Background: Bile peritonitis is a severe, nonseptic inflammatory response to bile in the peritoneal cavity. It may result from generalized or localized leakage of bile due to spontaneous rupture of the biliary system or as a complication of biliary tract inflammation, obstruction, manipulation, or trauma. Cytologically, bile in abdominal fluid appears as golden-green granular pigment.
Objective: The purpose of this report is to describe the atypical cytologic features of abdominal fluid in 3 dogs with bile peritonitis.
Methods: As part of a diagnostic workup, abdominal fluid was obtained from 3 dogs with bile peritonitis and analyzed. In 2 dogs, fluid bilirubin concentration was determined and Hall's bile stain, Alcian blue-periodic acid-Schiff stain, and Mayer's mucicarmine stain were applied to direct smears of the fluid.
Results: Acellular mucinous fibrillar material in clumps and lakes was the prominent cytologic finding in the abdominal fluid from all 3 dogs. Bile pigment was not observed. Fluid from the 3 dogs contained increased numbers of inflammatory cells, predominantly neutrophils. Total protein concentration ranged from 2.9 to 5.6 g/dL. Fluid total bilirubin concentration was greater than twice that of the concurrent serum bilirubin concentration. Based on results of the special stains, the amorphous material was positive for mucosubstances, but was negative for bilirubin. In all dogs, bile peritonitis originated from a rent in the common bile duct.
Conclusions: Bile peritonitis with fibrillar mucinous material in abdominal fluid has not been described previously in dogs. The material was similar to "white bile" observed in humans and experimentally in dogs as a sequela to extrahepatic biliary tract obstruction. When mucinous material is observed in abdominal fluid from dogs and the fluid bilirubin concentration is greater than twice the serum bilirubin concentration, rupture of the extrahepatic biliary tract should be suspected.  相似文献

Agricultural practices have strong impacts on soil microbes including both the indices related to biomass and activity as well as those related to community composition. In a grassland restoration project in California, where native perennial bunchgrasses were introduced into non-native annual grassland after a period of intensive tillage, weeding, and herbicide use to reduce the annual seed bank, microbial community composition was investigated. Three treatments were compared: annual grassland, bare soil fallow, and restored perennial grassland. Soil profiles down to 80 cm in depth were investigated in four separate layers (0-15, 15-30, 30-60, and 60-80 cm) using both phospholipid ester-linked fatty acid (PLFAs) and ergosterol as biomarkers in addition to microbial biomass C by fumigation extraction. PLFA fingerprinting showed much stronger differences between the tilled bare fallow treatment vs. grasslands, compared to fewer differences between restored perennial grassland and annual grassland. The presence or absence of plants over several years clearly distinguished microbial communities. Microbial communities in lower soil layers were little affected by management practices. Regardless of treatment, soil depth caused a strong gradient of changing habitat conditions, which was reflected in Canonical Correspondence Analysis of PLFAs. Fungal organisms were associated with the presence of plants and/or litter since the total amount and the relative proportion of fungal markers were reduced in the tilled bare fallow and in lower layers of the grassland treatments. Total PLFA and soil microbial biomass were highly correlated, and fungal PLFA biomarkers showed strong correlations to ergosterol content. In conclusion, microbial communities are resilient to the grassland restoration process, but do not reflect the change in plant species composition that occurred after planting native bunchgrasses.  相似文献

Medulloblastoma (MB) is the most common malignant brain tumor of children. To identify the genetic alterations in this tumor type, we searched for copy number alterations using high-density microarrays and sequenced all known protein-coding genes and microRNA genes using Sanger sequencing in a set of 22 MBs. We found that, on average, each tumor had 11 gene alterations, fewer by a factor of 5 to 10 than in the adult solid tumors that have been sequenced to date. In addition to alterations in the Hedgehog and Wnt pathways, our analysis led to the discovery of genes not previously known to be altered in MBs. Most notably, inactivating mutations of the histone-lysine N-methyltransferase genes MLL2 or MLL3 were identified in 16% of MB patients. These results demonstrate key differences between the genetic landscapes of adult and childhood cancers, highlight dysregulation of developmental pathways as an important mechanism underlying MBs, and identify a role for a specific type of histone methylation in human tumorigenesis.  相似文献

