A quantitative polymerase chain reaction (qPCR) assay was developed for the detection and quantification of Edwardsiella ictaluri in channel catfish Ictalurus punctatus pond water using modifications to a published E. ictaluri-specific qPCR assay and previously established protocols for the molecular detection of myxozoan parasites in catfish ponds. Genomic DNA equivalents indicative of the number of bacteria in a sample were determined and standard curves correlating to bacterial numbers were established. The assay was found to be highly repeatable and reproducible, with a linear dynamic range of five orders of magnitude. There was no interference of the assay from the presence of large quantities of nontarget DNA. Known quantities of bacteria were added to sample volumes of 40 or 500 mL of pond water collected from several different ponds. The minimum level of detection was approximately 100 cell equivalents (CE) in 40 (2.5 CE/mL) or 500 mL of pond water (0.2 CE/mL). Sample volumes of 40 mL yielded the most consistent results, which were not significantly different from those obtained from broth culture alone. Cell equivalents determined by qPCR in 40-mL pond water samples spiked with known quantities of bacteria were within one order of magnitude of the actual number of cells added. Repetitive element-based polymerase chain reaction analysis of archived isolates demonstrated the genetic homogeneity of E. ictaluri, and consistent amplification of these isolates by qPCR analysis demonstrated the stability of the PCR target. The assay described here provides a reliable method for the detection and quantification of E. ictaluri in pond water and will be an invaluable tool in epidemiological studies. Additionally, the assay provides a way to evaluate the effects that vaccination, antibiotic treatments, and restricted feeding practices have on E. ictaluri populations during an outbreak. Information obtained with these tools will aid in optimizing disease management practices designed to maximize productivity while minimizing losses.  相似文献

Enteric septicemia of catfish (ESC), caused by Edwardsiella ictaluri, is the most problematic bacterial disease affecting catfish aquaculture in the southeastern United States. Efforts to develop an effective ESC vaccine have had limited industrial success. In commercial settings, ESC vaccines are typically administered by immersion when fry are transferred from the hatchery to rearing ponds. While this approach is a practical method of mass delivery, this strategy administers vaccines to very young fish, which lack a fully developed immune system. To circumvent this limitation, an oral vaccination strategy was evaluated as a means of immunizing catfish at the fingerling stage of production, when fish possess a more complete immune arsenal. A virulent E. ictaluri isolate (S97-773) was attenuated by successive passage on media containing increasing concentrations of rifamycin. In laboratory trials, cultured vaccine was diluted and mixed with feed (100 mL diluted vaccine/454 g feed). This mixture was then fed to Channel Catfish Ictalurus punctatus fingerlings. Two separate dilutions of cultured vaccine (1:10 and 1:100) were used to create the vaccine–feed mixture, equating to estimated doses of 5 × 107 and 5 × 106 CFU/g of feed, respectively. After 30 d, catfish were exposed by immersion (1 × 106 CFU/mL) to the virulent parental strain of E. ictaluri. The target dose (1:100 dilution, ~5 × 106 CFU/g of feed) offered exceptional protection (relative percent survival = 82.6–100%). In addition, negligible deaths occurred in fish vaccinated at 10 times the target dose (1:10 dilution, ~5 × 107 CFU/g of feed). In pond trials, antibody production increased 18-fold in orally vaccinated fish. When compared with nonvaccinated controls, vaccination significantly improved survival, feed fed, feed conversion, biomass produced, and total harvest. This research demonstrates Channel Catfish can be successfully immunized in a commercial setting against E. ictaluri with a single dose of an orally delivered, live attenuated, E. ictaluri vaccine.

Anticoccidials are compounds that are widely used as feed additives to prevent and treat coccidiosis. They are licensed for use in a prescribed concentration and during a certain time interval for broilers and pullets but not for laying hens. It was shown in the past that carry-over at the feeding mill is found to be the main reason for the presence of residues in eggs. An animal experiment was set up to investigate the effect of carry-over at the feeding mill on the presence of residues of anticoccidials in eggs. For the compounds diclazuril, robenidine, halofuginone and nicarbazin in combination with narasin, two concentration levels were tested: the maximum allowed concentration for broilers (100%) and a concentration corresponding to 5% carry-over during feed preparation. Also dimetridazole was included in the experiment but only at one concentration level. Eggs were sampled during treatment (14 days) and for a period of 30 days after withdrawal of the anticoccidial-containing feed. Residues were determined, and deposition and depletion curves were generated. Analyses were performed by ELISA and LC-MS/MS. For all compounds, substantial residues could be found in the 5% groups, which points out the risk of carry-over at the feeding mill. The distribution of the residues between egg yolk and white was determined by analyzing both fractions.  相似文献

