Lignan production in callus and cell suspension cultures ofDaphne odora is reported for the first time. The cell suspension culture produced pinoresinol, lariciresinol, secoisolariciresinol, matairesinol, and wikstromol. The eating quality of cooked rice is important and determines its market price and consumer acceptance. To comprehensively describe the variation of eating quality in 183 rice germplasm accessions, we evaluated 33 eating-quality traits including amylose and protein contents, pasting properties of rice flour, and texture of cooked rice grains. All eating-quality traits varied widely in the germplasm accessions. Principal-components analysis (PCA) revealed that allelic differences in the Wx gene explained the largest proportion of phenotypic variation of the eating-quality traits. In 146 accessions of non-glutinous temperate japonica rice, PCA revealed that protein content and surface texture of the cooked rice grains significantly explained phenotypic variations of the eating-quality traits. An allelic difference based on simple sequence repeats, which was located near a quantitative trait locus (QTL) on the short arm of chromosome 3, was associated with differences in the eating quality of non-glutinous temperate japonica rice. These results suggest that eating quality is controlled by genetic factors, including the Wx gene and the QTL on chromosome 3, in Japanese rice accessions. These genetic factors have been consciously selected for eating quality during rice breeding programs in Japan.  相似文献

Seedling ofWisteria floribunda, which belongs to the twiner type of climbing plants, were planted in nursery and the effect of support on the growth of the seedlings was tested. The seedlings were grown with 240 cm-height and 120 cm-height supports (the culms of a dwarf bamboo), and without support. Positive effect of the supports and its length were observed for stem length ofW. floribunda at the end of the growing season. The positive effect of support was also observed for total dry weight. Total dry weight with 240 cm-height support reached more than the double of the seedlings without support. These effects of the support were attributed to die-back behavior of the twining stem, which detached from the support or did not encounter the support. Diameter of the stems without support showed a rapid decrease toward the stem tip, while that with support were relatively constant when they were twining around the support. The existence of the support also affected the dry matter allocation in this species; supported seedlings allocated larger proportion of biomass to current-year stems than unsupported seedlings, although the allocation to the roots was smaller in the supported treatments. Support availability may be critical for the establishment and further growth of the seedlings ofW. floribunda which regenerate at the forest edge and canopy gaps. Stereochemistry and biosynthesis of guaiacylglycerol-8-O-4′-(sinapyl alcohol) ether (GGSE), an 8-O-4′ neolignan, which consists of coniferyl and sinapyl alcohol moieties, in Eucommia ulmoides were investigated. Four 8-O-4′ neolignans, GGSE, syringylglycerol-8-O-4′-(coniferyl alcohol) ether (SGCE), guaiacylglycerol-8-O-4′-(coniferyl alcohol) ether (GGCE), and syringylglycerol-8-O-4′-(sinapyl alcohol) ether (SGSE), were synthesized. Their erythro and threo diastereomers were separated through acetonide derivatives, intermediates of the synthesis, and identified by means of nuclear magnetic resonance (NMR) spectroscopy. All of the erythro-acetonide derivatives have larger coupling constants (ca 9 Hz) for the C7-H resonances than those of the threo ones (1.5–2 Hz). In the case of the four 8-O-4′ neolignans, the C7-H coupling constants of the threo-isomers are not smaller than those of the erythro ones. GGSE isolated previously from this plant was identified as the erythro isomer by comparison of the 13C-NMR data with synthetic erythro-GGSE and threo-GGSE and the other 8-O-4′ neolignans mentioned as above. Administration of a mixture of [8-14C]coniferyl alcohol and [8-14C]sinapyl alcohol to excised shoots of E. ulmoides was carried out and the incorporation of 14C into erythro-[14C]GGSE was found to be higher than that in threo-[14C]GGSE. We previously reported that the release of O2 from isolated pea cell walls was enhanced by a 70-kDa glycoprotein elicitor but was suppressed by mucin-type glycopeptide suppressors (supprescins A and B) prepared from pycnospore germination fluid of Mycosphaerella pinodes, causal agent of Mycosphaerella blight of pea. Here, we show that superoxide dismutase (SOD) in the apoplast fluid/cell wall of pea seedlings responds to the fungal elicitor and suppressor molecules. In a pharmacological study and with internal amino acid sequencing, the apoplastic SOD in a pea cultivar Midoriusui was found to be a Cu/Zn type SOD. We cloned a full-length cDNA of the Cu/Zn-SOD and designated it as PsCu/Zn-SOD1. An increase in PsCu/Zn-SOD1 mRNA and the PsCu/Zn-SOD1 protein was induced by treatment with the elicitor more intensively than by wounding. Such induction by the elicitor or wounding, however, was inhibited by the concomitant presence of supprescins. The SOD activity of recombinant PsCu/Zn-SOD1 was regulated directly by these signal molecules in a manner similar to their effect on the SOD activity in the apoplastic fluid and in the cell wall-bound proteins. Based on these findings, we discuss a role for PsCu/Zn-SOD1 in the pea defense response. Pythium species were isolated from seedlings of strawberry with root and crown rot. The isolates were identified as P. helicoides on the basis of morphological characteristics and sequences of the ribosomal DNA internal transcribed spacer regions. In pathogenicity tests, the isolates caused root and crown rot similar to the original disease symptoms. Multiplex PCR was used to survey pathogen occurrence in strawberry production areas of Japan. Pythium helicoides was detected in 11 of 82 fields. The pathogen is distributed over six prefectures.  相似文献

Potato dextrose broth (PDB), one of the most popular culture media for fungi, can be made in the laboratory from potato extract and glucose (handmade PDB) and also bought as powder (commercial PDB). We compared growth of Fusarium oxysporum in handmade and commercial PDBs. For seven strains tested, bud cell formation and fungal biomass were higher in handmade PDB than in commercial PDB. Gel filtration analyses revealed that handmade PDB contained carbohydrates with molecular weights >20 MDa, while most carbohydrates in commercial PDB were <45 kDa. Freshly prepared, raw potato starch had a molecular weight >20 MDa and enhanced bud cell formation by five of seven strains when added to commercial PDB. The addition of glucose to commercial PDB promoted biomass production but not bud cell formation. Furthermore, in a semisynthetic medium containing raw potato starch, bud cell formation was enhanced for all strains tested. These results indicate that the high molecular carbohydrates present in raw potatoes enhanced bud cell formation by Fusarium oxysporum.  相似文献
