The role of different factors such as biological material (tissues, organs) and trophic condition (overfeeding or not) in the metal accumulation was studied in three genotypes of ducks (Pekin, Muscovy, and Mule) under breeding conditions. Results showed that overfeeding decreased the concentration in Cd, Cu, and Zn through the dilution process. In contrast, mercury concentration increased with this method. A relation between lipidic metabolism of genotypes and the distribution of this metal in biological material was found. Domestic ducks were little contaminated, but a low chronic contamination in Cd was observed, probably coming from the food. Due to the low levels of contamination observed in these breeding ducks, they can be considered as a good control for further contamination studies and comparison with accumulation levels recorded in the field. The impact of feeding condition on accumulation showed the importance of taking into account the life cycle of birds before studying their contamination and the impact of pollutants. 相似文献

Sulfonamide antibiotics reach soil via manure and adversely affect microbial diversity. Clear effects of these bacteriostatic, growth‐inhibiting antibiotics occur in the presence of a parallel input of microbial activity stimulating manure. Natural hot spots with already increased soil microbial activity are located in the rhizosphere, comprising microorganism such as Pseudomonas with plant growth promoting and pathogenic strains. The hypothesis was therefore that the antibiotic activity of sulfonamides is promoted in the rhizosphere even in the absence of manure, followed by shifts of the natural plant‐specific microbial community structure. This was evaluated by a laboratory experiment with Salix fragilis L. and Zea mays L. After 40 d of incubation, sub‐areas such as non‐rhizosphere soil, rhizosphere soil and plant roots were sampled. Effects on microbial community structure were analyzed using 16S rRNA gene fragment patterns of total bacteria community and Pseudomonas. Selected exoenzymes of N‐, P‐, and C‐cycling were used to test effects on microbial functions. Compared to the factors soil sub‐area and sulfadiazine (SDZ) content, plant species had the largest influence on the bacterial community structure and soil exoenzyme activity pattern. This was also reflected by an up to 1.5‐fold higher acid phosphatase activity in samples from maize‐ compared to willow‐planted soil. A multiplex real-time PCR was designed to detect and differentiate equid herpesvirus 1 (EHV-1) and equid herpesvirus 4 (EHV-4). The PCR targets the glycoprotein B gene of EHV-1 and EHV-4. Primers and probes were specific to each equine herpesvirus type and can be used in monoplex or multiplex PCRs, allowing the differentiation of these two closely related members of the Alphaherpesvirinae. The two probes were minor-groove binding probes (MGB) labelled with 6-carboxy-fluorescein (FAM) and VIC for detection of EHV-1 and EHV-4, respectively. Ten EHV-1 isolates, six EHV-1 positive clinical samples, one EHV-1 reference strain (EHV-1.438/77), three EHV-4 positive clinical samples, two EHV-4 isolates and one EHV-4 reference strain (EHV-4 405/76) were included in this study. EHV-1 isolates, clinical samples and the reference strain reacted in the EHV-1 real-time PCR but not in the EHV-4 real-time PCR and similarly EHV-4 clinical samples, isolates and the reference strain were positive in the EHV-4 real-time PCR but not in the EHV-1 real-time PCR. Other herpesviruses, such as EHV-2, EHV-3 and EHV-5 were all negative when tested using the multiplex real-time PCR. When bacterial pathogens and opportunistic pathogens were tested in the multiplex real-time PCR they did not react with either system. The multiplex PCR was shown to be sensitive and specific and is a useful tool for detection and differentiation of EHV-1 and EHV-4 in a single reaction. A comprehensive equine herpesvirus disease investigation procedure used in our laboratory is also outlined. By using CBA/J mice as responders, the immunodepressive effect of seven antibacterial chemotherapeutic agents was tested, i.e. penicillin (pen), streptomycin (str), erythromycin (erm), kanamycin (kan), tetracycline (tet), colistin (col) and chloramphenicol (chl). Lymphocytotoxicity power, as well as the ability of each drug to influence secondary humoral (against sheep red blood cells or diphtheria anatoxin) or cell-mediated (against PPD and Coxsackie A9 virus) immunity were searched.
Erm, col and chl markedly depressed humoral and cell-mediated immune responsiveness in vitro after in vivo treatment with non-cytotoxical amounts. The Th lymphocyte supplementation of B reactive spleen cell population recovered the immune capacity in col and chl in vivo-treated groups, but not in erm-treated group. 相似文献

Hardwoods contain a substantial amount of hemicellulose-type oligosaccharides, the chemical structures of which are typically categorized in the xylan saccharide class. The efficient and selective isolation of these hemicelluloses is regarded as one of the most critical obstacles to overcome for their eventual biomaterial and bioenergy utilization. Therefore, the objective of the current work was to perform a fundamental exploration of the function of extraction pH, temperature, and time on the final chemical properties of the extracted hemicelluloses. The extraction chemistries employed varied pH and showed that acidic conditions provided a higher extractive yield versus alkaline conditions. An alkaline environment gave higher yields than an acidic environment and also resulted in much higher lignin removal from wood, especially at high temperature. In general, control of the pH chemistry leads to a change in the carboxylic acid groups present in the extracts. When the extraction pH changed from acidic to alkaline, the total carboxylic acid group content in the hemicelluloses removed significantly increased due to more acetic acid group hydrolysis and increased lignin degradation. 相似文献