The effects of long-term feeding of cottonseed meal on growth, testis development, and sperm motility of male channel catfish Ictulurus punctatus were studied. Brood-sized male channel catfish were stocked into 0.04-ha earthen ponds in April 1992 at the rate of 120 fish per pond. The fish were fed a diet (32% protein floating catfish feed) containing either 0, 25, 375, or 52% cottonseed meal to satiation daily, except for winter months when fish were fed on days when the water temperature was 15 C or above. Fish were harvested in July 1994. Feed consumption was similar for fish regardless of dietary treatment. Feed conversion was higher for fish fed the two highest levels of cottonseed meal and weight gain was depressed in these groups. Testis weight, gonosomatic index, and sperm motility were not negatively affected by high levels of dietary cottonseed meal indicating that reproductive capacity was not diminished. Thus it wonld appepr that up to 52% cottonseed meal could be used in the diets of brood-sized channel catfish unless maximum gain is important. In a practical situation, growth rates of brood-sized channel catfish are not as important as reproductive performance.  相似文献

A major development concern of the 1970s and 1980s was ‘the other energy’ or ‘woodfuel crisis’. Woodfuel use in developing regions of Africa, Asia, and Latin America was believed to be a key factor in tropical deforestation, and the loss of forests was projected to result in widespread woodfuel shortages. In recent years, however, it has become apparent that woodfuel production is seldom a direct cause of deforestation (although it may be a by‐product), and that most woodfuel demand is met by trees and shrubs growing outside of forest areas. In addition, earlier predictions of serious woodfuel shortages have not materialized. But in some regions, including the Philippines, woodfuel use is still regularly pointed to as a primary cause of deforestation, and woodfuel shortages are still predicted for the immediate future. In particular, commercial woodfuel demand for household and commercial use is said to be contributing to over‐exploitation of forest resources. This paper examines the connections between commercial woodfuel demand and deforestation on the Island of Cebu in the central Philippines. Cebu is widely perceived as an environmental disaster area on the brink of ecological collapse, and tree cutting for woodfuel production is often blamed for contributing to this condition. In contrast, this paper will argue that environmental conditions on Cebu are improving, that tree planting and management is widespread among smallholder farmers and other landowners, and that commercial demand for woodfuel and other wood products is one of the main factors driving increased reforestation on the island. A commercially available heterogeneous, solid-phase tube enzyme-linked immunoassay (ELISA) was modified and validated for the measurement of serum cortisol in rainbow trout Oncorhynchus mykiss . The assay is accurate and precise. Resting and stress-elevated serum cortisol concentrations were measured in rainbow trout with a sensitivity of 1.5 ng/ml. Fish held in net-pens at a density of 0.4 kg/m3/cm had a resting cortisol level of 16.5 ± 3.8 ng/ml (mean ± SE). At 3 h postdisturbance, serum cortisol levels were not affected by the removal of fish from adjacent net-pens with dip nets or by the use of 200 mg/L tricaine methanesulfonate (MS-222) as an anesthetic for obtaining samples. However, an acute stress (60 s removal from water) elevated serum cortisol levels to 73.7 ± 9.4 ng/ml.  相似文献

During the spring of 2002, a low pathogenic avian influenza (LPAI) A (H7N2) virus caused a major outbreak among commercial poultry in Virginia and adjacent states. The virus primarily affected turkey flocks, causing respiratory distress and decreased egg production. Experimentally, turkeys were more susceptible than chickens to H7N2 virus infection, with 50% bird infectious dose titers equal to 10(0.8) and 10(2.8-3.2), respectively. Comparison of virus shedding from the cloaca and oropharynx demonstrated that recent H7N2 virus isolates were readily isolated from the upper respiratory tract but rarely from the gastrointestinal tract. The outbreak of H7N2 virus raised concerns regarding the availability of vaccines that could be used for the prevention and control of this virus in poultry. We sought to determine if an existing commercial avian influenza (AI) vaccine prepared from a 1997 seed stock virus could provide protection against a 2002 LPAI H7N2 virus isolated from a turkey (A/turkey/Virginia/158512/02 [TV/02]) in Virginia that was from the same lineage as the vaccine virus. The inactivated AI vaccine, prepared from A/chicken/ Pennsylvania/21342/97 (CP/97) virus, significantly reduced viral shedding from vaccinated turkeys in comparison with sham controls but did not prevent infection. The protective effect of vaccination correlated with the level of virus-specific antibody because a second dose of vaccine increased antiviral serum immunoglobulin G and hemagglutination inhibition (HI) reactivity titers in two different turkey age groups. Serum from CP/97-vaccinated turkeys reacted equally well to CP/97 and TV/02 antigens by HI and enzyme-linked immunosorbent assay. These results demonstrate the potential benefit of using an antigenically related 1997 H7N2 virus as a vaccine candidate for protection in poultry against a H7N2 virus isolate from 2002.  相似文献
