Summary A new method is presented for anther culture. Anthers are cultivated on cubes of solid medium, surrounded by liquid medium. This allows changes of media composition at any time. When embryos or callus were produced, the liquid initiation medium was removed and replaced with regeneration medium. The best yield of embryos and the best regeneration frequency were obtained when the initiation medium contained 2,4-dichlorophenoxyacetic acid (2,4-D). Regeneration of shoots from callus was stimulated on double layer medium, with the callus placed on top of a solid medium, partly submerged in the overlaying liquid medium. The best gelling agent for shoot multiplication media was agarose, but gellan gum was a good alternative. Both the production of flower buds and embryogenesis in anther cultures was inhibited by an increased concentration of CO2 in the air surrounding the donor plants.  相似文献

Field-cured onions cv. Hyskin ( Allium cepa L.) supplied with organic nitrogen fertilizer were studied. The fertilizer was applied by broadcasting and harrowing, broadcasting and rotary cultivation, or placement between rows. Nitrogen dynamics were monitored throughout the growing season by soil sampling. Variation in quercetin content in the onion scales was analyzed by HPLC. The organically fertilized onions were compared with inorganically fertilized onions grown in the same field. Inoculation with arbuscular mycorrhizal fungi (AMF) in the row at sowing or during commercial transplant production was tested but did not significantly affect mycorrhizal root colonization levels in the field. Onions that received no fertilizer at all or that had fertilizer placed between rows had better establishment, probably due to more favorable soil nitrogen concentrations for seedling emergence. Broadcast application led to higher nitrogen concentration in the root zone, resulting in fewer but larger individual onions. Quercetin levels were not significantly altered as a result of nitrogen fertilizer source (inorganic or organic), application method, or mycorrhizal inoculation. However, variation between years was significant, with quercetin levels in 2004 almost twice as high as those in 2005.  相似文献

The behavior of antioxidants in emulsions is influenced by several factors such as pH and emulsifier type. This study aimed to evaluate the interaction between selected food emulsifiers, phenolic compounds, iron, and pH and their effect on the oxidative stability of n-3 polyunsaturated lipids in a 10% oil-in-water emulsion. The emulsifiers tested were Tween 80 and Citrem, and the phenolic compounds were naringenin, rutin, caffeic acid, and coumaric acid. Lipid oxidation was evaluated at all levels, that is, formation of radicals (ESR), hydroperoxides (PV), and secondary volatile oxidation products. When iron was present, the pH was crucial for the formation of lipid oxidation products. At pH 3 some phenolic compounds, especially caffeic acid, reduced Fe(3+) to Fe(2+), and Fe(2+) increased lipid oxidation at this pH compared to pH 6. Among the evaluated phenols, caffeic acid had the most significant effects, as caffeic acid was found to be prooxidative irrespective of pH, emulsifier type, and presence of iron, although the degrees of lipid oxidation were different at the different experimental conditions. The other evaluated phenols were prooxidative at pH 3 in Citrem-stabilized emulsions and had no significant effect at pH 6 in Citrem- or Tween-stabilized emulsions on the basis of the formation of volatiles. Throughout the northern hemisphere old forests with high abundance of dead wood are rare features in most landscapes today, and the loss of dead wood constitutes a serious threat to the existence of many species. This study, using field surveys and dendrochronology, examines the relationship between wood-inhabiting fungi and past forest utilisation along a gradient of early logging activity. Data were collected in three late-successional Scots pine forests in northern Sweden. Nonmetric Multidimensional Scaling (NMS) was then used to assess differences in species composition among the forests. Our results show that minor forest logging (22-26 cut stumps ha−1) carried out a century ago may have continuing effects on forest characteristics, including dead wood dynamics and the wood-inhabiting fungal community - especially the abundance of red-listed species. The most important effects are lower numbers of logs in early and intermediate stages of decomposition. Additionally, numbers of species (including red-listed species) can be high in forests that have been subject to low levels of logging. Overall, the high species numbers recorded in this study (= 60-87) show that old, low-productivity pine forests harbour a considerable fraction of the total diversity of Basidiomycetes in northern Fennoscandian boreal forests. We conclude that the formation of a framework linking forest history and environmental data is vital for understanding the ecology and formulating goals for future management of these forests.  相似文献