OBJECTIVE: To determine the methicillin-resistant profile of staphylococcal isolates from the skin of dogs with pyoderma. ANIMALS: 90 dogs with pyoderma. PROCEDURE: Staphylococci isolated from dogs with pyoderma were tested for susceptibility to methicillin by use of a standard disk diffusion test with oxacillin disks. The DNA extracted from the isolates was tested for the mecA gene that encodes the penicillin-binding protein 2a (PBP2a) by use of a polymerase chain reaction (PCR) assay. The expression of PBP2a was determined with a commercial latex agglutination assay. Species of staphylococcal isolates were identified by use of morphologic, biochemical, and enzymatic tests. RESULTS: Most of the isolated staphylococci were methicillin-susceptible, coagulase-positive Staphylococcus intermedius isolates. Whereas only 2 of 57 S. intermedius isolates were resistant to methicillin, approximately half of the isolates had the mecA gene and produced PBP2a. Staphylococcus schleiferi was the second most common isolate. Widespread resistance to methicillin was found among S. schleiferi isolates. More coagulase-negative S. schleiferi isolates were identified with mecA gene-mediated resistance to methicillin, compared with coagulase-positive S. schleiferi isolates. CONCLUSIONS AND CLINICAL RELEVANCE: The latex agglutination assay for the detection of PBP2a expression coupled with the PCR assay for the mecA gene may provide new information about emerging antimicrobial resistance among staphylococcal isolates.  相似文献

Sea turtles interact with a variety of fishing gears across their broad geographic distributions and ontogenetic habitat shifts. Cumulative assessments of multi-gear bycatch impacts on sea turtle populations are critical for coherent fisheries bycatch management, but such estimates are difficult to achieve, due to low fisheries observer effort, and a single-species, single-fishery management focus. We compiled the first cumulative estimates of sea turtle bycatch across fisheries of the United States between 1990 and 2007, before and after implementation of fisheries-specific bycatch mitigation measures. An annual mean of 346,500 turtle interactions was estimated to result in 71,000 annual deaths prior to establishment of bycatch mitigation measures in US fisheries. Current bycatch estimates (since implementation of mitigation measures) are ∼60% lower (137,800 interactions) and mortality estimates are ∼94% lower (4600 deaths) than pre-regulation estimates. The Southeast/Gulf of Mexico Shrimp Trawl fishery accounts for the overwhelming majority of sea turtle bycatch (up to 98%) in US fisheries, but estimates of bycatch in this fishery are fraught with high uncertainty due to lack of observer coverage. Our estimates represent minimum annual interactions and mortality because our methods were conservative and we could not analyze unobserved fisheries potentially interacting with sea turtles. Although considerable progress has been made in reducing sea turtle bycatch in US fisheries, management still needs improvement. We suggest that sea turtle bycatch limits be set across US fisheries, using an approach similar to the Potential Biological Removal algorithm mandated by the Marine Mammal Protection Act.  相似文献

Yeast bread is a major contributor of sodium in the American diet. Because of its functional impact on dough rheology and the quality of the final baked product, simply reducing the level of sodium chloride (salt) in the formula or replacing it with salt substitutes has found minimal success. The objective of this study was to determine the effect of sea salt containing 57 or 64% less sodium than common sea salt on the breadmaking properties and consumer acceptability of bread. The sodium content of the salt had no effect on dough strength, mixing time, gas production, loaf volume, or crumb grain. The flavor and overall liking of breads containing sea salt with 57 and 64% less sodium content were scored only slightly lower than bread containing the control salt by an untrained panel of 118 consumers. No difference in texture and no unacceptable flavor notes in the bread made with reduced‐sodium salts were reported. Thus, it appears that use of reduced‐sodium sea salt is a satisfactory alternative to reduce the sodium content of bread.  相似文献

