The rasH2 mouse was developed as a model for carcinogenicity studies in regulatory science. Its phenotype is stable during high-volume production and over successive generations. To produce rasH2 mice, three strains of mice (C57BL/6J-TgrasH2, C57BL/6J, and BALB/cByJ) were maintained individually. Since the homozygous c-HRAS genotype is lethal, hemizygous transgenic mice were maintained by crossing with inbred C57BL/6J mice. After breeding, male B6-transgenic mice were mated with female BALB/cByJ mice to obtain transgenic mice. Pups that were rasH2-Tg (tg/wt) or rasH2-Wt (wt/wt) were confirmed by genotyping. Frozen embryos were preserved by the Central Institute for Experimental Animals (CIEA) and sent to two facilities, CLEA Japan and Taconic Biosciences, where the mice were produced. Production colonies are created in both facilities and supplied to customers worldwide. To prevent genetic drift, the colonies were renewed for up to 10 generations, and renewals were carried out four times every five years from 2005 to 2021. To ensure the uniformity and maintenance of the phenotype of rasH2 mice, the carcinogen susceptibilities were monitored in every renewal of colonies by CIEA based on a standard protocol of the short-term carcinogenicity study using the positive control compound N-methyl-N-nitrosourea (MNU). Furthermore, simple carcinogenicity monitoring targeting the forestomach, the organ most sensitive to MNU, was performed approximately once a year. Based on the optimally designed production and monitoring systems, the quality of rasH2 mice with reproducibility and stability of carcinogenicity is maintained and supplied globally.  相似文献

We investigated the soybean productivity of double cropping and relay intercropping in farmer’s fields for three years using two black soybean cultivars of Kurozukin and Tanbaguro, which are used for the first and second crop, respectively. Kurozukin is the early maturing cultivar for vegetable soybean harvest and Tanbaguro is the late-maturing cultivar for the harvest of vegetable soybean and seeds. The yield of the first crop (Kurozukin) was similar to the mono cropping regardless of cropping patterns. However, the yield of the second crop (Tanbaguro) was affected by cropping patterns. The yield of Tanbaguro in double cropping was prone to decrease by late sowing. The late sowing was induced by the late sowing and late harvesting of Kurozukin because of the low temperature in April and the large amount of precipitation in rainy season, respectively. In relay intercropping that Tanbaguro was sown between the rows of Kurozukin at about one month before the harvest of Kurozukin, the yield of Tanbaguro was similar to the mono cropping and the competition with Kurozukin was not observed. Thus, the land equivalent ratio value of double cropping was lower than that of relay intercropping. These results suggest that relay intercropping is more useful cultivation system than double cropping to increase the annual soybean production.  相似文献

Summary Triticum aestivum cv. Norin 26 with Aegilops crassa, Ae. juvenalis or Ae. vavilovii cytoplasm (all D2 type) has been studied relative to its photoperiodic response of male sterility and fertility restoration patterns. Alloplasmic lines of Norin 26 with a D2 type cytoplasm showed almost complete male sterility under long-day conditions (15 h), but high male fertility under short-day conditions (14.5 h). No significant influence of temperature on reduction in male fertility was observed. Thus, this type of male sterility is called photoperiod-sensitive cytoplasmic male sterility (PCMS). The PCMS is expressed in the form of pistillody of stamens. Histological studies revealed that there were incomplete ovule-like structures instead of tapetal cells and pollen grains in the pistillate stamens. The floret differentiation stage of the plant is the stage that is sensitive to photoperiod. The PCMS can be used as a new means for hybrid wheat production, named two-line system. The PCMS line is maintained and multiplied by self-fertilization under short-day conditions, and hybrid seed can be produced by crossing the PCMS line with a pollinator line under long-day conditions. Seven kinds of angiotensin I-converting enzyme (ACE) inhibitory peptides were isolated from the hydrolysates of wakame (Undaria pinnatifida) by Protease S "Amano" (from Bacillus stearothermophilus) by using three-step high-performance liquid chromatography (HPLC) on a reverse-phase column. These peptides were identified by amino acid composition analysis, sequence analysis, and liquid chromatography-mass spectrometry (LC-MS), as Val-Tyr (IC(50) = 35.2 microM), Ile-Tyr (6.1 microM), Ala-Trp (18.8 microM), Phe-Tyr (42.3 microM), Val-Trp (3.3 microM), Ile-Trp (1.5 microM), and Leu-Trp (23.6 microM). These peptides have resistance against gastrointestinal proteases in vitro. Each peptide was determined to have an antihypertensive effect after a single oral administration in spontaneously hypertensive rats (SHR). Among them, the blood pressure significantly decreased by Val-Tyr, Ile-Tyr, Phe-Tyr, and Ile-Trp in a dose of 1 mg/kg of body weight (BW). The present study showed that antihypertensive effect in the hydrolysates of wakame by Protease S "Amano" was attributed to these peptides.  相似文献

