Comparative studies of two cultivated and sixteen wild species of the genus Oryza were carried out using one- and two-dimensional gel electrophoresis for variation in their seed proteins for interrelationships of these species. A number of polypeptides in the range of molecular weight 13–110 kDa were seen to vary. Under reducing conditions, polypeptides spread over the regions of mol. wt. 33–40.5, 25–27 and 19–21.5 kDa exhibited maximum variation in their patterns. Two-dimensional gel electrophoresis revealed the occurrence of disulphide-linked glutelin polypeptide pairs of mol. wt. 60, 58, 52 and 25 kDa breaking into a large and a small subunit each in the range of mol. wt. 18–40.5 and 16–25 kDa respectively in Oryza sativa. The number of such polypeptide pairs varied from 2 to 6 in different species and also in O. sativa showed variation in mol. wts. of their constituent subunits. The UPGMA dendrogram revealed that most of the Oryza species occurred in different clusters and subclusters and thus did not share very close relationships. The undisputed and closest relationship observed was that of cultivated rice O. sativa with the O. rufipogon followed by that with O. nivara. The African cultivated O. glaberrima occurring on the nearest branch of the same subcluster, thereby, supporting the phylogenetic of these species suggested in earlier studies. Eight diploid species and seven tetraploid species were included in one part of the dendrogram while the remaining two species with AA genome i.e. O. glumaepatula and O. meridionalis and one with FF i.e. O. brachyantha stood separately from these as scattered in the group of seven tetraploid species with BBCC, CCDD and HHJJ genomes. The tetraploids O. alta, O. latifolia and O. grandiglumis with CCDD genomes which occurred on the farthest part were distantly related with O. sativa. The cyanogen bromide peptide maps and two dimensional gel electrophoresis also supported the closest relationship between O. sativa and O. rufipogon. 相似文献

Summary A preliminary evaluation for resistance to chili thrips, Scirtothrips dorsalis Hood of 41 and 194 pepper (Capsicum annuum L.) germplasms during 1987 and 1988, respectively, indicated chili accessions may be a promising source of resistance. In contrast, all sweet pepper accessions tested were highly susceptible. A number of chili accessions produced a moderate yield, in spite of a high thrips infestation indicating tolerance to S. dorsalis. Rating for thrips damage was more reliable and efficient than estimating thrips numbers in screening pepper accessions for resistance to thrips. Highly significant, positive correlation between ratings at the seedling stage and final rating for thrips damage indicated the feasibility of screening pepper accessions at the seedling stage. Comparing the similarities in rating among accessions resulted in 40 distinct groups. The variance-covariance matrix of the data from these 40 groups was subjected to principal component analysis. This accounted for 56 and 18 per cent of the variation across the two principal axes, respectively. Projection of chili and sweet pepper accessions along these two axes revealed three distinct clusters. About 80 per cent of chili accessions formed the first cluster, 58 per cent of the sweet pepper accessions formed the second cluster and a third cluster exhibited intermediate ratings for thrips damage. The significance of these findings in relation to geographical divergence and resistance to thrips among pepper accessions is discussed. 相似文献

Northeast India is rich in Citrus genetic diversity representing several wild and cultivated species. Besides commercially cultivated species, several wild, semi-wild and domesticated species namely Citrus indica, C. macroptera, C. ichangensis, C. latipes, C. megaloxycarpa and C. assamensis are found to grow in Northeast India. These species have great potential in improvement of Indian Citrus industry being source of genes for combating biotic and abiotic stresses. The natural diversity of these genetic resources is shrinking at alarming rate due to large scale deforestation, shifting cultivation practiced in these areas and climate change scenario in this important hotspot of biodiversity. Socio-economic importance of each species have been studied during surveys and interesting domestication trends were observed based on their cultural and economic significance, which led to “in situ on farm” conservation of C. indica, C. macroptera and threat to C. megaloxycarpa and C. ichangensis. Consequently, genetic resources of most of these species are facing severe threat of extinction necessitating policy interventions and adoption of dynamic conservation and management strategies. Species specific conservation strategy especially through “in situ on farm” conservation has been proposed with suitable sites in Northeastern Indian states based on population structure and suitability of habitat. Farmers’ support through “The Biological Diversity Act, 2002” and “The Protection of Plant Varieties and Farmers’ Rights Act, 2001” of India would bring the required impact on management of these complex genetic resources of Citrus. In the present study, we have documented the current status, socio-economic potential, domestication trends and associated traditional knowledge of these wild and semi-wild Citrus species occurring in Northeast India and enumerated suitable conservation strategies and desired policy interventions for their dynamic conservation and sustainable utilization. 相似文献

