Dripper clogging is a major drawback of microirrigation systems that must be addressed to improve their efficiency and durability. Particle-induced clogging is first studied in situ. The experiments consist in observing in real conditions the behavior of a series of drippers fitted on an agricultural plot in the south of France. The plot is supplied from a canal with Durance River water. The latter is loaded with sediments that gradually clog drippers and filters. Water analysis reveal that physicochemical clogging prevails over biological clogging. This characterization helps in setting in-lab experiment protocol. Indeed, besides field observation of clogging, laboratory analyses of both the irrigation water and the clogging material are performed with reactive and inert clay: smectite and an illite–calcite mix. A surprising tendency is observed: Salt concentration in smectite seeded water decreases the clogging, whereas it increases agglomerate size. Computational fluid dynamic simulations are carried out to investigate the impact of particles on flow behavior. Results demonstrate that clay particles interacting with the flow govern the complex structure of the fluid velocity fields inside the dripper labyrinth channel. 相似文献
At the field scale, the co-existence of different farming production systems requires strategies to prevent gene flow between adjacent crops. Oilseed rape produces pollen dispersed by wind and insects and the risks of pollen mediated gene flow are significant for this crop. Cleistogamy, the trait of non-opening flowers, could be used to reduce pollen flow. Cleistogamous oilseed rape genotypes were obtained by INRA in France and were bred in order to improve the stability of this trait. In this paper, we examine the reliability of the cleistogamous trait for two inbred lines. The flower opening level was measured at different dates during the flowering period in six field experiments (three sites × 2 years). The results showed that some flowers were partially opened with rates varying from 0.5% to 33% principally depending on genotypes, trials (site and year) and recording dates. Given that other studies have shown that cleistogamy could reduce pollen dispersal, we consider that, even when partially unstable, cleistogamy could be beneficially used in combination with other means in a containment strategy. 相似文献
The fungus Cladosprium fulvum is a biotrophic leaf pathogen of tomato. The fungus develops in the intercellular space without forming specialized feeding structures and does not affect the leaf tissue. The outcome of the C. fulvum-tomato interaction can be described by the gene-for-gene model. Failure of infection is expressed by a hypersensitive response. Two fungal proteins, ECP1 and ECP2, have been isolated and their corresponding genes have been cloned. In a compatible interaction including many physiological races ECP1 and ECP2 are highly produced and a role in pathogenicity is suggestive. The ecp1 gene shows some homology with tumor necrosis factor receptors (TNFRs) while the ecp2 gene shows no homology with sequences known in data bases. However, disruption of one of the two genes showed no reduced pathogenicity of the fungus. Two race-specific elicitors, AVR4 and AVR9, have been isolated and their corresponding genes have been cloned. The avirulence genes Avr4 and Avr9 are only present in C. fulvum avirulent on Cf-4 and Cf-9 cultivars, respectively. The expression of these two genes is, like the expression of the ecp genes, highly induced when the fungus grows in planta. Disruption of the Avr9 gene in wild type avirulent races leads to virulence on tomato genotypes carrying the complementary resistance gene Cf-9. A single base-pair change in the avirulence gene Avr4 leads to virulence on tomato genotypes carrying the Cf-4 resistance gene. Field strains of Botrytis cinerea Pers ex Fr, the causal agent of grey mould diseases, were collected from French vineyards between 1993 and 2000. Several phenotypes have been characterized according to the inhibitory effects of fungicides towards germ-tube elongation and mycelial growth. Two types of benzimidazole-resistant strains (Ben R1 and Ben R2) could be detected; negative cross-resistance to phenylcarbamates (e.g. diethofencarb) was only found in Ben R1. Benzimidazole resistance was related to point mutations at codon 198 (Ben R1) or 200 (Ben R2) of the beta-tubulin gene. Most dicarboximide-resistant strains were also weakly resistant to aromatic hydrocarbon fungicides (e.g. dicloran) but remained sensitive to phenylpyrroles (e.g. fludioxonil). These resistant field strains (Imi R1) contained a single base pair mutation at position 365 in a two-component histidine kinase gene, probably involved in the fungal osmoregulation. Three anilinopyrimidine-resistant phenotypes have been identified. In the most resistant one (Ani R1), resistance was restricted to anilinopyrimidines, but no differences were observed in the amino-acid sequences of cystathionine beta-lyase (the potential target site of these fungicides) from Ani R1 or wild-type strains. In the two other phenotypes (Ani R2 and Ani R3), resistance extended to various other groups of fungicide, including dicarboximides, phenylpyrroles and sterol biosynthesis inhibitors. This multi-drug resistance was probably determined by over-production of ATP-binding cassette transporters. The hydroxyanilide fenhexamid is a novel botryticide whose primary target site is the 3-keto reductase involved in sterol C-4 demethylations. Apart from the multi-drug-resistant strain Ani R3, three other fenhexamid-resistant phenotypes have been recognized. For two of them (Hyd R1 and Hyd R2) fenhexamid-resistance seemed to result from P450-mediated detoxification. Reduced sensitivity of the target site could be the putative resistance mechanism operating in the third resistant phenotype (Hyd R3). Increased sensitivity to inhibitors of sterol 14 alpha-demethylase recorded in Hyd R1 strains was related to two amino-acid changes at positions 15 and 105 of this enzyme. 相似文献
A sheep-pox virus strain has been adapted and multiplied in primary lamb kidney cell cultures. The main characteristics of the strain have been verified in vitro after clones were isolated, and the results confirmed its identity. The safety and the potency of the strain have been investigated in sheep.