While food abundance is known to limit bird populations, few studies have simultaneously examined both temporal and habitat-related variation in food abundance for ground-foraging birds. We surveyed ground-dwelling arthropods throughout the breeding season and in three forest harvest types (clearcut, selection, and shelterwood stands managed for spruce, fir and hemlock) at the Penobscot Experimental Forest (PEF), Bradley, Maine, USA. Pitfall trapping during 2003 and 2004 was used to measure seasonal and habitat-related variation in abundance of ground-dwelling arthropod food items for hermit thrushes. Arthropods were sorted into relevant taxonomic and size class categories based on the known diet of hermit thrushes. Habitat variables included forest management type, stand-level structure and composition of tree species, and microhabitat measures in a 1 m2 area around the pitfall traps. We used redundancy analysis to find stand-level and microhabitat measures that best explained variation in arthropod abundance during 2004. The majority of arthropod captures comprised three groups: ants (Formicidae, 44%), ground beetles (Carabidae, 13%), and spiders (Araneae, 11%). There was significant temporal variation in captures for many taxa. Notably, there were as many as six times more Araneae in early compared to late season samples suggesting the importance of Araneae to ground-foraging birds. Carabidae were more abundant in selection versus either shelterwood or clearcut stands on one sample date. Conversely, Formicidae and Curculionidae were more abundant in shelterwood plots on one and three sampling dates, respectively. Overall, our data provided little evidence that forest management type impacts arthropod food abundance, in stands 5–20 years post-harvest. Abundance of Carabidae was positively correlated with the number of softwoods less than 5 cm DBH and the number of hardwoods greater than 10 cm DBH, while abundance of Formicidae and Homopterans was negatively associated with these same measures. Several taxa were negatively associated with percent of the 1 m2 around the pitfall that was shaded, including Araneae, Carabidae, Formicidae, and Staphylinidae. These results suggest that less-shaded forest microhabitats are important areas of increased food abundance for ground-foraging birds, regardless of forest management type or tree composition. Consequently, forest managers seeking to maximize food abundance for ground-foraging birds in managed spruce-fir forests should consider retaining some large diameter hardwood trees and use management techniques that reduce forest floor shading.  相似文献

Controlled chemical modifications of single-walled carbon nanotubes (SWCNTs) that tune their useful properties have been sought for multiple applications. We found that beneficial optical changes in SWCNTs resulted from introducing low concentrations of oxygen atoms. Stable covalently oxygen-doped nanotubes were prepared by exposure to ozone and then light. Treated samples showed distinct, structure-specific near-infrared fluorescence at wavelengths 10 to 15% longer than displayed by pristine semiconducting SWCNTs. Dopant sites harvest light energy absorbed in undoped nanotube regions by trapping mobile excitons. The oxygen-doped SWCNTs are much easier to detect and image than pristine SWCNTs because they give stronger near-infrared emission and do not absorb at the shifted emission wavelength.  相似文献

Molecular diagnostic techniques have been developed to differentiate the Ascochyta pathogens that infect cool season food and feed legumes, as well as to improve the sensitivity of detecting latent infection in plant tissues. A seed sampling technique was developed to detect a 1% level of infection by Ascochyta rabiei in commercial chickpea seed. The Ascochyta pathogens were shown to be genetically diverse in countries where the pathogen and host have coexisted for a long time. However, where the pathogen was recently introduced, such as A. rabiei to Australia, the level of diversity remained relatively low, even as the pathogen spread to all chickpea-growing areas. Pathogenic variability of A. rabiei and Ascochyta pinodes pathogens in chickpea and field pea respectively, appears to be quantitative, where measures of disease severity were based on aggressiveness (quantitative level of infection) rather than on true qualitative virulence. In contrast, qualitative differences in pathogenicity in lentil and faba bean genotypes indicated the existence of pathotypes of Ascochyta lentis and Ascochyta fabae. Therefore, reports of pathotype discrimination based on quantitative differences in pathogenicity in a set of specific genotypes is questionable for several of the ascochyta-legume pathosystems such as A. rabiei and A. pinodes. This is not surprising since host resistance to these pathogens has been reported to be mainly quantitative, making it difficult for the pathogen to overcome specific resistance genes and form pathotypes. For robust pathogenicity assessment, there needs to be consistency in selection of differential host genotypes, screening conditions and disease evaluation techniques for each of the Ascochyta sp. in legume-growing countries throughout the world. Nevertheless, knowledge of pathotype diversity and aggressiveness within populations is important in the selection of resistant genotypes.  相似文献
