Industrial proteins from agriculture of either animal or vegetable origin, including their peptide derivatives, are of great importance, from the qualitative and quantitative point of view, in food formulations (emulsions and foams). A fundamental understanding of the physical, chemical, and functional properties of these proteins is essential if the performance of proteins in foods is to be improved and if underutilized proteins, such as plant proteins (and their hydrolysates and peptides derivatives), are to be increasingly used in traditional and new processed food products (safe, high-quality, health foods with good nutritional value). In this contribution we have determined the main physicochemical characteristics (solubility, composition, and analysis of amino acids) of a sunflower protein isolate (SPI) and its hydrolysates with low (5.62%), medium (23.5%), and high (46.3%) degrees of hydrolysis. The hydrolysates were obtained by enzymatic treatment with Alcalase 2.4 L for DH 5.62 and 23.5% and with Alcalase 2.4 L and Flavorzyme 1000 MG sequentially for DH 46.3%. The protein concentration dependence on surface pressure (surface pressure isotherm), a measure of the surface activity of the products (SPI and its hydrolysates), was obtained by tensiometry. We have observed that the degree of hydrolysis has an effect on solubility, composition, and content of the amino acids of the SPI and its hydrolysates. The superficial activity and the adsorption efficiency were also affected by the degree of hydrolysis.  相似文献

The effects of three fungicide residues (cyprodinil, fludioxonil, and pyrimethanil) on the aromatic composition (acids, alcohols, and esters) of Vitis vinifera white wines (var. Airén) inoculated with three Saccharomyces cerevisiae strains (syn. bayanus, cerevisiae, and syn. uvarum) are studied. The aromatic exponents were extracted and concentrated by adsorption-thermal desorption and were determined by gas chromatography using a mass selective detector. The addition of the three fungicides at different doses (1 and 5 mg/L) produces significant differences in the acidic fraction of the aroma, especially in the assays inoculated with S. cerevisiae, although the final contents do not exceed the perception thresholds. The lower quality wines, according to isomeric alcohol content [(Z)-3-hexen-1-ol and 3-(methylthio)propan-1-ol] are those obtained by inoculation with S. cerevisiae(syn. bayanus) and addition of cyprodinil. The addition of fungicides in the assays inoculated with S. cerevisiae (syn. bayanus) produces an increase in the ethyl acetate and isoamyl acetate contents, which causes a decrease in the sensorial quality of the wine obtained.  相似文献

Background  Arsenic (As) availability in natural environment is related to the element’s adsorption and desorption processes in soils. Total As is better related to available As in temperate soils than in tropical soils. In tropical soils, total As is not very significant in terms of availability, therefore justifying the necessity for studies into As dynamics. Knowledge of As dynamics in soil as well as development of new analytical methodologies involving tropical soils are insufficient and necessary for future mitigation projects. Objective  The objectives of this study were: (1) To adjust methodologies which may assist in understanding arsenate dynamics in tropical soils and substrates; (2) To evaluate the adsorption and desorption of arsenate in soils and substrate samples, and to find a minimum value of arsenate available in soil which is lethal to sorghum plants. Material and Methods  Samples of three soils from Minas Gerais State (YL, RYL, and CS) and two sulfide substrates of gold mining (B1 and B2) were used in the assays. All the material was physically and chemically characterized. Remaining As (As-rem) and remaining P (P-rem) of each material, along with MACP and MACAs (using the Langmuir isotherms), were obtained. After agitation to obtain MACP and MACAs, arsenate was extracted by anionic resin and Mehlich-III to evaluate arsenate desorption of the material retained on the filter paper. Subsequently, arsenate desorption curves for the different materials were obtained, and arsenate availability was determined through a bioassay with sorghum plants. Samples of soils and substrate B1 were incubated with six levels of As doses. Plants were grown under greenhouse conditions for 30 days. The plants were then harvested, dried and weighed. Available As in the soils and substrate was determined by Mehlich-III. Results and Discussions  As-rem level decreased from YL (sandy) to RYL (clayey) soil samples, which always showed lower values than P-rem. Among the soils and substrates evaluated, RYL showed the highest MACAs and MACP, followed by CS, YL and Bl. The results were in accordance with the values observed for As-rem and P-rem and confirm the idea that the ability of the assayed materials to remove As from the soil/substrate solution is higher than the ability to remove P. On the other hand, the binding energy (a) between soil/substrate and As is weaker than the binding energy of P. Given the fact that the studied soils present a real ability to remove As from the solution, only a small part of As would be unavailable considering MACAs as a reference. As-Mehlich-III values were higher than As-resin for substrate Bl. Mehlich-III seemed to be more appropriate to extract labile forms of arsenate in substrate B1 as well as in the soils. Available As by Mehlich-III (26.9 mg/dm3) was considered a reference of As LCL to sorghum plants. CC50 was sensitive to the buffering capacity of each soil, showing values varying from 1.34 mg/dm3 As (clay soil with lower As-rem) to 12.31 mg/dm3 As (sandy soil with higher As-rem). Conclusions  The adaptation of the As-rem and MACAs methodologies was satisfactory and of great value in the study of adsorption, desorption and As availability for soils and mining substrate. Mehlich-III was also satisfactory to estimate available As and was sensitive to soil buffering capacity. Nevertheless, resin can also be used as an alternative. MACAs varied among soils and was higher than MACp. However, As showed higher lability than P. Using Mehlich-III, we determined the value corresponding to CC50 that showed a good reference of toxicity to available As. Outlook  The environmental implications of the As behavior are quite serious. Beside the fact that arsenate is removed very fast from the soil solution, an anthropogenic input of the element, being part of the soil quantity factor, may remain in a reversible form for a long time. As may therefore return to the soil solution and becomes available to plants, animals and the entire environment. Considering that CC50 is the maximum contents of available As the environment can tolerate to allow some vegetal biomass production, the maximum capacity of As immobilization in each soil is reduced when compared to the soils’ MACAs values. Therefore, the maximum and safe values of reference to be used in the evaluation of incidental discharge of the element in soils must be reduced.  相似文献