The inoculation of the strain to sheep was followed by a post-vaccinal reaction materialised by a nodule at the site of inoculation and an increase of temperature by about 1°C. No reactions adversely affecting pregnancy have been noted. Immunisation was demonstrated by an increase in the level of neutralising serous antibodies and protection against the pathogenic virus. The immunity tended to decrease during the second year after primovaccination and a yearly booster vaccination appeared to be necessary. Primovaccination of lambs over 2 months of age produced a better immunity, especially when the lambs were born from vaccinated ewes.
This strain forms the active principle of a freeze-dried vaccine containing no adjuvant of the immunity. 相似文献
Fusarium wilt, caused by Fusarium oxysporum, is a major disease of jojoba, causing serious economic losses. This study was aimed at characterizing the Fusarium populations associated with jojoba in Israel. Fifty Fusarium isolates used in this study included 23 isolates from the 1990s (“past”) and 27 recently isolated (“recent”). All the isolates were characterized by arbitrarily primed (ap)-PCR and 16 representatives were additionally delineated using multilocus (tef1, rpb1, rpb2) phylogeny and evaluated for their pathogenic potential. Consequently, 88% of the isolates were identified and characterized to the F. oxysporum species complex. The remaining 12% grouped within the F. fujikuroi, F. solani, and F. redolens species complexes. Variations in the infection rate (16.7%–100%), disease symptoms (0.08–1.25, on a scale of 0–3), and fungal colonization index (0.67–2.17, on a scale of 0–4) were observed within the tested isolates, with no significant differences between the past and recent isolates. The representative isolates were assigned to 11 groups based on ap-PCR. Pathogenicity tests showed that isolates from Groups II, IV, and V were the most aggressive, whereas isolates from Groups III, VIII, and IX were the least aggressive. Among the tested isolates, F. oxysporum sensu lato was the most aggressive, followed by F. proliferatum, while F. nygamai was the least aggressive. This study demonstrates the complexity and genetic diversity of Fusarium wilt on jojoba in Israel, indicating possible multiple introductions of infected germplasm into the country. 相似文献
Soil Bulk Density (BD) is an extremely important variable because it is an important site characterization parameter, and it is highly relevant for policy development because it is mandatory for calculating soil nutrient stocks. BD can influence soil chemical properties, land-use planning and agronomic management. The 2018 Land Use and Coverage Area Frame Survey (LUCAS) saw the unprecedented collection of BD core analysis in a subset of the locations in Europe and the United Kingdom where soil physical and chemical properties were analysed in the 2009 and the 2015 sampling campaigns. Here, we integrated the LUCAS 2018 BD sampling campaign with the mass fraction of coarse fragments previously determined in LUCAS 2009–2015 in order to provide a dataset of the volume fraction of coarse fragments and the BD of the fine earth and improve soil organic carbon (SOC) stock estimation accuracy for topsoil. BD data sampled at 0–10 and 10–20 cm were averaged to harmonize the BD with the mass fraction of coarse fragments measured in 2009, 2012 and 2015. Samples were from cropland, grassland and woodland soils, which accounted for 41%, 21% and 30%, respectively, of the total number of selected sites (n = 6059); ‘bareland’, and ‘shrubland’ accounted for 3% of the sites each, whereas ‘artificial land’ accounted for <1%. Only six samples were classified as ‘wetland’. The dataset was produced assuming the mass density of the coarse fraction to be constant across all LUCAS soil samples. We also estimated the SOC stocks associated with LUCAS 2018 BD and SOC content measurements and showed that correcting the BD by the coarse mass fraction instead of the coarse volume fraction generates SOC stock underestimation. We found the highest deviations in woodlands and shrublands. We showed that, when SOC stock is computed with coarse mass fraction, the error compared with the computation by volume may vary depending on the SOC and coarse mass fraction. This may imply a SOC stock underestimation for European soils. This dataset fits into the big framework of LUCAS soil properties monitoring and contributes both to soil awareness and soil research and assessments, which are two important objectives of the Soil Strategy and the European Soil Observatory (EUSO). 相似文献
