Since 2002, papaya black rot has been spreading over several islands of Okinawa Prefecture, Japan. To devise a prevention strategy for the disease, microbiological research on the pathogen was conducted. Twelve strains were isolated from papaya infected with black rot showing symptoms such as water-soaked lesions on stems and petioles, black spots on fruits, and rotted leaves turning yellow with necrotic spots. Through Koch's postulates, we confirmed that the isolated strains caused papaya black rot. Bacteriological assays showed that the strains have characteristics different from the type strains of Erwinia mallotivora, E. papayae, and E. psidii. Moreover, 16S rDNA sequence similarity searches showed that the isolated strains had less than 98.6% similarity with type strains. Additionally, phylogenetic analysis of 16S rDNA sequences suggested that the isolated strains were possibly a novel species belonging to the genus Erwinia, as the strains formed an independent cluster and had low sequence similarity with the type strains. Earlier studies indicated that papaya black rot is caused by E. cypripedii. Therefore, we propose to add the Erwinia sp. isolated in this study to the list of papaya black rot pathogens.  相似文献

We investigated for dynamics of Campylobacter clones on 2 different managerial broiler farms. Campylobacter isolates were differentiated by resistance typing and molecular typing methods. On farm I, the same C. jejuni clones resistant to fluoroquinolone and oxytetracycline were isolated after one and half years again and another susceptible clone was invaded. The susceptible clone was isolated again after half year. Broiler flocks on the farm may be repeatedly infected with a few C. jejuni clones. On farm II, new clones including antimicrobial resistant one, were often invaded. The change of predominant C. jejuni clone in each flock on both the farms was observed, in the absence of antimicrobial selective pressure.  相似文献

We performed a field experiment in thebiodegradation of heavy oil spilled from the Russian tankerNakhodka on a beach in the Sea of Japan. We collectedoil-contaminated cobbles and treated half with nitrogen andphosphorus slow-release fertilizers to stimulate microbialdegradation of the oil; the other half acted as unfertilizedcontrols. The cobbles were placed in porous acrylic vessels andsubmerged. We monitored changes in the oils, macronutrients,microbial community structure and amount of chlorophyll a. There were no significant differences in these criteriabetween the fertilized and unfertilized vessels, apart from anincrease in chlorophyll a in the fertilized vessels.However, there was a major intrinsic degradation of semi-volatile oil compounds in the unfertilized vessels; this occurred at a rate similar to that in the fertilized vessels, despite the low concentration of macronutrients in the seawater at the site.  相似文献

The Concentration of nitric oxide can be monitored by a new device in which the Zeeman effect is used to shift an absorption line of nitric oxide into coincidence with a laser line of carbon monoxide. The absorption is modulated by a small, oscillating magnetic field. This device is specific for nitric oxide and is not subject to interference from other gases.  相似文献

Tyrosinase, trypsin, and tryptase are known to play important roles in melanin production of human skin. This paper describes the study of the inhibitory effect of Balanophora fungosa on melanin. The 50% EtOH extract obtained from B. fungosa indicated an inhibitory effect on mushroom tyrosinase activity with an IC(50) value of 15 μg/mL. Bioassay-guided fractionation of the active extract resulted in the isolation of four known compounds. Their structures were identified as 1-O-(E)-caffeoyl-3-O-galloyl-4,6-(S)-HHDP-β-d-glucopyranose (1), 1-O-(E)-caffeoyl-3,4,6-tri-O-galloyl-β-d-glucopyranose (2), caffeoyl-β-d-glucopyranose (3), and abietin (4) on the basis of spectroscopic analyses and comparison of their spectral data with those in the literature. Compounds 1 and 2 prevented pigmentation of melanin in a three-dimensional cultured human skin model. Furthermore, compounds 1 and 2 indicated inhibitory activities against trypsin and tryptase.  相似文献

A guinea pig (9-week-old) that had been placed in a control group for a pharmacological test was found to have a single nodule on the surface of the right ventricular wall. In a transverse section of the heart after fixation, a whitish mass was found that extended from the subendocardium to the subepicardium of the right ventricular wall. Histopathological examination revealed a spongy network consisting of vacuolated spaces in the myocardium of the right ventricle extending to the myocardium and subepicardium of the right atrium. The vacuolated space was PAS-positive. Immunohistochemical examinations revealed that the lesions contained striated fibers that were positive for anti-desmin and anti-myoglobin. Electron micrographs revealed the lesions resulting in affected striated muscle fibers and accumulations of many glycogen granules. Based on the findings, the lesions were diagnosed as a cardiac rhabdomyoma. This is the first report of application of immunohistochemical examinations to diagnosis of cardiac rhabdomyoma in the guinea pig.  相似文献

The chemical characteristics of the Urauchi River water in Iriomote Island National park, Okinawa, Japan have been studied. Concentrations of PO4 3?, NO2 ?, and NH4 + were barely detectable. We compared the concentration ratios of Mg2+/Na+, HCO3 ?/Na+, and Ca2+/Na+ in the Urauchi River to those of 60 large rivers in the world and indicated that the chemical composition of the river is most likely formed by the binary mixing of sea salt components and silicate rock weathering components. Although rock weathering in the catchments area is driven by both H2CO3 and H2SO4, the role of H2CO3 is dominant. The percentages of the concentration of each cation in the river water are almost the same as those of other rivers with drainage areas consisting of silicate rock and sandstone. Thus, the Urauchi River shows the typical chemical characteristics of a river in a silicate rock area that includes sandstone.  相似文献
