C. coli and to determine whether the 16S rRNA sequence types correlated with genotypes generated by PFGE analysis of SmaI restricted genomic DNA of the strains.
Methods
The 16S rRNA genes of 45 strains of C. jejuni and two C. coli strains isolated from broilers were sequenced and compared with 16S rRNA sequences retrieved from the Ribosomal Database Project or GenBank. The strains were also genotyped by PFGE after digestion with SmaI.
Results
Sequence analyses of the 16S rRNA genes revealed nine sequence types of the Campylobacter strains and the similarities between the different sequence types were in the range 99.6–99.9%. The number of nucleotide substitutions varied between one and six among the nine 16S rRNA sequence types. One of the nine 16S rRNA sequence profiles was common to 12 of the strains from our study and two of these were identified as Campylobacter coli by PCR/REA. The other 10 strains were identified as Campylobacter jejuni. Five of the nine sequence types were also found among the Campylobacter sequences deposited in GenBank. The three 16S rRNA genes in the analysed strains were identical within each individual strain for all 47 strains.
Conclusion
C. jejuni and C. coli seem to lack polymorphisms in their 16S rRNA gene, but phylogenetic analysis based on 16S rRNA sequences was not always sufficient for differentiation between C. jejuni and C. coli. The strains were grouped in two major clusters according to 16S rRNA, one cluster with only C. jejuni and the other with both C. jejuni and C. coli. Genotyping of the 47 strains by PFGE after digestion with SmaI resulted in 22 subtypes. A potential correlation was found between the SmaI profiles and the 16S rRNA sequences, as a certain SmaI type only appeared in one of the two major phylogenetic groups. 相似文献
Disease has become an increasingly important issue for wildlife management over the past two decades. Adequate surveillance is fundamental for disease prevention and control, thus there is an increasing need for diagnostic assays for wildlife management. The objective of this study was to evaluate the performance of a field-portable biosensor adapted for rapid detection of specific antibodies in tortoise plasma that reflect a history of exposure to Mycoplasma agassizii, which is an agent of tortoise upper respiratory tract disease. Banked plasma samples were tested in two blinded trials, and the parameters that define the reliability of a diagnostic test were estimated based on externally validated tortoise plasma controls. The mean sensitivity of the biosensor (ability to identify exposed tortoises in the group of all exposed individuals) was 78%; the mean specificity (unexposed individuals with negative test result, out of all unexposed individuals tested) was 73%; the mean positive predictive value (exposed individuals with positive test, out of all individuals with positive test) was 82%; the mean negative predictive value (unexposed individuals with negative test, out of all individuals with negative test) was 68%. In a 15-min field-portable format, the biosensor was able to discriminate between true seropositive (n=34) and true seronegative (n=23) tortoise plasma with overall accuracy of 84%. The goals established for the tortoise population can help managers decide whether potential diagnostic errors should impact management decision-making, and whether the benefits of the field-portable format of the biosensor assay outweigh any potential disadvantages. 相似文献
An Internet-based questionnaire among horse owners was carried out to identify factors affecting the incidence of insect bite hypersensitivity (IBHI) among horses in the Netherlands. Information was obtained for 794 horses of various breeds, but the breed distribution was not representative for the Dutch horse population. Of the horses for which information was available, 56% suffered from IBH and 44% did not. The most common clinical symptoms were pruritus, scaling, and hair loss, occurring mainly at the base of the tail and along the mane. Breed, age, region (and local habitat), stabling, type of bedding in the stable, use of the horse, deworming frequency, and season were associated with significant differences in IBH incidence. Knowledge of the factors influencing the incidence of IBH may make it possible to reduce the number of animals affected and help alleviate symptoms in affected horses. 相似文献
This study aimed to compare feed efficiency measures of Nellore beef cattle on different residual intake and gain (RIG) classes. We used data from 610 animals weighing on average 236.33 kg and average of 283 days of age from feedlot performance tests carried out between 2005 and 2012. Animals were grouped based on RIG into three different classes: high RIG (>?mean?+?0.5 standard deviation (SD), most efficient; n?=?193), medium RIG (mean?±?0.5 SD; n?=?235), and low RIG (<?mean – 0.5 SD, least efficient; n?=?182). Residual feed intake (RFI), residual gain (RG), feed conversion ratio (FCR), feed efficiency (FE), relative growth rate (RGR), and Kleiber ratio (KR) of animals in each RIG class were compared by Tukey test at 1% of probability. Phenotypic correlations between variables were evaluated as well. Animals on high RIG class showed lower dry matter intake (P?<?0.01) and higher average daily gain (P?<?0.01) than low RIG animals. Consequently, high RIG animals had lower FCR (P?<?0.01) and higher FE (P?<?0.01) than those animals in low RIG class. The most efficient animals based on RIG were also the most efficient animals based on RG and RFI. RIG was negatively correlated to dry matter intake (P?<?0.01) and FCR (P?<?0.01), and a positive correlation was found between RIG and FE (P?<?0.01). Therefore, RIG appears to be a good parameter to select animals with reduced dry matter intake and high productive performance.