Residue levels of pyrifenox, pyridaben, and tralomethrin were determined in unprocessed and processed tomatoes, grown in a experimental greenhouse, to evaluate the effect of three different household processes (washing, peeling, and cooking) and the "unit to unit" variability of these pesticides in tomatoes. The study was carried out on 11 greenhouse tomato samples collected during a 5 week period in which two successive treatments with the studied pesticides were applied. Residue levels in unprocessed and processed tomato samples were determined by means of ethyl acetate extraction and gas chromatography-electron capture detection determination. The washing processing factor results were 0.9 +/- 0.3 for pyridaben, 1.1 +/- 0.3 for pyrifenox, and 1.2 +/- 0.5 for tralomethrin, whereas the peeling processing factors were 0.3 +/- 0.2 for pyridaben and 0.0 +/- 0.0 for both pyrifenox and tralomethrin. The average loss of water in the tomato pure samples during the cooking process was approximately 50%; the cooking processing factors were 2.1 +/- 0.8 for pyridaben, 3.0 +/- 1.1 for pyrifenox, and 1.9 +/- 0.8 for tralomethrin. The unit-to-unit variability factors were determined on three different greenhouse samples analyzing 10 different units of unprocessed tomatoes from each sample. In all cases, the unit-to-unit variability factor results were within the range of 1.3-2.2.  相似文献

Tristimulus Colorimetry was applied to characterize the color of Valencia late orange juices. Color measurements were made against white background and black background. The profile of the main carotenoids related to the color of the juices was determined by HPLC. Significant correlations (p < 0.05) between b*, Cab* and h(ab) and the content of beta-cryptoxanthin, lutein + zeaxanthin and beta-carotene were found. The correlations between the color parameters L*, a*, b*, Cab* and h(ab) and the carotenoids content were also explored by partial least squares. The results obtained have shown that it is possible to obtain equations, by means of multiple regression models, which allow the determination of the individual carotenoid levels from the CIELAB color parameters, with R2 values always over 0.9. In this sense, equations have been proposed to calculate the retinol equivalents (1 RE = 1 microgram retinol = 12 micrograms beta-carotene = 24 micrograms alpha-carotene = 24 micrograms beta-cryptoxanthin) of the orange juice analyzed as a function of the color parameters calculated from measurement made against white and black backgrounds. The average RE per liter of juice obtained by HPLC was 51.07 +/- 18.89, whereas employing these equations, average RE values obtained were 51.16 +/- 1.36 and 51.21 +/- 1.70 for white background and black background, respectively.  相似文献

(13)C NMR spectra of oil fractions obtained chromatographically from 66 vegetable oils were obtained and analyzed to evaluate the potential use of those fractions in predicting oil stabilities and to compare those results with oil stability prediction by using chemical determinations. The oils included the following: virgin olive oils from different cultivars and regions of Europe and north Africa; "lampante" olive, refined olive, refined olive pomace, low-erucic rapeseed, high-oleic sunflower, corn, grapeseed, soybean, and sunflower oils. Oils were analyzed for fatty acid and triacylglycerol composition, as well as for phenol and tocopherol contents. By using stepwise linear regression analysis (SLRA), the chemical determinations and the (13)C NMR data that better explained the oil stability determined by the Rancimat were selected. These selected variables were related to both the susceptibility of the oil to be oxidized and the content of minor components that most contributed to oil stability. Because (13)C NMR considered many more variables than those determined by chemical analysis, the predicted stabilities calculated by using NMR data were always better than those obtained by using chemical determinations. All these results suggest that (13)C NMR may be a powerful tool to predict oil stabilities when applied to chromatographically enriched oil fractions.  相似文献

