(1) Background: G protein-coupled inward-rectifier potassium (GIRK) channels, especially neuronal GIRK1/2 channels, have been the focus of intense research interest for developing drugs against brain diseases. In this context, venom peptides that selectively activate GIRK channels can be seen as a new source for drug development. Here, we report on the identification and electrophysiological characterization of a novel activator of GIRK1/2 channels, AsKC11, found in the venom of the sea anemone Anemonia sulcata. (2) Methods: AsKC11 was purified from the sea anemone venom by reverse-phase chromatography and the sequence was identified by mass spectrometry. Using the two-electrode voltage-clamp technique, the activity of AsKC11 on GIRK1/2 channels was studied and its selectivity for other potassium channels was investigated. (3) Results: AsKC11, a Kunitz peptide found in the venom of A. sulcata, is the first peptide shown to directly activate neuronal GIRK1/2 channels independent from Gi/o protein activity, without affecting the inward-rectifier potassium channel (IRK1) and with only a minor effect on KV1.6 channels. Thus, AsKC11 is a novel activator of GIRK channels resulting in larger K+ currents because of an increased chord conductance. (4) Conclusions: These discoveries provide new insights into a novel class of GIRK activators.  相似文献

We evaluated the effect of supplementation of a wheat bran (WB) diet with fresh carrots (FC) or wet brewer’s grains (WBG) on the growth performance and chemical composition of Tenebrio molitor larvae. Additionally, a digestibility trial was performed to determine the nutritional value of the raw materials used. The control diet was based on WB-only. Two other diets were formulated in which WB was supplemented with FC (FC diet) or with WBG (WBG diet). The experiment was conducted in trays (12 per treatment) and lasted 90 d. Larval weight, feed intake, and excreted feces were controlled in each tray the experiment. The digestibility trial was performed from 48 to 62 d post-hatch. Results showed digestibility coefficients of ashes, crude protein, and gross energy were significantly higher in FC diet compared with the other diets. Consequently, both digestible energy (DE) and digestible protein (DP) contents of FC were also significantly higher than those obtained for WB and WBG diets (on av. +1.12 megajoule [MJ] DE and +9.15 g DP per kg dry matter [DM]; P < 0.003). Mealworms fed FC diet showed significantly higher final weight and average daily gain than those fed the WBG diet (+12.4%), being higher in WBG than in WB diet (+3.5%). Dietary treatment did not affect DM, ashes, ether extract, and crude protein content of mealworms obtained. Some dietary effects on amino acid and fatty acid composition were observed. This study provides novel data and a unique experimental approach to assess the nutritional value of raw materials in mealworms.  相似文献

Increased flour yield in hard wheat is associated with increased endosperm rheology index, calculated from strength and stiffness as measured by the SKCS. A study of the fractured endosperm of hard wheat varieties grouped according to similar rheology index values was performed using environmental scanning electron microscopy (ESEM). Differing microstructures and fracture patterns were observed between each group. Specifically, the group representing high rheology index had a greater concentration of small starch granules in prismatic cells. Samples of diverse wheat germplasm were grown at two sites and subjected to laboratory milling. Starch granule size distribution (SGSD) analysis using a laser diffraction method was undertaken on a subset of samples in triplicate representing a range in flour yield. The results supported an hypothesis for a significant influence of SGSD on flour yield of hard wheat varieties. In addition, a significant part (R2>0.40 (p<0.05) at two sites) of the association appeared to be under genetic control. Results indicate a more even gradation of distributions involving an increase in the sample volume % of small granule (types B and C) and decrease in type A granules. This was associated with increased rheology index values and higher flour yield. The ratio of type A:C starch granules accounted for up to 58% (p<0.05) of the variation in flour yield in the samples studied. Thus, rheological parameters measured using a rapid SKCS screening method can be linked to the genetic regulation of SGSD with implications for the improvement of commercial processing performance of hard wheat.  相似文献

The indigenous pitanga (Eugenia uniflora L.) is now marketed and commercially processed in Brazil. In the present work, the carotenoids of the pitanga fruit from two states and at two stages of ripening, as well as of processed pitanga products (frozen pulp and juice, the brands being designated as A, B and C) commercialized in Campinas, São Paulo, were determined by HPLC. As compared to ripe pitanga from Medianeira, Paraná, those from Campinas had significantly higher (all-E)-lycopene (14.0 vs. 71.1 μg/g), (13Z)-lycopene (1.1 vs. 5.0 μg/g) and (all-E)-γ-carotene (1.6 vs. 3.8 μg/g) levels. Significant increases in most of the carotenoids occurred from the partially ripe to the ripe fruits, with (all-E)-lycopene doubling its concentration in fruits from both states. Pitanga was found to be one of the richest fruit sources of carotenoids, particularly lycopene, but the processed products had much lower lycopene content. The mean (all-E)-lycopene concentration was 16.6 μg/g for frozen pulp brand A, 23.0 μg/g for bottled juice brand B and 25.6 μg/g for bottled juice brand C. Optimization of processing is therefore needed to guarantee better retention of this important carotenoid.  相似文献