Blood lactate measurements are commonly used in exercising horses to determine the onset of lactate accumulation and in colic patients to assess clinical status and to indicate prognosis. To study the usability of a portable blood lactate meter based on dry chemistry (Lactate Pro), the data from this instrument were compared to data from a laboratory-used lactate meter based on wet chemistry (ABL 605 blood gas analyzer [ABL]). Heparinized blood samples were obtained from horses participating in a jumping experiment (n = 9), from horses cantering at maximal speed on a racetrack (n = 7), and from patients admitted to the Department of Equine Sciences at Utrecht University for severe colic (n = 13). Seventeen of these samples were tested in duplicate on both instruments to determine the repeatability of the measurements. Blood lactate concentrations measured with the Lactate Pro ranged from 0.8-17.6 mmol/liter and with the ABL from 1.0-18.6 mmol/liter. The correlation between lactate concentrations obtained using the Lactate Pro and values from the ABL was 0.90, and the relationship was represented by the following formula: y = 0.90 . x + 0.36, indicating a linear relationship between values produced by the ABL and Lactate Pro. The repeatability for the Lactate Pro was high (0.997), which is comparable to the ABL (0.999). 相似文献
To study whether airborne pollution can be detected in overbank sediments, samples collected from five overbank sediment profiles in eastern Finnmark, Norway, at 1 cm depth intervals, were subjected to chemical analysis and 210Pb dating. The studied drainage basins constitute parts of an area polluted by emissions from two Ni-Cu smelters in Russia. In the most polluted catchment area, the surface sample showed a 5-fold higher Ni concentration and a 3-fold higher Cu concentration than the pre-industrial sediments at depth. The increases started at the same time as the smelters. Slight Ni increases were also detected in the neighbouring drainage basin, while no significant concentration change was seen in drainage basins previously shown to be only weakly influenced by the smelter emissions. In the most polluted drainage basin, the increase in Ni accumulation rate did not equal the airborne deposition rate. Selective surface erosion of fine grained particles with adhering airborne Ni has probably caused excess Ni accumulation in both overbank and lake sediments. On the contrary, opening of minerogenic point sources may dilute the pollutant concentrations in the drainage sediments. Thus, dating of the sediment profiles is necessary to determine the airborne pollutant accumulation rates. However, dating is not necessary to map the resultant concentration increase, that may show the increased exposure of humans and biota in contact with the sediments. 相似文献
Thirty-five young cats were studied by echocardiography from the 2nd to 12th weeks of life to analyze correlation between body weight, body surface area, age and heart rate with fourteen echocardiographic parameters. There was a positive linear correlation (r = 0.49-0.78) between the independent variables (body weight, body surface area, age) and left ventricular wall thickness and diameter, aortic diameter and left atrial diameter, whereas there was a negative correlation (r = -0.39 and r = -0.43) between the heart rate and left ventricular diameter during systole and diastole. No linear dependence of the fractional shortening, ejection fraction, percentage thickening of the interventricular septum and left ventricular posterior wall, LA/AO ratio, and the ratio IVSED/LVWED to the independent variables was observed. 相似文献
