New icetexane diterpenes (1-2); 8, 11, 13-icetexatriene-10-hydroxy, 11, 12, 16-tri acetoxyl (1) and 8, 11, 13-icetexatriene-7, 10, 11-dihydroxy-12, 13-dihydrofuran (2) along with six known compounds namely acetoxy syranzaldehyde (3), syranzaldehyde (4), coniferaldehyde (5), lupeol (6), betulin (7), and 4-(4-methoxy phenyl)-2-butanone (8) were isolated from the roots of Premna tomentosa. The structures of compounds 1 and 2 were established by detailed spectral analysis using UV, IR, 1H NMR, 13C NMR, 1D, 2D and Mass. The newly isolated compounds were screened for rat intestinal α-glucosidase inhibitory and free radical (DPPH) scavenging potentiality. The new icetexane diterpenes (1, 2) and compound 3 were found to have significant α-glucosidase inhibitory and also free radical scavenging (DPPH) activities.  相似文献

Analysis of differential pore-volume distribution (DPVD) patterns of a commercial Clinoptilolite has been conducted experimentally using an analyzer by measuring the N2 adsorption isotherm. The commercial material was fractionated by sieving through sequential sieves: <125 µ (Z8; fine), 125–250 µ (Z9; medium), and >250 µ (Z10; coarse). The DPVD of the fractions were calculated from the hysteresis loop according to the adsorption and desorption curves of Dollimore-Heal (DH) model. The adsorption cycle had produced dissimilar differential pore volume distribution patterns. For the 10-nm pore width, fine and medium fractions had a maximum pore volume up to 0.0016 cm3/g nm, while the coarse fraction had a little greater value up to 0.002 cm3/g nm. Moreover, the medium fraction had too many hills and valleys in the DPVD. However, the desorption cycle–based DH analysis did not show any marked variation in the DPVD pattern.  相似文献

Platelets are generated from megakaryocytes (MKs) in mammalian bone marrow (BM) by mechanisms that remain poorly understood. Here we describe the use of multiphoton intravital microscopy in intact BM to visualize platelet generation in mice. MKs were observed as sessile cells that extended dynamic proplatelet-like protrusions into microvessels. These intravascular extensions appeared to be sheared from their transendothelial stems by flowing blood, resulting in the appearance of proplatelets in peripheral blood. In vitro, proplatelet production from differentiating MKs was enhanced by fluid shear. These results confirm the concept of proplatelet formation in vivo and are consistent with the possibility that blood flow-induced hydrodynamic shear stress is a biophysical determinant of thrombopoiesis.  相似文献

The hatching and mortality response of 0- to 48-h-old eggs of field strains of the stored-product insects Cryptolestes ferrugineus (Stephens), Lasioderma serricorne (F) and Oryzaephilus surinamensis (L) following phosphine fumigation for 24, 48 or 120 h at 27 (+/- 2) degrees C was investigated. Hatching was delayed and reduced in the first few days in a phosphine-resistant strain of C ferrugineus that was treated with 2.0-7.0 mg litre(-1) doses for 48 h (5-80% mortality) and with 1.0-2.0 mg litre(-1) for 120 h (44-84% mortality). In both the exposures there were significant increases in hatching on later days when compared with the corresponding controls. Developmental delay was, however, not evident in susceptible strains of C ferrugineus, L serricorne and O surinamensis that were exposed to phosphine for 24 h.  相似文献

Concerns have been raised that the amphibian larval stages are particularly at risk and may be vulnerable to adverse effects of pesticides. The present study reports acute toxicity of cypermethrin at 24, 48, 72 and 96 h through static renewal bioassay test for Duttaphrynus melanostictus. The LC50 values were 5.15, 4.55, 3.95, and 3.34 μg/L for 24, 48, 72, and 96 h respectively. At sublethal concentration (0.33 μg/L) behavioral, morphological and biochemical changes were studied. The behavioral and morphological anomalies observed in the present study are typical signs of cyano pyrethroid poisoning. Significant changes were observed in total, soluble, and structural proteins. The depletion of all the protein fractions observed in this investigation led to progressive protein oxidation and catabolism of proteins. Decreased protein level has resulted in a marked elevation of free amino acid levels at all time intervals. The induction of catalase, glutathione-S-transferase activities and elevation in the levels of hydrogen peroxide, reduced glutathione, and malondialdehyde eventually lead to oxidative damage of biomolecules, showing that the generation of reactive oxygen species and oxidative stress are involved in the toxicity induced by cypermethrin. Indicating increased susceptibility of tadpoles. Thus, an exposure to cypermethrin at sublethal concentration had catastrophic effect on tadpoles of D. melanostictus.  相似文献

Summary Semilooper resistant transgenic castor plants were produced through Agrobacterium-mediated genetic transformation method. Two castor cultivars, Jyothi and VP1 were transformed using the super-binary vector pTOK233 carrying gus A and hpt genes. Putative transformants were regenerated following selection on the hygromycin containing medium. GUS positive primary transformants, when subjected to Southern analysis, revealed stable integration of gus A into their genomes. In the T1 generation, a monogenic segregation ratio of 3 GUS positive: 1 GUS negative plants was observed. Furthermore, transformation experiments were carried out with the Agrobacterium pSB111 super-binary vector carrying a synthetic delta endotoxin gene cryIAb and the herbicide resistance gene bar both driven by cauliflower mosaic virus 35S promoter. Putative transformants were regenerated through selection on the phosphinothricin containing medium and Basta tolerant transformants were subjected to molecular analysis. PCR analysis revealed the presence of both bar and cryIAb genes in the Basta tolerant primary transformants. Southern analysis of PCR positive plants with cryIAb probe showed a 3 Kb band upon HindIII digestion and a > 6 Kb band with BamHI digestion, thus suggesting stable integration of cryIAb intact expression cassette and independent nature of the transformants. The primary transformants subjected to ELISA disclosed varied levels of Cry protein. These transgenics expressing cryIAb – when bioassayed against freshly hatched semilooper larvae – induced substantial (> 88%) insect mortality. Southern analysis of 2T1 plants revealed the presence of cryIAb gene, indicating stable inheritance of the transgene into the next generation. In T1, all the Southern-positive plants for cryIAb invariably exhibited tolerance to Basta, denoting co-segregation of both bar and cryIAb genes. Transgenics, expressing cryIAb exhibited ample resistance against the castor semilooper.  相似文献
