The present study evaluated the effect of supplementing the medium used to mature equine oocytes in vitro with oestrous mare serum (EMS) or horse follicular fluid (HFF). To this end, 144 ovaries were obtained from mares aged 16-21 months and transported to the laboratory in Dulbecco's phosphate buffered saline (D-PBS) at 30 degrees C. Oocytes were harvested from the ovaries by slicing, and then selected for in vitro maturation (IVM) according to the number of cumulus cell layers and the characteristics of the cytoplasm. The selected oocytes were washed three times in TCM199 medium plus HEPES (TCM-199H) or in the same medium plus glutamine (TCM-199G), then matured in vitro in six study groups established according to the in vitro maturation (IVM) treatment to see possible interactions between HEPES and glutamine on other supplements: Ten percent EMS was added to two of these media (TCM-199H+EMS and TCM-199G+EMS) and 10% HFF was added to the media in two other groups (TCM-199H+HFF and TCM-199G+HFF). IVM was performed at 38.5 degrees C for 40 h in a controlled atmosphere (5% CO2, 95% relative humidity). The findings indicate that the presence of EMS or HFF in the TCM-199H medium gives rise to the best results in terms of the proportions of oocytes reaching maturity (37.7% and 36.8%, respectively). The values obtained with EMS and HFF were statistically similar to each other but differed from the other treatments. The media containing glutamine led to the highest proportions of degenerated oocytes.  相似文献

Critical illness, anesthesia, primary cardiovascular disease, and exercise may result in marked hemodynamic alterations. Measuring cardiac output (CO) is central to defining these alterations for both clinician and researcher. In the past 10 years, several new methods of measuring CO have been developed for the human medical market. Some of these methods are now validated in the horse and are in clinical use. The Fick method has been used in equine research for more than a century. It depends on simultaneous measurement of mixed venous (pulmonary arterial) and peripheral arterial oxygen content and oxygen uptake by the lungs. The technique is technically demanding, which restricts its clinical use. Indicator dilution techniques, with indocyanine green, cold (thermodilution), or lithium as the marker, have also been widely used in the horse. The indocyanine technique is cumbersome, and thermodilution requires right heart catheterization, which is not a benign procedure, making both of these methods less than ideal for clinical use. Lithium dilution requires catheterization of a peripheral artery and a jugular vein. It has recently been validated in anesthetized adult horses and neonatal foals. Doppler echocardiography is a noninvasive ultrasound-based technique. More accurate measurements are obtained with transesophageal than with transthoracic measurements; however, both methods require considerable technical expertise. Bioimpedance and pulse contour analysis are 2 new methods that have yet to be validated in the horse. With the currently available technology, lithium dilution appears to be the method of measuring CO best suited to the equine clinic.  相似文献

Sixteen Chilean flamingos, Phoenicopterus chiles, and 10 red-tailed hawks, Buteo jamacensis, were vaccinated in the pectoral muscle with 0.2 ml of a commercially produced killed West Nile virus vaccine intended for use in horses. Half the birds of each species received a booster vaccination 3 weeks after the first injection. Three weeks after the booster vaccination, none of 13 birds surveyed had detectable antibody to West Nile virus.  相似文献

  • 1. River restoration is now widely undertaken and may be considered an increasingly important aspect of river management. Recent developments in European legislation (Habitats Directive and the Water Framework Directive) should give further impetus to river restoration across EU member states, as this legislation places greater emphasis on the processes that support river ecology.
  • 2. Traditional river restoration approaches have been reach‐focused and opportunistic, capitalizing on flood defence works and the cooperation of sympathetic landowners.
  • 3. Furthermore, many schemes have been species‐ or habitat‐driven and, thus, have sought to recreate channel forms believed to be favoured by particular species or associated with particular habitats. Such approaches have not always given due attention to the underlying geomorphological processes that create channel form and, consequently, the schemes have not been self‐sustaining, requiring continued management input.
  • 4. Consistent with the ethos of the Water Framework Directive, an eco‐hydromorphic approach to river restoration is proposed here. This approach views spatial and temporal heterogeneity as fundamental characteristics of fluvial systems and advocates recreating a framework within which natural processes, such as sediment transport and nutrient dynamics, can occur.
  • 5. Mesoscale habitat approaches are considered as one possible way to plan and achieve this framework, providing the potential to link the wide range of spatial and temporal scales that characterize river systems.
  • 6. It is argued that river restoration will only be sustainable if it is undertaken within a process‐driven and strategic framework with inputs from a wide range of specialists. Such an approach needs to be reviewed constantly in light of appraisal and monitoring of previous river restoration schemes.

This study examines parents’ perspectives on their children working on their family dairy farms in Wisconsin. The objective of this focus group study is (1) to gain insights on why children work on their family farms, (2) to identify those benefits that parents perceive that they and their children gain from their children working on-farm, (3) to determine the concerns that parents have about their children working, (4) to identify ways to improve the safety of children on family farms, and (5) to understand how US agricultural policy impacts family decisions to use their children’s labor on their farms. The two focus groups reveal that fathers and mothers have different concerns and different perceptions regarding the benefits gained from having their children work on farms. The findings suggest that in response to US agricultural policy, parents are increasingly reliant upon their children’s labor. Children work the longest hours on economically stressed farms. Lydia Zepeda is a fellow of the Center for World Affairs and the Global Economy and professor of Consumer Science at the University of Wisconsin-Madison. She earned her MS and PhD in Agricultural Economics at the University of California at Davis. Her research interests include agricultural technology adoption, risk perceptions, household decision-making, and consumer food demand. Jongsoog Kim is a research fellow at Korea Women’s Development Institute in Seoul, Korea. She earned her PhD in Human Ecology at the University of Wisconsin-Madison.
