A DNA vaccine against contagious agalactia was developed for the first time, encoding the P48 of Mycoplasma agalactiae. Specific immune responses elicited in BALB/c mice were evaluated. Both total IgG and IgG1 were detected in mice vaccinated with pVAX1/P48. Proliferation of mononuclear cells of the spleen, levels of gamma interferon, interleukin-12, and interleukin-2 mRNAs were enhanced in immunized animals. Results indicate that pVAX1/P48 vaccination induced both Th1 and Th2 immune responses. Nucleic acid immunization could be a new strategy against M. agalactiae infections and may be potentially used to develop vaccines for other Mycoplasma diseases. 相似文献

Impaired insulin sensitivity is increasingly recognised in cats, but sequences of genes involved in insulin-signalling are largely undetermined in this species. In this study, extended feline mRNA sequences were determined for the adiponectin, glucose transporter-1 (GLUT1), GLUT4, peroxisome proliferative activated receptor-gamma1 (PPARgamma1), PPARgamma2, plasminogen activator inhibitor-1 (PAI-1), monocyte chemoattractant protein-1 (MCP-1) and insulin receptor genes. Conserved dog-specific primers identified from human-dog mRNA alignments were used to amplify feline cDNA in the polymerase chain reaction (PCR). The feline sequences determined by this method were used to design feline-specific primers suitable for real-time PCR for quantification of gene expression in insulin sensitive tissues of healthy cats. Partial sequences of feline mRNAs had 86-95% identity with dog and human genes. Expression of adiponectin, GLUT1, GLUT4, PPARgamma1, PPARgamma2, PAI-1 and insulin receptor mRNA was detected and quantified in subcutaneous and visceral fat and skeletal muscle, whereas MCP-1 mRNA was detected in adipose tissue but not in skeletal muscle. Further characterisation of genes related to glucose metabolism in cats will provide additional insights into insulin-signalling mechanisms in this species. 相似文献

A 6-year-old intact female corn snake (Elaphe guttata guttata) was presented with a 3-week history of anorexia. Coelomic radiographs revealed a 9 x 4 cm soft tissue opacity suggestive of a right ovarian enlargement. The mass (9 x 5 x 4 cm) was surgically removed, and multiple smears from tissue sections were stained with Diff-Quik. Multiple tissue samples from the mass were collected and fixed in formalin. Cytologic specimens were moderately cellular and contained light pink amorphous background material. The cells were primarily spindle-shaped with moderate to marked anisocytosis and anisokaryosis. Cells sometimes were round to polygonal, and rarely were arranged in small clusters. Macrophages occasionally were observed. Histologic specimens consisted of a highly cellular mass composed of pleomorphic, spindle-shaped cells and, occasionally, round to polygonal cells arranged in irregular fascicles. The neoplastic cells were immunoreactive for cytokeratin (AE1/AE3), smooth muscle actin, and skeletal muscle actin, but did not stain for vimentin or desmin. On the basis of the morphologic and immunohistochemical results, a diagnosis of ovarian undifferentiated carcinoma was made. In this report, we describe the challenges of using immunohistochemistry to diagnose this uncommon type of tumor in reptiles. 相似文献

BACKGROUND: The increased sophistication of imaging techniques in veterinary medicine allows the detection of a wide variety of intracranial and intraspinal lesions; however, imaging often does not provide a definitive diagnosis for nervous system (NS) lesions. Cytology is emerging as a useful diagnostic tool for obtaining a fast and accurate assessment of NS lesions, but little information is available for dogs and cats. OBJECTIVES: The purpose of this study was to assess the accuracy of cytologic evaluation of squash samples from NS lesions in dogs and cats and to consider cytology-based diagnostic guidelines and sources of misdiagnosis. METHODS: Cytologic specimens from masses localized in the central and peripheral NS taken during surgery or postmortem examination were classified into 3 groups according to the final histopathologic diagnosis: Group 1 = completely correct diagnosis, when the cytologic diagnosis and final histologic diagnosis were exactly correlated; Group 2 = partial correlation, when the cytologic diagnosis only partially correlated with the final histologic diagnosis, and Group 3 = no correlation, when the cytologic diagnosis was incorrect and there was no correlation with the general histologic type of lesion. The diagnostic accuracy of cytopathology was calculated by considering the histopathologic diagnosis as the "gold standard," and calculating a 95% confidence interval (CI). RESULTS: A total of 42 animals (33 dogs and 9 cats) were included in the study. The cytologic diagnoses were classified in Group 1 for 32 cases (76%; 95% CI 0.63-0.89), in Group 2 for 6 cases (14%; 95% CI 0.04-0.25), and in Group 3 for 4 cases (10%; 95% CI 0.006-0.18). Considering both complete and partial correlation as an adequate result, cytologic diagnosis was satisfactory in 90% of biopsies. CONCLUSIONS: Although the current series of cases is relatively small, cytologic evaluation of squash preparations can be considered a fairly accurate and reliable tool in the diagnosis of NS lesions. 相似文献

