Objective  Microbial communities are a central component of trophic dynamics and biogeochemical processes on coastal systems, since most of the processes in sediments are mediated by microorganisms and carried out by enzymes. Microorganisms play a key role in decomposition processes in salt marsh sediments, although the significance of microbial dynamics is largely unexplored. A culture-dependent (Ecoplate) and a culture-independent (extracellular enzyme activity [EEA]) approaches were evaluated in their ability to distinguish the catabolic potential among sediments from Tagus estuary salt marshes with different proximities to anthropogenic sources. Methods  Ecoplate was used to analyse the salt marsh community-level physiological profiles (CLPPs). Results were expressed as the net area under the curve for each of the 31 response wells over a 3-day incubation period in two sediment horizons. The catabolic profiles for salt marsh samples were analysed by Principal Component Analysis (PCA) and hierarchic clustering methods. EEA was analysed by fluorescein diacetate (FDA) hydrolysis in two sediment horizons. The FDA is catalysed by extracellular enzymes, i.e. esterases, lipases and partially by proteases. Results were expressed as μg g−1d wt h−1. Results and Discussion  In this study the CLPP and EEA data were not generally correlated. In Corroios salt marsh only in surface sediments higher net areas corresponded to higher extracellular enzymatic activity, and in Alcochete deep sediments lower net areas corresponded to lower enzymatic activity. Although EEA profiles more directly reflect the inherent activity of resident community in each salt marsh sample, the CLPP profiles provide better assessments of diversity. Papillomaviruses (PVs) have been shown as being the etiologic agents of various benign and malignant tumours in many vertebrate species. In dolphins and porpoises, a high prevalence of orogenital tumours has recently been documented with at least four distinct novel species-specific PV types detected in such lesions. Therefore, we generated the immunological reagents to establish a serological screening test to determine the prevalence of PV infection in Atlantic bottlenose dolphins [(Tursiops truncatus (Tt)]. Using the baculovirus expression system, virus-like particles (VLPs) derived from the L1 proteins of two TtPV types, TtPV1 and TtPV2, were generated. Polyclonal antibodies against TtPV VLPs were produced in rabbits and their specificity for the VLPs was confirmed. Electron microscopy and enzyme-linked immunosorbent assay (ELISA) studies revealed that the generated VLPs self-assembled into particles presenting conformational immunodominant epitopes. As such, these particles are potential antigen candidates for a TtPV vaccine. Subsequently, the VLPs served as antigens in initial ELISA tests using sera from six bottlenose dolphins to investigate PV antibody presence. Three of these sera were derived from dolphins with genital tumour history and showed positive PV ELISA reactivity, while the remaining sera from lesion-free dolphins were PV antibody-negative. The results suggest that the developed screening test may serve as a potential tool for determining PV prevalence and thus for observing transmission rates in dolphin populations as the significance of PV infection in cetaceans starts to unfold.  相似文献

This study was designed to evaluate the effects of two feeding methods on milk yield, composition and fatty acid (FA) profile obtained from dual‐purpose cattle. Twenty‐four Aosta Red Pied cows beyond peak of lactation were assigned to two groups and fed hay and concentrates in the proportions 0.69 and 0.31 on a dry matter basis for 10 weeks. Concentrates were offered separately from forages 6 times a day (separate ration, SR) or as a total mixed ration (TMR). The feeding method did not significantly influence dry matter intake (16.8 vs. 16.9 kg/head/day for SR‐ and TMR‐fed cows, respectively), milk yield (17.4 vs. 17.5 kg/head/day), milk fat, protein and lactose contents (36.4 vs. 35.2, 33.5 vs. 32.8, and 47.3 vs. 47.4 g/kg) and yields (607.9 vs. 613.4, 567.4 vs. 572.7 and 805.5 vs. 829.7 g/head/day). The overall milk FA profile was very similar between groups. Milk concentrations of FA used as indirect markers of rumen function (C18:2 t10c12, odd‐ and branched‐chain FA) and the extent of ruminal biohydrogenation were comparable (P > 0.05) between SR‐ and TMR‐fed cows, suggesting that ruminal pH did not vary considerably as a consequence of the feeding strategy applied.  相似文献

The objective of this study was to evaluate the effects of supplementation strategy on performance, nutritional and metabolic characteristics of Nellore suckling female calves, on grazing Urochloa decumbens during the rainy-dry transition period. Forty-four Nellore female calves, averaging 147.6?±?1.34 kg (4 months), were used. A single supplement 20% of crude protein of dry matter was provided, consisting of daily supplement at 0.0, 0.2, 0.4, or 0.6% of body weight. A positive linear effect (P?<?0.05) on final body weight and average weight gain of female calves was observed with increased supplementation. Supplementation increased consumption, in kilograms per day, of dry matter (DM), organic matter (OM), crude protein (CP), digested dry matter, and total digestible nutrients (TDN), all calculated as % of dry matter. There was no effect of supplementation on the consumption in kilograms per day of neutral detergent fiber corrected for ash and protein (apNDF) (P?>?0.05). Supplementation increased, in a quadratic way, the total apparent digestibility coefficient of DM, OM, CP, apNDF, and TDN (P?<?0.05). There was no effect of supplementation (P?>?0.05) on non-esterified fatty acid concentrations. There was a positive linear effect (P?<?0.05) of supplementation on insulin concentrations. In this study, it is concluded that linearly increasing the feeding level of a supplement with 20% crude protein (% of DM) in the range of 0.2–0.6% of body weight improves the performance, nutritional and metabolic characteristics of the animals, considering these pasture characteristics.  相似文献