Recent reports have shown that dietary phosphatidylcholine (PC) has various beneficial biological effects. Omega 3 polyunsaturated fatty acids (omega3 PUFAs) have also been reported to have lipid-lowering effects in animal models and human studies. In the present study, we investigated the effect of omega3 PUFAs containing PC (omega3-PC) on obesity-related disorders in Otsuka Long-Evans Tokushima fatty (OLETF) rats. Rats were fed semisynthetic diets that contained either 5% corn oil and 2% egg-PC or 5% corn oil and 2% omega3-PC for 4 weeks. During this 4 week feeding of the omega3-PC, the OLEFT rats showed a decrease of omental white adipose tissue weight. In addition, the omega3-PC diet significantly decreased liver weight and hepatic lipid levels in OLETF rats. These changes were attributable to the significant suppression of fatty acid synthase activity and significant enhancement in the activities of carnitine palmitoyltransferase and peroxisomal beta-oxidation. Moreover, the omega3-PC diet reduced serum glucose levels concomitant with the increase of serum adiponectin levels. These results show that compared with egg-PC, omega3-PC can prevent or alleviate obesity-related disorders through the suppression of fatty acid synthesis, enhancement of fatty acid beta-oxidation, and increase of the serum adiponectin level in OLETF rats.  相似文献

Objective We aimed to examine the effects of cannabidiol (CBD)-containing hemp oil without delta-9-tetrahydrocannabinol (THC) as a supplemental treatment for canine atopic dermatitis (CAD), as well as its adverse effects, and effects on concurrent drug use in dogs.Animal In this retrospective case series, 8 dogs with CAD were diagnosed by veterinary dermatologists certified by the Japanese Society of Veterinary Dermatology.Procedure The medical records of dogs supplemented with CBD-containing hemp oil were evaluated with respect to signalment, physical examination, plasma C-reactive protein concentrations, pharmacologic management, the CAD Extent and Severity Index (4th iteration), and the Pruritus Visual Analog Scale.Results Overall, CBD, used as a supplement in combination with other drugs, was well-tolerated over a wide dose range and decreased the occurrence of pruritus in dogs with CAD when ingested twice a day.Conclusion This study provides the first report of supplementation with CBD without THC that was effective in controlling pruritic behavior in dogs with CAD.Clinical relevance Further controlled studies are required to investigate the dose range, efficacy, and safety.  相似文献

The enantiomers of 6-fluoro-, 6-bromo-, and 6-iodopericosine A were synthesized. An efficient synthesis of both enantiomers of pericoxide via 6-bromopericosine A was also developed. These 6-halo-substituted pericosine A derivatives were evaluated in terms of their antitumor activity against three types of tumor cells (p388, L1210, and HL-60) and glycosidase inhibitory activity. The bromo- and iodo-congeners exhibited moderate antitumor activity similar to pericosine A against the three types of tumor cell lines studied. The fluorinated compound was less active than the others, including pericosine A. In the antitumor assay, no significant difference in potency between the enantiomers was observed for any of the halogenated compounds. Meanwhile, the (−)-6-fluoro- and (−)-6-bromo-congeners inhibited α-glucosidase to a greater extent than those of their corresponding (+)-enantiomers, whereas (+)-iodopericosine A showed increased activity when compared to its (−)-enantiomer.  相似文献

The clay mineralogy of 22 samples of the Ap horizons of Ando soils was determined by a combination of methods. Of these samples, 15 did and 7 did not contain allophane and imogolite. Opaline silica was found in 4 samples, whereas aluminum—humus complexes, iron oxides and layer silicates were found in all samples. The presence of allophane and imogolite and the absence of opaline silica in a few Ap horizons was related to mixing of A1 horizons and subsoils by cultivation and to lower supplies of organic matter relative to the amounts of aluminum released from volcanic ash by weathering. The contents of 2:1 and 2:1:1 layer silicates and their intergrades were larger in soils in which quartz predominated in fine fractions. It was inferred that aluminum bound with humus and in allophane-like constituents, rather than aluminum in allophane and imogolite, is important in reactions with phosphate and fluoride.  相似文献

This study evaluated four commercially available biotechnologically processed soybean meals (SBMs) that were included as the primary protein sources in a nonfishmeal diet for juvenile rainbow trout Oncorhynchus mykiss. The processed SBMs were Afcep SBM fermented with mixed bacteria predominantly Bacillus spp., DaBomb SBM fermented with Lactobacillus acidophilus, PepSoyGen SBM fermented with Aspergillus oryzae and Bacillus subtilis, and HP300 SBM treated enzymatically. Fishmeal (FM) and common heat-treated and hulled SBM were used in the reference diets. Sodium dodecyl sulfate (SDS) polyacrylamide gel electrophoresis (PAGE) analysis revealed that the protein components of Afcep SBM appeared to have relatively small molecular sizes compared with the other SBM products. After a 10-week feeding trial, growth of fish fed the common SBM diet was depressed, and abnormal liver and distal intestinal morphologies and bile acid characteristics were observed. The Afcep SBM diet showed approximated growth performance and comparable physiological conditions relative to the FM diet, while only limited improvements were observed in fish fed the other processed SBMs. These findings suggest that Afcep SBM, with its smaller molecular mass proteins, is the most promising processed SBM in nonfishmeal diets for rainbow trout.  相似文献
