Much remains to be discovered about the population genetic structure of parasites, despite the importance of such knowledge to understanding the processes involved in the spread of drug resistance through populations. Here we present a study of population genetic diversity in Trichostrongylus tenuis, an avian parasitic nematode infecting both poultry and game birds, where anthelmintic use is common. We examined diversity of nicotinamide dehydrogenase subunit 4 (nad4) mtDNA sequences within and between seven locations: five in the UK (red grouse hosts), one in Iceland (domestic goose) and one in Norway (willow grouse). Within-UK comparisons showed high nucleotide diversity (pi=0.015, n=23) but no structure between locations (Phi(ST)=0.022, P=0.27), with over 97% of variation distributed within-hosts. The highest diversity was found in Iceland (pi=0.043, n=4), and the lowest in Norway (pi=0.003, n=4). Differentiation between countries was considerable (Phi(CT)=0.44, P<0.05), in spite of the potential mixing effects of gene flow via migrating wild hosts and the poultry trade. However, significant pairwise F(ST) values were found only between Norway and the other locations. Phylogenetic analysis provided statistical support for a separate clade for Norwegian samples only, with unresolved diversity leading to a star-shaped relationship between Icelandic and UK haplotypes. These results suggest that Norwegian T. tenuis are isolated, but that there is some connectivity between UK and Icelandic populations. Although anthelmintic resistance has not yet been reported for T. tenuis, the population structure is such that emerging resistance has the potential to spread by gene flow over a large geographic scale.  相似文献

The aim of the study was to investigate the ability of vitamin C to increase the antioxidative and immunomodulating potential in healthy dogs. Fifteen dogs were tested for the effects of orally administered vitamin E (60 mg dl‐α tocopheryl acetate) in combination with vitamin C (0, 30 or 60 mg ascorbic acid crystalline). Three treatments (0, 30, 60 mg vitamin C) were tested in a 3 × 3 cross‐over study in three periods of 36 days. Pre‐prandial blood samples were taken for analysis of vitamins C, E, A, retinyl palmitate and stearate, antioxidant status [Thiobarbituric acid reactive substances (TBARS) and uric acid], biochemical and haematological analysis. Subpopulations of lymphocytes, mitogen‐induced peripheral blood mononuclear cell proliferation (PBMC) and serum IgA and IgG concentrations were determined. There was a trend (p = 0.056) for an increased plasma vitamin C concentration by vitamin C supplementation. There was no evidence that dietary treatment altered neither the other plasma vitamin concentrations nor TBARS and uric acid concentrations nor the subpopulations of the lymphocytes except for the number of CD4+ lymphocytes that increased with vitamin C supplementation. There was no effect of vitamin C on serum IgA and IgG concentration. A significant time × treatment interaction was demonstrated on PBMC’s to pokeweed, with an increase observed by 30 mg vitamin C supplementation but a decrease by 60 mg vitamin C supplementation. There was no clear evidence for an effect of dietary vitamin C on antioxidative capacity in healthy dogs fed a diet with vitamin E concentrations well above the recommendations. Yet, a limited number of immunological parameters were slightly affected.  相似文献

The purpose of this study was to investigate the impact of eggshell calcium (Biomin H® dietary supplement) and its combinations with alfacalcidol (1α-hydroxyvitamin D3) and menaquinone-7 (vitamin K2) on ovariectomy-induced bone loss in rats. Adult female rats (n = 48) were divided into 6 groups of 8 individuals each: sham-operated rats (SHAM); ovariectomized (OVX) rats untreated; OVX rats treated with Biomin H® (BIO); OVX rats simultaneously receiving Biomin H®, vitamin D3 (BIO + D3); OVX rats simultaneously treated with Biomin H®, vitamin K2 (BIO + K2) and OVX rats treated with Biomin H®, vitamin D3, vitamin K2 (BIO + D3 + K2) during 8 weeks. Biochemical parameters, bone mineral density (BMD), bone mineral content (BMC) and femoral bone microstructure were determined. Plasma calcium and phosphate were increased in BIO + D3 and BIO + D3 + K2 groups as compared to OVX. Alkaline phosphatase was elevated in OVX, BIO versus SHAM, BIO + D3 + K2 groups. When compared to OVX group, decreased urine deoxypyridinoline was observed in all treated groups and femoral BMD, BMC were higher in BIO, BIO + D3, BIO + D3 + K2 groups. The BIO + K2 rats had similar densitometrical values than OVX individuals. Microcomputed tomography revealed increased trabecular relative bone volume (due to an increase in trabecular number) in BIO + D3, BIO + D+ K2 as compared to OVX. The higher relative bone volume in BIO + D3, BIO + D+ K2 groups was also accompanied by an increase in bone surface. In the cortical bone, an enhanced periosteal bone apposition was identified in BIO, BIO + D3, BIO + K2, BIO + D+ K2 groups. The rats from BIO + D+ K2 group had a higher area of primary osteon's vascular canals. In BIO + D3, BIO + K2, BIO + D+ K2 groups, an increased area of secondary osteons was determined in comparison with OVX. Our results indicate the beneficial effect of triple application of Biomin H®, vitamin D3, vitamin K2, as well as simultaneous administration of Biomin H®, vitamin D3 on the inhibition of ovariectomy-induced bone loss in a rat model of osteoporosis.  相似文献

