This study assessed the protective efficacy of oral vaccination with 2 experimental attenuated Salmonella Typhimurium-vectored vaccines for necrotic enteritis in protecting chickens against intestinal colonization by common serovars of Salmonella belonging to the 4 major serogroups affecting chickens. Birds were vaccinated orally with 1 × 108 colony-forming units (CFU) of 1 of the vaccine strains χ9241 and χ9352, which express a plasmid-encoded partial recombinant hypothetical protein gene (tHP) of Clostridium perfringens, at days 1 and 7 of age, and then were challenged at 14 d of age with 106 CFU of Salmonella serovars Anatum, Enteritidis, Heidelberg, Kentucky, or Typhimurium (representative serovars of serogroups B, C, D, and E). Birds were necropsied at 4 wk of age, and samples were collected to determine reduction in tissue and intestinal colonization. The chickens vaccinated with χ9241-tHP showed reduced colonization by Salmonella Enteritidis (serogroup D) and by Salmonella Heidelberg and Salmonella Typhimurium (serogroup B) compared with the control birds. No reduction in colonization was observed in the chickens vaccinated with χ9352-tHP. There was an association between the efficacy of these vaccine strains in protecting against necrotic enteritis, assessed on an earlier occasion, and their efficacy in protecting against Salmonella colonization. Thus, the choice of an attenuated Salmonella Typhimurium vaccine vector for delivery of heterologous antigens to chickens should be based partly on the vaccine’s value in protecting against colonization by serovars within serogroups B and D. We characterized Influenza A/H5N1 virus that caused the first outbreak of highly pathogenic avian influenza (HPAI) in chickens in Bhutan in 2010. The virus was highly virulent to chicken, killing them within two days of the experimental inoculation with an intravenous pathogenicity index (IVPI) of 2.88. For genetic and phylogenetic analyses, complete genome sequencing of 4 viral isolates was carried out. The isolates revealed multiple basic amino acids at their hemagglutinin (HA) cleavage site, similar to other "Qinghai-like" H5N1 isolates. The receptor-binding site of HA molecule contained avian-like amino acids ((222)Q and (224)G). The isolates also contained amino acid residue K at position 627 of the PB2 protein, and other markers in NS 1 and PB1 proteins, highlighting the risk to mammals. However, the isolates were sensitive to influenza drugs presently available in the market. The sequence analysis indicated that the Bhutan viruses shared 99.1-100% nucleotide homology in all the eight genes among themselves and 2010 chicken isolate from Bangladesh (A/chicken/Bangladesh/1151-11/2010) indicating common progenitor virus. The phylogenetic analysis indicated that the Bhutan isolates belonged to sub-clade 2.2.3 (EMA 3) and shared common progenitor virus with the 2010 Bangladesh virus. Based on the evidence of phylogeny and molecular markers, it could be concluded that the outbreaks in Bhutan and Bangladesh in 2010 were due to independent introductions of the virus probably through migratory birds. 相似文献

Compositional transformation of South African semiarid grasslands and savannas owing to changes in soil nutrient status and fire-linked attributes is often reported. However, mechanisms of change are not fully understood. Currently, plant-derived smoke has attracted much attention as a fire-related cue responsible for stimulating germination and seedling growth. However, there is very little documentation on how these fire-linked factors, such as smoke, and soil macronutrients, such as nitrogen (N), phosphorus (P), and potassium (K), interact to effect seedling growth of grasses. In this study, smoke-responsive (Themeda triandra) and less smoke-responsive species (Eragrostis curvula and Panicum maximum) were tested with different concentrations and combinations of smoke-water and smoke-isolated butenolide with or without added N, P, or K under greenhouse conditions. In the absence of N, P, or K, smoke-water and butenolide treatments enhanced a number of seedling growth parameters of T. triandra. In contrast, exclusion of N from the nutrient solution significantly reduced shoot length, seedling weight, root volume, and vigor index of E. curvula at all tested concentrations of smoke-water and butenolide solutions compared to the control. In the presence of N, P, and K, smoke-water and butenolide suppressed seedling growth of P. maximum, whereas the absence of one of these macronutrients had a small promotory effect on some parameters. Pregnancy-related changes in oxidative metabolism of several xenobiotics including pesticides were examined in the hepatic microsomes of CD1 mice. The effect of pregnancy on hepatic microsomal cytochrome P-450-catalyzed substrate oxidation was found to be dependent upon the type of reaction examined. Not all substrates undergoing the same reaction showed identical changes during pregnancy. Those enzyme activities which exhibited a decline in specific activity during pregnancy generally exhibited no change in total hepatic capacity. Enzymes posting no change in specific activity throughout gestation generally showed large increases in total hepatic activity. Phorate S-oxidation was catalyzed by both microsomal flavin-containing monooxygenase (MFMO) and cytochrome P-450. A survey of sheep and goat producers in the state of Maharashtra, India, was undertaken to ascertain the extent and economic impact of sheep pox and goat pox (SGP). One thousand one hundred and sixteen owners were interviewed. Eighty owners (7.2%) reported that they had experienced an outbreak of the disease in the previous 6 years. The results showed that, while producers ranked SGP below other infectious diseases such as foot-and-mouth disease, rinderpest and enterotoxaemia, when SGP occurred it had a major impact, with average morbidity and mortality rates of 63.5% and 49.5%, respectively. Modelling studies suggested it would take about 6 years for a flock or herd to recover from an outbreak, with average annual losses in income of 30–43%, depending on flock type and the owner's actions. Statewide, it is estimated that around 5000 flocks and herds are affected by SGP annually in Maharashtra, costing up to INR 107.5 million. The highest losses occurred in the Aurangabad region. 相似文献

Investigations were carried out on Helicoverpa armigera Nuclear Polyhedrosis Virus (HaNPV) isolates collected from different parts of India to study their genetic diversity through
molecular characterization and their assessment in the IPM of Helicoverpa armigera on chickpea under field conditions. Variations were found in the DNA profiling of different HaNPV isolates. The similarity
matrix relating the different isolates showed similarity co-efficients ranging from 0.38 to 0.82. The highest genetic similarity
index of 0.82 was seen between the isolates from Raichur and Guntur followed by 0.77 between the isolates from Coimbatore
and Raichur. Field evaluation of different HaNPV isolates against H. armigera in the chickpea ecosystem at the Main Agricultural Research Station, University of Agricultural Sciences, Dharwad, Karnataka
for 2 years revealed that the performance of the Gulbarga and Coimbatore isolates was better than that of others in terms
of their ability to cause greater larval mortality of H. armigera with a greater yield of chickpea. The Incremental Benefit Cost Ratio was highest with the Gulbarga (2.88) and Coimbatore
(2.83) isolates, followed by the Dharwad isolate (2.39).