Canine leishmaniasis (CL) is a systemic parasitic disease with a wide variability of response to specific therapy: the majority of patients apparently improve with treatment, some of them respond but later relapse, and few of them do not respond at all. It has been demonstrated that the immune response plays a key role in the development and outcome of Leishmania infection in the dog and in the response to the treatment, although this response is not well understood. Some authors have suggested that ill dogs show a reduction in the percentage of circulating CD4+ lymphocytes and in the CD4+/CD8+ ratio, both of which normalize after treatment and clinical recovery. The present paper discusses the variation of the different lymphocyte subpopulations (CD3, CD4, CD8, CD21) of peripheral blood mononuclear cells (PBMC) in 28 dogs diagnosed with CL and submitted to conventional treatment with meglumine antimoniate (Glucantime) for 1 month and with allopurinol (Zyloric) for 1 year, in order to evaluate the usefulness of these parameters as indicators of the immunological condition of the ill animals and of the prognosis of their evolution during the treatment. It is concluded that circulating lymphocyte subpopulations are similar in dogs with leishmaniasis and in healthy dogs and that there is no correlation between the clinical status or response to therapy and the values of the counts of the different lymphocyte subpopulations. Therefore, the percentage of different lymphocyte subpopulations cannot be used as a parameter to predict the evolution of an individual patient in a clinical context.  相似文献

In North America, the only endemic focus for Angiostrongylus vasorum (French heartworm) was historically thought to occur in the southeastern part of the island of Newfoundland. However, reports of A. vasorum infection in wild canids in West Virginia, USA, and Nova Scotia, Canada, suggest the introduction of the parasite to mainland North America. We screened for A. vasorum in coyotes from across southern Ontario. Additionally, we evaluated the performance of ELISAs for detection of circulating A. vasorum antigen (Ag-ELISA) and antibodies against A. vasorum (Ab-ELISA) designed for use in sera or blood of foxes for use with coyotes in this region. Autopsies were performed on 397 coyotes, and lung tissue extract prepared from each carcass was tested via both ELISAs. The sensitivity and specificity for both tests were estimated in the absence of a gold standard using a 2-test single population Bayesian model; sensitivity and specificity priors were based on the performance of the assays in foxes in Switzerland. Eight coyotes tested positive for A. vasorum antigen; no animal was antibody positive. The estimated sensitivity and specificity of the Ag-ELISA were 90.8% (95% credible interval [CrI]: 83.8–95.6%) and 95.5% (95% CrI: 93.4–97.2%), respectively. For the Ab-ELISA, the estimated sensitivity and specificity were 41.9% (95% CrI: 32.1–51.9%) and 98.0% (95% CrI: 96.3–99.0%), respectively. Based on these findings and negative postmortem data for the same animals, there is insufficient evidence to suggest the presence of A. vasorum in southern Ontario coyotes.  相似文献

In dogs, a mean broncho‐arterial ratio of 1.45 ± 0.21 has been previously defined as normal. These values were obtained in dogs under general inhalational anesthesia using a single breath‐hold technique. The purpose of the study was to determine whether ventilation technique and bronchial diameter have an effect on broncho‐arterial ratios. Four healthy Beagle dogs were scanned twice, each time with positive‐pressure inspiration and end expiration. For each ventilation technique, broncho‐arterial ratios were grouped into those obtained from small or large bronchi using the median diameter of the bronchi as the cutoff value. Mean broncho‐arterial ratios obtained using positive‐pressure inspiration (1.24 ± 0.23) were statistically greater than those obtained at end expiration (1.11 ± 0.20) P = 0.005. There was a strong positive correlation between bronchial diameter and broncho‐arterial ratios for both ventilation techniques (positive‐pressure inspiration rs = .786, P < 0.0005 and end expiration rs = .709, P < 0.0005). Mean broncho‐arterial ratio for the large bronchi obtained applying positive‐pressure inspiration was 1.39 cm ± 0.20 and during end expiration was 1.22 cm ± 0.20. Mean broncho‐arterial ratio for the small bronchi obtained during positive‐pressure inspiration was 1.08 cm ± 0.13 and during end expiration was 1.01 cm ± 0.13. There was a statistically significant difference between these groups (F = 248.60, P = 0.005). Findings indicated that reference values obtained using positive‐pressure inspiration or from the larger bronchi may not be applicable to dogs scanned during end expiration or to the smaller bronchi.  相似文献
