It has been postulated that osmotic effects on plant tissue are mediated by abscisic acid (ABA). Hybrid larch (Larix kaempferi (Lambert) Carr. x L. decidua Mill.) plantlets, transformed with the ABA-inducible wheat Em promoter associated with the Gus reporter gene, were axenically inoculated with two ectomycorrhizal fungi: Cenococcum geophilum Fr., considered tolerant to water stress, and Laccaria bicolor (Marie) Orton, considered less tolerant to drought. The mycorrhizal and non-mycorrhizal transgenic plantlets were subjected to osmotic stress by adding polyethylene glycol (PEG) to the culture medium. In the presence of PEG, L. bicolor and C. geophilum reduced shoot water potential and turgor potential, but increased host osmotic potential. Treatment of plantlets with PEG induced a significant increase in endogenous ABA concentrations. Laccaria bicolor and C. geophilum behaved similarly and significantly decreased the ABA response of plantlets to PEG treatment. Moreover, inoculation with either fungus regulated the ABA response of the plantlets even when the fungus was separated from the host by a cellophane sheet that prevented mycorrhiza formation. Although the wheat Em promoter was inducible in larch plantlets, it was not regulated by endogenous ABA. Induction of the wheat Em promoter in larch plantlets depended on organ type, with maximum induction in the root apex. Induction of the Em promoter was significantly decreased by mycorrhizal inoculation.  相似文献

Summary Lepyrus palustris Scop., a pest of the basket-willow has in Hungary one generation in a year. The overwintered adults appear in the early spring and cause occasionally severe damages on the young shoots, especially in the southern parts of Hungary. The females lay 100–140 eggs during the months May–June on the earth surface near to the willow-stocks. The larvae enter the soil and damage outside on the main roots, which can cause severe losses mainly in the young plantations. The 4th instar larva pupate in the soil. The young adults appear in July, the damage caused by them in the second part of the summer is not more important. The best control has been achieved applying Wofatox dust (3 per cent) against the overwintered adults at the beginning of the vegetation period.  相似文献

In Spain, many Pinus halepensis Mill. forests have been seriously affected by significant forest fires in the past decade, in 1994 alone, more than 100 000 ha were burned in Eastern Spain. In order to study the reproductive characteristics of P. halepensis after forest fires, six locations were selected in four areas affected by serious fires in the summer of 1994, and the following different precipitation zones were studied: dry-subhumid, dry and semi-arid. Ten years after the fires, data relevant to the production of pine cones: serotinous (grey), mature (brown), immature (green) and opened cones, was collected from areas with natural post-fire regeneration. Various cone and seed characteristics, such as pine cone seed number and weight, and germination percentage, were measured. The results showed greater production of cones and strobili in high-density sites. The biggest cone sizes (volume) and seed number per cone were related to site quality (dry-subhumid precipitation zone). Also, viability and germination percentages were higher with better site quality, with significant differences in values for serotinous and non-serotinous cones. Despite these differences, the canopy seed bank was large enough to ensure regeneration in this area for this age stand.  相似文献

This study describes the distribution of galanin (Gal) and galanin receptor 2 (GalR2) in the pre‐optic area (POA) of the female guinea pig. Frozen sections were undergone for a routine immunofluorescence labelling. Gal and GalR2 display immunoreactivity in all parts of the pre‐optic area. Gal shows reactivity both in perikarya and fibres, whereas GalR2 was observed only in perikarya. Gal‐ and GalR2‐immunoreactive (‐ir) perikarya were the most numerous in the medial pre‐optic area (MPA) with the highest reactivity in its dorsal part. In the median pre‐optic nucleus (MPN) and periventricular pre‐optic nucleus (PPN), only single Gal‐ and GalR2‐ir neurons were observed. The highest density of Gal‐ir fibres was revealed in the PPN and the lowest in the lateral pre‐optic area (LPA). The results of this study indicate that the distribution pattern of Gal containing neurons overlaps well with the distribution pattern of GalR2‐positive neurons, especially in the MPA. This may suggest GalR2‐dependent activity in this brain region.  相似文献

Our previous study revealed the expression of substance P (SP) and calcitonin gene‐related peptide (CGRP) in sensory distal ganglion of the vagus (nodose ganglion) neurons in the pig. As these neuropeptides may be involved in nociception, the goal of these investigations was to determine possible expression of vasoactive intestinal polypeptide (VIP), SP and CGRP in the pituitary adenylate cyclase‐activating polypeptide‐immunoreactive (PACAP‐IR) porcine nodose perikarya. Co‐expression of these substances was examined using a double‐labelling immunofluorescence technique. To reveal the ganglionic cell bodies, the pan‐neuronal marker protein gene product 9.5 (PGP 9.5) was used. Quantitative analysis of the neurons revealed that 67.25% of the PGP 9.5+ somata in the right‐side ganglion and 66.5% in the left side, respectively, co‐expressed PACAP‐IR. Moreover, 60.6% of the PACAP‐IR cells in the right‐side ganglion and 62.1% in the left, respectively, co‐expressed VIP. SP‐IR was observed in 52.2 and 39.9% of the right and left ganglia, respectively. CGRP was found in 27.7 and 34.1% of the right and left distal ganglion of the vagus, respectively. High level of co‐expression of PACAP with VIP, SP and CGRP in the distal ganglia of the vagus sensory perikarya directly implicates studied peptides in their functional interaction during nociceptive vagal transduction.  相似文献

The present assay attempts to evaluate the feasibility of using embryo transfer in small community farmers by in vivo study and by modelling the results obtained. From the total of 59 donor cows, 62.7% responded to treatment, with a significant difference (p = 0.002) in the percentage of the response between breeds, being 90.5% (19/21) in Holstein and 47.4% (18/38) in Brahman. A total of 283 embryos were graded as transferable, while 141 as non‐transferable, without difference in the percentage of transferable embryo by breed (p = 0.18). The mean of transferable embryos graded as class I and II was not different between Holstein and Brahman (p = 0.96 and p = 0.92, respectively); besides, no differences were observed in the other grades (non‐transferable). The highest difference in costs, regardless of its quality by breed, was seen in the lower levels of probable fertility of the embryo transferred, even reaching several hundred dollars. When modelling the expected costs for embryo produced and transferred, values can reach nearly $2000.00 when the probable fertility is only 10%. However, when the probable fertility was 60%, embryo cost was close to $300.00. This technology seems to be viable on average or high‐scale systems, having a superovulatory response between 60 and 80% with 4–6 transferrable embryos. Yet, in small‐scale farming, due to the reduced number of donors and/or recipients, the costs surpass the economical feasibility of the technique.  相似文献
