Background: The CBC is an essential test for assessing the health of rats used in drug development studies. Because of limited blood volume, estimates of cell counts from a blood smear would be valuable when other analytical methods of enumerating cells are not possible or available. Objective: The purpose of this study was to develop a statistical model to accurately estimate WBC, platelet (PLT), and RBC counts in blood smears from rats. Method: Blood smears and quantitative cell counts were obtained from vehicle‐treated male and female Fischer 344 rats (n=65) involved in a variety of studies. The numbers of WBCs, PLTs, and RBCs were estimated in 10 fields in the monolayer of smears using × 20 (WBC) or × 100 (PLT, RBC) objectives. Using a statistical model and the quantitative cell counts obtained on an ADVIA 120 hematology analyzer, formulas were developed to predict the quantitative counts from the estimates. Results: Data were log‐transformed before analysis. A formula was derived using the slope and intercept of the regression line between cell estimates and ADVIA counts to predict WBC, PLT, and RBC counts based only on estimates. A second formula was developed for situations in which limited quantitative analyses may be available, and resulted in even more accurately predicted counts from smear estimates. Conclusion: The formulas developed in this study can be a valuable tool in estimating cell counts from a blood smear when cell counting instruments are not available or when an instrument cell count needs to be verified. These formulas may be useful in the assessment of rat blood in discovery and lead optimization studies.  相似文献

We examined the effects of fertilization and gypsy moth defoliation on condensed tannin concentration (%CT) of hybrid poplar (Populus x canadensis cv 'Eugeneii') fine roots in the summers of 1997 and 1998. This factorial experiment included two defoliation treatments (defoliated and a foliated control) and fertilization treatments (100 kg nitrogen (N) ha(-1) and an unfertilized control). Gypsy moth (Lymantria dispar L.) populations were experimentally increased to obtain defoliation in the summers of 1996, 1997 and 1998; fertilization subplots were supplemented with NH4NO3 (100 kg N ha(-1)) in the spring of each year. Despite the severity of defoliation, the effects were small, and significant on only two sampling dates: in May 1997, when fine root %CT was 23% lower in the defoliated trees, and in November 1997, when trees in the defoliated unfertilized plots had 35% higher root %CT than trees in all other plots. Defoliation effects on root %CT did not follow the same seasonal pattern as defoliation effects on root starch content, N uptake capacity or leaf %CT. Regulation of root condensed tannin concentration appeared to be partially uncoupled from these traits. The small transient effects on root defense reflect the resilience of this early successional tree to severe early season defoliation.  相似文献

Continuing our search for natural product and natural product-based compounds for the control of off-flavor in catfish, 29 stilbene analogues were synthesized and evaluated for algicidal activity against the 2-methylisoborneol (MIB)-producing cyanobacterium Oscillatoria perornata. The cis and trans isomers of 4-(3,5-dimethoxystyryl)aniline showed moderate and selective algicidal activity toward O. perornata with the lowest observed inhibitory concentration and lowest complete inhibition concentrations of 10 muM. This is the first report on selective stilbene algicidal activity toward a MIB-producing cyanobacteria species.  相似文献

Protected areas have a central role to play in educating people about biodiversity and nature conservation. However, there is little evidence regarding how effective public education efforts related to protected areas have been. We used on-site questionnaires at Sites of Special Scientific Interest to determine whether visitors to sites were aware of their protected status. Less than one-third of visitors knew the site they were visiting was a SSSI. However, there was substantial variation in understanding across sites (8-43%). When we compared this variation to five possible predictor variables, only membership of conservation organisations was retained in all models in the 95% confidence set, with members of these groups being better informed about protected area designations. However, the predictive power of all models was low. These results represent a challenge for those aiming to reengage people with conservation and the countryside, and to educate them about its value and contribution.  相似文献