Starch has been the most popular and economic size material. Synthetic binders have also been developed to be used as size material to improve weaving loom efficiency. Some synthetic size materials have got restrictions in use mainly because of ecological reasons. In the recent years, many modifications have come up in the starch as a sizing agent. Different modification can give different properties, which can be suited for a particular application. In the present study, for comparative analysis of different varieties of natural starch, modified starch and synthetic size material have been evaluated and mechanical properties like cohesion power, adhesion power, abrasion resistance, bending rigidity etc. were studied. Total amino acid contents of several important U.S. potato cultivars grown in Maine, Idaho and The Red River Valley are listed along with nitrogen content. Differences in amino acid content among cultivars were roughly proportional to differences in total nitrogen. The effect of storage on amino acids in two cultivars was minor. The limiting essential amino acids compared to egg were MET and the sulphur amino acids.  相似文献

The effect of hydroxypropyl-β-cyclodextrin (HPβCD) in the dyeing of polyamide fabric was investigated under conditions that mimic industrial dyebaths where dye mixtures are used. The dyes used were Telon Red BN, Telon Yellow A-2R, and Telon Blue RR. The blue dye complexed with HPβCD whereas the other dyes did not do so under the conditions used. The dyeing results were compared with those obtained using a traditional retarding agent, Albegal B. The exhaustion dyeing data showed that HPβCD mainly affected the dyeing behavior of the blue dye that became encapsulated. In the case of the yellow and red dyes, changes in the overall kinetics of dyeing were observed, resulting in modified exhaustion profiles. In comparison, Albegal B retarded the blue dyeing process and increased the rate of exhaustion of the yellow dye while the exhaustion curve of red dye remained unchanged. The color uniformity of the dyed polyamide was improved when HPβCD was used. This resulted from the more controlled exhaustion rate during the critical phase of the dyeing process.  相似文献

Teton Russet is an early-maturing, medium-russeted, potato cultivar with high merit for both fresh-pack and processing. In early harvest trials in the Pacific Northwest, Teton Russet had total yields similar to Russet Norkotah, and higher than Ranger Russet and Russet Burbank. Marketable yield of Teton Russet in the early harvest trials was also comparable to or higher than Russet Norkotah in Washington and Oregon, and higher than Ranger Russet and Russet Burbank at these sites, as well as in Idaho. In full-season trials, while total yield of the earlier-maturing Teton Russet tended to be lower than Ranger Russet and Russet Burbank, marketable yield was generally higher than Russet Burbank across the majority of sites due to its higher percentage of U.S. No. 1 tubers. Teton Russet is suitable for processing, with acceptable fry color following up to 8 months of storage at 8.9 °C. Uniformity of fry color was also very consistent. Teton Russet has shown lower levels of the amino acid asparagine relative to Ranger Russet and Russet Burbank which may contribute to lower acrylamide levels in French fries and other processed potato products. Teton Russet is notable for having resistance to common scab (Streptomyces spp.) and Fusarium dry rot, and is moderately resistant to tuber net necrosis. Analyses have also shown Teton Russet to have significantly higher protein levels than Russet Norkotah, Ranger Russet, and Russet Burbank, as well as higher vitamin C content than Russet Norkotah and Russet Burbank. Teton Russet was released in 2011 by the USDA-ARS and the Agricultural Experiment Stations of Idaho, Oregon, and Washington, and is a product of the Pacific Northwest Potato Variety (Tri-State) Development Program.  相似文献

Eight grass silages were made from two contrasting varieties of perennial ryegrass, four silages each from a late-cut early-maturing (high dry matter, HDM) variety and from an early-cut, late-maturing (low dry matter, LDM) variety. The grass was ensiled without additive (untreated), or with formic acid, or with one of two enzyme mixtures of hemicellulases and cellulases (enzyme 1 and enzyme 2) for a period of 130 d.
Formic acid-treated silage had lower levels of lactic acid at both levels of dry matter than the other silages. Enzyme treatment of grass prior to ensilage resulted in reduced levels of cellulose, acid-detergent fibre and neutral-detergent fibre in LDM silages and lower acid-detergent fibre and neutral-detergent fibre in the HDM silages compared with the corresponding untreated and formic acid-treated silages. Voluntary intakes (g DM d−1) of untreated and enzyme-treated silages were significantly ( P <0·01) lower at both digestibilities compared with formic acid-treated silages (LDM: untreated, 982; formic, 1069; enzyme 1, 868; enzyme 2, 937; HDM: untreated, 931; formic, 1027; enzyme 1, 943; enzyme 2, 914). The organic matter, carbohydrate and nitrogen digestibility coefficients of LDM silages were significantly ( P <0·001) higher than those of HDM silages. There were no significant differences in any component digestibility related to silage additive.
Comparison of digestibility coefficients for constituents of the LDM silages fed to sheep or steers showed no differences between species.  相似文献

Giant piezoelectricity on Si for hyperactive MEMS   总被引:1,自引:0,他引:1  
Microelectromechanical systems (MEMS) incorporating active piezoelectric layers offer integrated actuation, sensing, and transduction. The broad implementation of such active MEMS has long been constrained by the inability to integrate materials with giant piezoelectric response, such as Pb(Mg(1/3)Nb(2/3))O(3)-PbTiO(3) (PMN-PT). We synthesized high-quality PMN-PT epitaxial thin films on vicinal (001) Si wafers with the use of an epitaxial (001) SrTiO(3) template layer with superior piezoelectric coefficients (e(31,f) = -27 ± 3 coulombs per square meter) and figures of merit for piezoelectric energy-harvesting systems. We have incorporated these heterostructures into microcantilevers that are actuated with extremely low drive voltage due to thin-film piezoelectric properties that rival bulk PMN-PT single crystals. These epitaxial heterostructures exhibit very large electromechanical coupling for ultrasound medical imaging, microfluidic control, mechanical sensing, and energy harvesting.  相似文献