The present study was undertaken with the objective of studying use and contribution of information sources in the purchase of agri-inputs by Indian farmers. The importance of an information source was measured in terms of its relevance as perceived by the farmers during the buying process. The study revealed that farmers preferred noncommercial, personal sources of information and the number of information sources used by them depended on the perceived importance of the product, the various options (brands) available to them, and credibility of the information sources. The personal characteristics of farmers did not have a significant influence on the number of information sources used by the farmers. 相似文献

Summary The merits and limitations of somatic cell techniques involving Agrobacterium-mediated transformation, direct gene transfer and protoplast fusion, are discussed in relation to the genetic improvement of forage and grain legumes. Whilst progress with legumes is limited compared to that with plants of other families such as the Solanaceae, the fact that many legumes are readily amenable to tissue culture now permits somatic cell techniques to be targetted to these species. Future development of the subject will necessitate close collaboration between molecular biologists and plant breeders to enable novel plants generated by in vitro technologies to be incorporated into conventional breeding programmes. 相似文献

Curcumin is a highly potent, nontoxic, bioactive agent found in turmeric and has been known for centuries as a household remedy to many ailments. The only disadvantage that it suffers is of low aqueous solubility and poor bioavailability. The aim of the present study was to develop a method for the preparation of nanoparticles of curcumin with a view to improve its aqueous-phase solubility and examine the effect on its antimicrobial properties. Nanoparticles of curcumin (nanocurcumin) were prepared by a process based on a wet-milling technique and were found to have a narrow particle size distribution in the range of 2-40 nm. Unlike curcumin, nanocurcumin was found to be freely dispersible in water in the absence of any surfactants. The chemical structure of nanocurcumin was the same as that of curcumin, and there was no modification during nanoparticle preparation. A minimum inhibitory concentration of nanocurcumin was determined for a variety of bacterial and fungal strains and was compared to that of curcumin. It was found that the aqueous dispersion of nanocurcumin was much more effective than curcumin against Staphylococcus aureus , Bacillus subtilis , Escherichia coli , Pseudomonas aeruginosa , Penicillium notatum , and Aspergillus niger . The results demonstrated that the water solubility and antimicrobial activity of curcumin markedly improved by particle size reduction up to the nano range. For the selected microorganisms, the activity of nanocurcumin was more pronounced against Gram-positive bacteria than Gram-negative bacteria. Furthermore, its antibacterial activity was much better than antifungal activity. The mechanism of antibacterial action of curcumin nanoparticles was investigated by transmission electron micrograph (TEM) analysis, which revealed that these particles entered inside the bacterial cell by completely breaking the cell wall, leading to cell death. 相似文献

Development of kunitz trypsin inhibitor (KTI)-free soybean is crucial for soy-food industry as the heat inactivation employed to inactivate the anti-nutritional factor in regular soybean incurs extra cost and affects protein solubility. In the presented work, a null allele of KTI from PI542044 was introgressed into cultivar ‘JS97-52’ (recurrent parent) through marker assisted backcrossing. Foreground selection in BC1F2, BC2F2 and BC3F2 was carried out using the null allele-specific marker in tandem with SSR marker Satt228, tightly linked with a trypsin inhibitor Ti locus. Background selection in null allele-carrying plants through 106 polymorphic SSR markers across the genome led to the identification of 9 KTI-free lines exhibiting 98.6% average recurrent parent genome content (RPGC) after three backcrosses, which otherwise had required 5–6 backcrosses through conventional method. Introgressed lines (ILs) were free from KTI and yielded at par with recurrent parent. Reduction of 68.8–83.5% in trypsin inhibitor content (TIC) in ILs compared to the recurrent parent (‘JS97-52’) was attributed to the elimination of KTI. 相似文献