Afforestation and deforestation are key land-use changes across the world, and are considered to be dominant factors controlling ecosystem functioning and biodiversity. However, the responses of soil microbial communities to these land-use changes are not well understood. Because changes in soil microbial abundance and community structure have consequences for nutrient cycling, C-sequestration and long-term sustainability, we investigated impacts of land-use change, age of stand and soil physico-chemical properties on fungal and bacterial communities and their metabolic activities. This study was carried out at four sites in two geographical locations that were afforested on long-established pastures with Pinus radiata D. Don (pine). Two of the sites were on volcanic soils and two on non-volcanic soils and stand age ranged from 5 to 20 y. Microbial communities were analysed by biochemical (phospho-lipid fatty acids; PLFA) and molecular (multiplex-terminal restriction fragment length polymorphism; M-TRFLP) approaches. Both site and stand age influenced microbial properties, with changes being least detectable in the 5-y-old stand. Land use was a key factor influencing soil metabolic activities as measured by physiological profiling using MicroResp. Pasture soils had higher microbial biomass (P < 0.001), and metabolic activities (P < 0.001), and basal respiration rates were up to 2.8-times higher than in the pine soils. Microbial abundance analysis by PLFA showed that the fungal to bacterial ratio was higher in the pine soils (P < 0.01). Community analysis suggested that soil bacterial communities were more responsive to site (principal component 1; P < 0.001) than to land use (principal component 5; P < 0.001). In contrast, the fungal community was more affected by land-use change (principal component 1; P < 0.001) than by site, although site still had some influence on fungal community structure (principal component 2; P < 0.001). Redundancy analysis also suggested that bacterial and fungal communities responded differently to various soil abiotic properties, land-use change and location of sites. Overall, our results indicate that the change in land use from pasture to P. radiata stands had a direct impact on soil fungal communities but an indirect effect, through its effects on soil abiotic properties, on bacterial communities. Most of the changes in bacterial communities could be explained by altered soil physico-chemical properties associated with afforestation of pastures.  相似文献

Animal health is a key factor affecting the economic efficiency of the dairy industry. Improvements in animal health are also of relevance to society more broadly, given important implications for animal welfare, food safety and quality. Although the economic gains of best practice with regard to animal health have been well documented, many farmers are not adopting optimal herd management techniques. This paper utilises nationally representative farm-level data from Ireland for 2013 to identify drivers and barriers to the adoption of best practice with regard to on-farm mastitis management. Exploratory factor analysis is used to derive measures of farmers’ attitudes towards animal health and mastitis and econometric techniques are employed to empirically assess the influence of these on the uptake of beneficial herd health management practices. A number of focus groups were also undertaken to complement the analysis. This paper concludes that farmers’ attitudes towards animal health are not a key driver in the uptake of best practice, although perceived disease risk is of relevance. A number of interesting issues arise in identifying barriers to the uptake of best practice, these include the possibility of routine inertia, i.e., farmers do not deviate from the routine developed around mastitis prevention until there is an indication of infection, as well as constraints around the availability of labour and time. Farmer behaviour with respect to mastitis management can thus be considered as reactionary as opposed to precautionary. This research highlights the valuable role of the extension agent but concludes that engagement around knowledge transfer and technology adoption is particularly complex.  相似文献

A combination of statistical techniques of analyses were used to evaluate the potential of International Commission on Illumination (CIE) lightness (L*), redness (a*) and yellowness (b*) colour space system and near-infrared spectroscopy (NIRS) to assess surface changes in relation with progressive decay of beech (Fagus grandifolia Ehrh.) by wood-inhabiting lignicolous fungi Inonotus hispidus, Trametes versicolor and Xylaria polymorpha. pH effects based modelling predictions of beech earlywood and latewood tissues were also included. Multivariate analysis techniques included response surface optimization, sample-specific standard error of prediction (SEP) method and projection to latent structures partial least squares (PLS) regression. Strong statistical relationships were derived for pH predictions with R2 values ranged: from 0.77 to 0.84 for I. hispidus; from 0.77 to 0.84 for T. versicolor and from 0.83 to 0.91 for X. polymorpha. R2 values for CIE-based L*a*b* colour space measurements ranged: from 0.43 to 0.69 (L*), 0.66 to 0.76 (a*), 0.42 to 0.53 (b*) for I. hispidus; from 0.59 to 0.69 (L*), 0.69 to 0.79 (a*), 0.64 to 0.79 (b*) for T. versicolor; and from 0.51 to 0.75 (L*), 0.89 to 0.94 (a*), 0.85 to 0.89 (b*) for X. polymorpha. Multivariate technical analysis (response surface analysis, sample-specific SEP, PLS regression) of CIE L*a*b* system and NIRS results should be able to characterize pH effects and surface changes of wood spalted by lignicolous fungi as a quick and reliable non-destructive method relevant to wood-spalting concerns and the forest products industry.  相似文献