A method for simultaneous determination of ginsenosides and polyacetylenes in Panax quinquefolium L. (American ginseng) roots was developed. The ginsenosides Rb1, Rb2, Rc, Rd, Re, Rg1, Ro, malonyl-Rb1, malonyl-Rc, and malonyl-Rd and the polyacetylenes falcarinol and panaxydol were extracted from fresh ginseng roots in a sequential extraction process with 100% methanol followed by 80% aqueous methanol and quantified simultaneously in extracts by high-performance liquid chromatography using diode array detection. Separations were achieved with a phosphate buffer-acetonitrile gradient system using an RP-C18 column. Except for Rd, the present extraction method resulted in similar or significantly higher concentrations of both ginsenosides and polyacetylenes in comparison to commonly used extraction methods for these compounds. The contents of polyacetylenes and ginsenosides were determined in the root hairs, lateral roots, and main roots of 6 year old ginseng plants. The total mean concentrations of ginsenosides and polyacetylenes in root hairs were 31.0 g/kg fresh weight (FW) and 2.6 g/kg FW, respectively, whereas the concentrations of these bioactive compounds in the main roots were significantly lower with total mean concentrations of 17.8 g/kg FW for ginsenosides and 0.6 g/kg FW for polyacetylenes. The concentration of individual and total ginsenosides and polyacetylenes did not differ significantly between main roots of different sizes. Consequently, it is possible to do quantitative screening for ginsenosides and polyacetylenes to breed ginseng roots with higher levels of bioactive compounds.  相似文献

Variation in quercetin content was investigated in field-cured onions (Allium cepa L.) that had been supplied with different nitrogen fertilizer levels and lifted at different developmental stages. Quercetin content varied significantly between years and was well correlated to global radiation in August. Field curing generally resulted in significant increases in quercetin content compared to levels at lifting. Nitrogen fertilizer level did not affect quercetin content, suggesting that nitrogen leakage from soil may be minimized without effects on flavonol content. Lifting time had minor effects on quercetin content in field-cured onions. Cultivar differences in quercetin content were significant but not consistent in all years. Quercetin content increased significantly less in dark environments compared to field curing, but some quercetin synthesis occurred regardless of light. Field curing with or without foliage still attached did not affect quercetin content, suggesting that no transportation from the foliage to the scales occurred.  相似文献

The aim of the present study was to determine the efficacy of the bone markers osteocalcin (OC) and carboxyterminal cross-linked telopeptide of type-I collagen (ICTP) in evaluating new bone formation in the dog, using commercially available immunoassay kits. Dogs were randomly divided into three groups and a circular external skeletal fixation system (CESF) was mounted on the tibia. In the first group a distraction osteogenesis procedure of the crus was performed. The second group received an osteotomy without crural lengthening, whereas the third group served as a sham-operated control. Bone formation was assessed using densitometric image analysis of crural radiographs. Despite significant differences in the amount of newly formed bone, this finding was not reflected in the plasma levels of OC and ICTP. In conclusion, OC and ICTP were not efficacious as markers of bone formation and resorption during osteogenesis in this canine model.  相似文献

Purpose: Root and root hairs of plants have been intensively studied in solution culture; however, correlation of such measurements in solution culture with development in soil is poorly understood. Therefore, the aim of this study is to study whether root and root hairs grown in solution culture can predict their behavior in soil and their correlation with macro- and micronutrients uptake of wheat genotypes.

Materials and methods: The growth of roots and root hairs as well as uptake of macro- and micronutrients of six spring wheat varieties was compared in solution culture under P stress and P abundance and in a low fertility soil.

Results and conclusions: Root length and surface area under P stress were significantly positively correlated with that in the low fertility soil, while no such correlation was apparent for root hair length and density. In absolute terms, the root length, surface area, root hair length and density of spring wheat varieties were substantially higher in soil than in solution culture, while the concentration and uptake of macro- and micronutrients in soil differed from solution culture in a complex way. The early uptake of macro- and micronutrients was intimately associated with root length and surface area as well as root hair length and density in soil but not in solution culture. Therefore, root length rather than root hair traits in low-P solution may be used to screen early root growth vigor in soil and thereby high nutrient uptake of wheat in low fertility soil.  相似文献

Current information on the root sucker ability of hybrid aspen (Populus tremula L. × P. tremuloides Michx.) is in most cases based on clone mixtures. In this study, we could separate the performance among clones by using two experimental sites with monoclonal plots of the crossing. The natural regeneration was followed for 2 years after harvest of the formerly planted stands, clear-cut at 22 and 25 years of age, respectively. We found that clonal differences were present in the number of root suckers produced per area unit and their biomass production. However, all included clones produced sufficiently many root suckers for a successful regeneration. To receive a more complete evaluation of the root sucker dynamics for future selection on the clonal level, further studies are needed where vitality, self-thinning and effects of thinning measures are coupled to the goal of the aspen forestry.  相似文献

This data set provides unique empirical data from triplets of Scots pine ( Pinus sylvestris L.) and European beech ( Fagus sylvatica L.) across Europe. Dendrometric variables are provided for 32 triplets, 96 plots, 7555 trees and 4695 core samples. These data contribute to our understanding of mixed stand dynamics.