Controlling gut inflammation is important in managing gut disorders in the piglet after weaning. Establishing patterns of inflammation markers in the time subsequent to weaning is important for future research to determine whether interventions are effective in controlling gut inflammation. The objective of this study was to evaluate the intestinal inflammatory response during the postweaning period in piglets. A 45-d study included 108 piglets (weaned at 22 d, body weight 5.53 ± 1.19 kg), distributed in 12 pens with nine pigs per pen. Histomorphometry, gene expression of pro- and anti-inflammatory cytokines, and the quantity of immunoglobulin (Ig) A producing cells were measured in jejunum, ileum, and colon on days 0, 15, 30, and 45 postweaning. Cytokine gene expression in peripheral blood mononuclear cells and Ig quantities were analyzed in blood from piglets on days 0, 15, 30, and 45 postweaning. Histomorphometrical results showed a lower villus length directly after weaning. Results demonstrated a postweaning intestinal inflammation response for at least 15 d postweaning by upregulation of IgA producing cells and IFN-γ, IL-1α, IL-8, IL-10, IL-12α, and TGF-β in jejunum, ileum, and colon. IgM and IgA were upregulated at day 30 postweaning. IgG was downregulated at day 15 postweaning. The results indicate that weaning in piglets is associated with a prolonged and transient response in gene expression of pro- and anti-inflammatory cytokines and IgA producing cells in the intestine.  相似文献

A longitudinal study was conducted to identify the morphological development of eyes and head of Jersey cattle at different ages. A total of 48 healthy Jersey cattle, 1 to 58 months of age, were included in the study. Ophthalmologic examinations were performed to identify healthy cattle to form the age groups: GI (1- to 11-month-old), GII (16- to 24-month-old) and GIII (25- to 58-month-old). The animals were physically restrained and examined by transcorneal ultrasound of both eyes; the axial length (AxL), corneal thickness (CO), lenticular thickness (L), depth of the anterior (AC) and vitreous (VC) chambers were evaluated. The cranial measurements obtained included the total, cranial, and nasal lengths and widths. Subsequently, the cephalic index (CI) was calculated. The AxL (GI: 2.83 cm; GII: 3.16 cm; GIII: 3.24 cm), AC (GI: 0.44 cm; GII: 0.53 cm; GIII: 0.53 cm), L (GI: 0.88 cm; GII: 1.01 cm; GIII: 1.04 cm) and VC (GI: 1.44 cm; GII: 1.55 cm; GIII: 1.59 cm) increased (p ≤ .001) according to age, but the CO (GI: 0.07 cm; GII: 0.08 cm; GIII: 0.08 cm) (p > .05) did not. The total length (GI: 30.83 cm; GII: 43.29 cm; GIII: 44.15 cm), cranial length (GI: 18.11 cm; GII: 23.82 cm; GIII: 22.69 cm), nasal length (GI: 12.72 cm; GII: 19.47 cm; GIII: 21.46 cm) and nasal width (GI: 26.22 cm; GII: 33.82 cm; GIII: 34.00 cm) increased (p < .001), and the CI (GI: 85.66 cm; GII: 78.15 cm; GIII: 77.02 cm) decreased (p < .001). The AxL correlated positively (p < .001) with the cranial length and width and negatively with the CI (p > .05). The ocular biometry of Jersey cattle correlates with their cranial measurements at different ages. With increasing age, the eyes and the head grow, establishing growth curves similar to those of other mammals.  相似文献

Modern and paleoclimate changes may have altered species dynamics by shifting species’ niche suitability over space and time. We analyze whether the current genetic structure and isolation of the two large American felids, jaguar (Panthera onca) and puma (Puma concolor), are mediated by changes in climatic suitability and connection routes over modern and paleoclimatic landscapes. We estimate species distribution under 5 climatic landscapes (modern, Holocene, last maximum glaciations [LMG], average suitability, and climatic instability) and correlate them with individuals’ genetic isolation through causal modeling on a resemblance matrix. Both species exhibit genetic isolation patterns correlated with LMG climatic suitability, suggesting that these areas may have worked as “allele refuges.” However, the jaguar showed higher vulnerability to climate changes, responding to modern climatic suitability and connection routes, whereas the puma showed a continuous and gradual transition of genetic variation. Despite differential responsiveness to climate change, both species are subjected to the climatic effects on genetic configuration, which may make them susceptible to future climatic changes, since these are progressing faster and with higher intensity than changes in the paleoclimate. Thus, the effects of climatic changes should be considered in the design of conservation strategies to ensure evolutionary and demographic processes mediated by gene flow for both species.  相似文献