BACKGROUND: Artifactual changes in blood may occur as a consequence of delayed analysis and may complicate interpretation of CBC data. OBJECTIVE: The aim of this study was to characterize artifactual changes in canine blood, due to storage, using the ADVIA 120 hematology analyzer. METHODS: Blood samples were collected into EDTA from 5 clinically healthy dogs. Within 1 hour after blood sample collection and at 12, 24, 36 and 48 hours after storage of the samples at either 4 degrees C or room temperature (approximately 24 degrees C), a CBC was done using the ADVIA 120 and multispecies software. A linear mixed model was used to statistically evaluate significant differences in values over time, compared with initial values. RESULTS: The HCT and MCV were increased significantly after 12 hours of collection at both 4 degrees C and 24 degrees C, and continued to increase through 48 hours. The MCHC initially decreased significantly at 12-24 hours and then continued to decrease through 48 hours at both temperatures. Changes in HCT, MCV, and MCHC were greater at 24 degrees C than at 4 degrees C at all time points. A significant increase in MPV and a decrease in mean platelet component concentration were observed at all time points at 24 degrees C. Samples stored at 24 degrees C for 48 hours had significantly higher percentages of normocytic-hypochromic RBCs, and macrocytic-normochromic RBCs, and lower platelet and total WBC counts. CONCLUSIONS: Delayed analysis of canine blood samples produces artifactual changes in CBC results, mainly in RBC morphology and platelet parameters, that are readily detected using the ADVIA 120. Refrigeration of specimens, even after 24 hours of storage at room temperature, is recommended to improve the accuracy of CBC results for canine blood samples. 相似文献

A case of eosinophilic granulomatous gastroenterocolitis and hepatitis in a 1-year-old male Siberian Husky is described. The dog presented with a history of diarrhea, weakness, lethargy, and anorexia of several months' duration. Hematologic and biochemical examinations, abdominal ultrasonography, computer tomography, and exploratory laparotomy were performed. Histopathologic examination of full-thickness biopsies from the gastrointestinal tract and liver revealed the presence of eosinophilic granulomatous lesions in the submucosa and tunica muscularis of stomach, jejunum, ileum, colon, and liver. Infectious agents were not detected by light microscopic and electron microscopic examination or by immunohistochemistry. On the basis of the findings, it is concluded that the disease in this dog represents an unusual manifestation of chronic idiopathic inflammatory bowel disease. 相似文献

Ethanolic extract from the fruits of Pimpinella anisoides, an aromatic plant and a spice, exhibited activity against AChE and BChE, with IC50 values of 227.5 and 362.1 μg/ml, respectively. The most abundant constituents of the extract were trans-anethole, (+)-limonene and (+)-sabinene. trans-Anethole exhibited the highest activity against AChE and BChE with IC50 values of 134.7 and 209.6 μg/ml, respectively. The bicyclic monoterpene (+)-sabinene exhibited a promising activity against AChE (IC50 of 176.5 μg/ml) and BChE (IC50 of 218.6 μg/ml). 相似文献

Coastal erosion is a widespread phenomenon on sandy coasts throughout the Mediterranean region; along the Thyrrenian coast of Tuscany (Italy), stone pine (Pinus pinea L.) stands originally planted for the protection of agricultural crops further inland are often damaged. In the present study, a pairwise comparison of stands at different distance from the sea at eroded and control sites highlighted the effects of coastal erosion alone on pine growth and function. Dendroecological analyses made it possible to determine the temporal dynamics of the phenomenon since 1930 and the interactions with climate, whilst additional structural (LAI, sapwood area) and functional (carbon isotope discrimination) measurements were used to discriminate between stress mechanisms. Salty winds, exacerbated by the removal of dunal vegetation, were found to be the most likely cause of the observed growth decline. The presence and, in more recent times, the reduction of surfactants in sea water played an important synergistic effect. The intrusion of salty water in the water table, on the contrary, played a marginal role at the site. Finally, stressed trees were more sensitive to the inter-annual variability in precipitation; at all sites, growth was stimulated by June, November and December precipitation in the current and two preceding years. 相似文献

A 'step-by-step' method was used to develop a simplified procedure for calculating pesticide residue levels on fruit at harvest by considering the application of the compound and the relevant routes of loss. The model is applicable to cases where the most important exposure route is by direct spray to the canopy of the crop and where uptake into the plant by the roots can be disregarded. The exposure dose is calculated by considering the proportion of total crop cover represented by the fruits. The loss processes considered are photodegradation, uptake, volatilization and washoff.The outputs of the model were compared with measured residues of pesticides on pear. Analysis of the model fit demonstrates that the model predicted the measured data with a good level of accuracy for four of seven investigated pesticides. The predicted/observed quotients are close to 1, as is the modelling efficiency, and there are no great differences between the predicted and observed values. Taking into account the extreme simplicity of the model and the complexity of the environmental processes considered, these results encourage further research into the modelling of residue behaviour in food commodities.The objectives of this work were to produce a tool to predict pesticide residues in products of plant origin, to complement monitoring of pesticide levels and to be useful in evaluating the effect of government policies on food safety. All predicted values were below the maximum levels fixed for pesticide residues in pear, as amended in Council Directives 86/362/EEC and 90/642/EEC. Loss of zoospores has happened independently several times in different phylogenic lines and has, it is claimed, no major
phylogenetic significance. But whether or not, how, and under which conditions plant pathogens retain the ability to produce
motile asexual spores has fundamental importance from an ecological and epidemiological perspective. Recent molecular investigations
of the early evolution of fungi and oomycetes are shedding light on the issue of zoospore loss in organisms able to cause
plant diseases. Zoospore loss may have accompanied the development of new forms of dispersal adapted to the terrestrial environment,
or the simplification processes which often follow the shift to parasitic or biotrophic life-forms. In this review we consider
hybridisation events between Phytophthora species, long distance dispersal of oomycetes, sporangia and zoospore survival, direct and indirect infection processes and
newly observed sporulating structures. These aspects are all relevant features for an understanding of the epidemiology of
zoosporic plant pathogens. Disease management should not be based on the presumption that the zoosporic stage is a weak link
in the life cycle. Oomycete plant pathogens show remarkable flexibility in their life cycles and ability to adapt to changing
environmental circumstances.