The key process for immune response development is the recognition of bacteria by the immune system of the host based on the sensing of pathogen-associated molecular patterns (PAMP). One of the most important PAMP is the lipopolysaccharide (LPS) molecule, a complex molecule present in the outer membrane of Gram negative bacteria. In this study we were interested in how different parts of the LPS of Salmonella enterica serovar Enteritidis are recognized by porcine neutrophils. To this aim, we constructed S. Enteritidis mutants with rfaL and rfaC genes disabled in the attachment of the O-antigen and in the synthesis of the inner oligosaccharide core of LPS, respectively. We found that in the absence of serum, both the rfa mutants associated with neutrophils and stimulated them for reactive oxygen species (ROS) production significantly more than the wild-type strain. Addition of polymyxin B, which neutralized lipid A, the endotoxic moiety of LPS, effectively decreased the association of the wild-type strain and the rfaC mutant with neutrophils, but not the rfaL mutant. This indicates that the oligosaccharide core newly exposed on the surface in the rfaL mutant, protected from interaction in the wild-type strain by the O-antigen but completely absent in the rfaC mutant, may represent a new ligand for porcine neutrophils that cannot be neutralized by polymyxin B.  相似文献

Confocal laser scanning microscopy (CLSM) is the method of choice to study interfacial biofilms and acquires time-resolved three-dimensional data of the biofilm structure. CLSM can be used in a multi-channel modus where the different channels map individual biofilm components. This communication presents a novel image quantification tool, PHLIP, for the quantitative analysis of large amounts of multichannel CLSM data in an automated way. Ninety potted red spruce saplings enclosed in open-top fumigation chambers were exposed to O3 (charcoal-filtered air, non-filtered air, 1.5 × ambient, or 2 × ambient) and simulated rain (pH 3.1, 4.1 or 5.1) for two growing seasons (June to October). Foliage was sampled for mineral nutrients, total soluble sugars, and starch in September 1988 at the end of the second season of exposures. The development of cold tolerance of individual trees was assessed using the electrolyte leakage technique. Ozone and simulated rain treatments had marginal effects on mineral nutrient concentrations of current and 1 yr old foliage. Ozone did not affect foliar carbohydrate levels but the simulated rain treatments of pH 3.1 tended to depress levels of total soluble sugars and starch in 1 yr old foliage and of starch in current year's foliage. During mid to late October, the current year's foliage of trees receiving rain of pH 3.1 was less tolerant to cold than the current year's foliage of trees receiving rain of pH 4.1 or 5.1. Following the October period, trees in all three acidic rain treatments had similar tolerances to cold.  相似文献

This paper addresses the development of organic agriculture in the Czech Republic, which is seen as a success story among post-communist countries. The relatively short history of organic farming and specific contextual factors raises questions about the nature and meaning of Czech organic farming. The goal of this study was to find out how farmers view their own practice, interpret its symbolic value, and construct its content. This empirical study uses Q methodology aimed at the identification of the collectively-shared perspectives belonging engaged actors. Data were gathered through semi-standardized interviews with Czech farmers registered in official organic scheme. The analysis emphasized three components, which are considered as three distinct perspectives possessed by organic farmers; that is, (1) organic farming as a way of life, (2) as an occupation, and (3) as a production of food of an alternative quality compared to conventional food. Each viewpoint entails a different understanding of what organic farming means; each then—when considered together—comprises the meaning of organic agriculture in the Czech Republic. The presented classification of the farmers holding the viewpoints contributes to the ongoing theoretical discussion regarding the nature of the current organic sector, its development and potential conventionalization.  相似文献

DNA replication in eukaryotes requires nucleosome disruption ahead of the replication fork and reassembly behind. An unresolved issue concerns how histone dynamics are coordinated with fork progression to maintain chromosomal stability. Here, we characterize a complex in which the human histone chaperone Asf1 and MCM2-7, the putative replicative helicase, are connected through a histone H3-H4 bridge. Depletion of Asf1 by RNA interference impedes DNA unwinding at replication sites, and similar defects arise from overproduction of new histone H3-H4 that compromises Asf1 function. These data link Asf1 chaperone function, histone supply, and replicative unwinding of DNA in chromatin. We propose that Asf1, as a histone acceptor and donor, handles parental and new histones at the replication fork via an Asf1-(H3-H4)-MCM2-7 intermediate and thus provides a means to fine-tune replication fork progression and histone supply and demand.  相似文献
