A reverse-phase liquid chromatographic method for the assay of sulfamethazine (SMZ) in feeds is described. Feed samples are extracted with 50% methanol solution, centrifuged, filtered, and diluted when necessary, and chromatographed on a C-18 column. Samples are eluted with a mobile phase of 20% methanol and 80% of a solution containing acetic acid and tetramethylammonium chloride. The average recovery from spiked samples was 97.2% with a coefficient of variation of 1.2%. Linearity was very good (correlation coefficient 0.9997). Within-day and between-day coefficients of variation averaged 1.3 and 2.6%, respectively. The results for samples assayed by this method compared closely with the results from the same extracts assayed by the AOAC colorimetric method. 相似文献

Modern agriculture involves mechanized operations which affect crop growth and yields through changes in the soil environment. Field research to study the phenomena involved in the soil-machine-crop system is usually site-specific with respect to edaphic, climatic and management factors. Field experiments are often expensive to conduct, and generalization to other sites or practices is usually tenuous at best. Computer-based techniques to simulate machine-soil-plant (or crop) systems can test many combinations of site characteristics and management practices, and screen the most promising combinations. Once these promising combinations are validated, the simulation techniques, together with stochastically-supported data bases can be used to predict tillage management systems suitable for specific sites and crops.Current efforts to model plant response to soil conditions created by tillage and machines have serious shortcomings, because the link between tool action, subsequent soil structure, and the parameters that describe fluxes of water, heat and gas need further development for deterministic modeling. Machine-soil models focus on efficiency of the tillage operation, while soil-plant models focus on soil structure and related parameters influencing plant growth. While current models for simulating the machine-soil-crop system have limitations, an approach to bridge between the two model applications offers great promise and should be a high research priority. When used with improved measurements of tool action and of soil structural properties before and after tillage and traffic, including position or placement of the crop residues, current models can be doubly effective for improving tillage management. 相似文献

The differential distributions of IgG1 and IgG2 were determined in prepartum and lactating bovine mammary tissue by indirect immunofluorescence. IgG1 was found predominately within the alveolar epithelial cells and lumens of prepartum tissue whereas IgG2 was largely confined to the stromal area surrounding the alveoli. Both IgG subclasses were confined predominately to the stroma in lactating tissue. Few IgG containing stromal cells were readily distinguished in any of the mammary tissue used in this study. 相似文献

Lactoferrin (LF) is an iron-binding protein present in both colostrum and secondary granules of polymorphonuclear neutrophils (PMNs). We hypothesized that supplemental LF enhances neutrophil function in neonatal calves. Newborn calves were assigned to receive colostrum (C), colostrum + LF (CLF, 1 g/kg), or milk replacer + LF (MRLF, 1 g/kg). Serum (LF and IgG) and whole blood (neutrophil isolation) samples were obtained prior to treatment (day 0) and at 24 hours and 9 days of age. Serum IgG concentrations (mean +/- SD) in C, CLF, and MRLF calves at 24 hours were 1,911 +/- 994 mg/dL, 2,181 +/- 625 mg/dL, and 0 mg/ dL, respectively. Serum LF concentrations in C, CLF, and MRLF calves on day 0 were 324 +/- 334 ng/mL (range 0-863 ng/mL), 135 +/- 158 ng/mL (range 0-429 ng/mL), and 318 +/- 337 ng/mL (range 0-964 ng/mL), respectively. LF concentrations in C, CLF, and MRLF calves at 24 hours were significantly higher (P < .05), at 1,564 +/- 1,114 ng/mL (range 335-3,628 ng/mL, 2,237 +/- 936 ng/mL (range 31-3,287 ng/mL), and 3,189 +/- 926 ng/mL (range 1,736-4,120 ng/mL), respectively. Cytochrome c reduction in opsonized zymosan-treated or phorbol ester-treated cells was not significantly affected by supplemental LF provided at birth.
