Abstract Abstract: A 9‐year‐old Arabian mare was evaluated for a 7‐day history of malaise. Results of a CBC included a leukocyte concentration within the reference interval (8.4 × 103/μL, reference interval 6.0–14.0 × 103/μL) with an apparent degenerative left shift (segmented neutrophils 1.2 × 103/μL, reference interval 2.5–7.5 × 103/μL; hyposegmented neutrophils 1.8 × 103/μL, reference interval 0.0–0.2 × 103/μL). Serum clinical chemistry results included increased aspartate transaminase, alkaline phosphatase, and gamma‐glutamyltransferase activities. A presumptive diagnosis of hepatitis or cholangiohepatitis was made. The horse was treated with antimicrobials and the malaise quickly resolved. However, in a recheck CBC on day 13, the apparent degenerative left shift remained. Further evaluation of the blood smear revealed many hyposegmented granulocytes with coarse mature chromatin and normal cytoplasmic features. On the basis of the microscopic examination, the horse was diagnosed with Pelger‐Huët anomaly. The patient's offspring was subsequently also diagnosed with Pelger‐Huët anomaly on the basis of blood film examination. Neutrophil, eosinophil, and basophil mean nuclear scores in both affected horses (mare, range 1.5–2.6; offspring, range 1.6–3.2) were lower than those in 2 unrelated Arabian horses (range, 2.8–5.0) and 5 non‐Arabian control horses (range, 2.8–5.0). Results of immunophenotyping and phagocytosis/oxidative burst assays via flow cytometry showed no difference in the expression of myeloid–specific or adhesion molecules or in neutrophil function between affected and control horses. This is the second known report of equine Pelger‐Huët anomaly, both of which affected Arabian horses.  相似文献

Marsupial young are born in an under-developed state without mature immune responses. Prior to the maturation of an immune system, marsupial young are heavily reliant upon immune factors secreted in the milk to defend them against potential microbial pathogens in the environment. In this study, we identified and characterized the immunoglobulin heavy chain constant regions, light chains, polymeric Ig receptor (pIgR), J chain, neonatal Fc receptor (alpha chain) (FcRn) and the chemokine CCL28 from the model marsupial species, the tammar wallaby (Macropus eugenii). Low levels of conservation were seen in motifs in C and Cγ associated with receptor binding and or transcytosis, and this may have potential implications for functionality. We evaluated the expression of immunoglobulin genes in the tammar mammary gland throughout lactation and found that two periods of increased expression of immunoglobulin genes occur. These two periods coincide with the birth of the young, and with its first emergence from the pouch. This increased expression may represent a strategy for maternal immunological protection of the pouch young.  相似文献

Evidence is presented that corn (maize) and sorghum starch granules have channels that connect a central cavity to the external environment. A mutant sorghum starch with numerous, large surface pores was treated with a methanolic solution of merbromin and sectioned. Light, fluorescence, and compositional backscattering electron microscopy revealed channels connecting an internal cavity to the external surface in most granules. Cavities and channels could also be seen in whole corn and sorghum starch granules treated with merbromin in methanol and viewed by fluorescence microscopy. Treatment of sorghum starch granules with an aqueous solution of merbromin revealed that the molecule penetrated the granule matrix under even slightly swelling conditions. Light microscopy showed cavities in unstained, whole, dry corn and sorghum starch granules mounted in immersion oil.  相似文献

Determining the optimum light conditions for sea urchins reared in land‐based systems is vital for the future use and assessment of possible commercial systems of sea urchin farming. The effects of two different light regimes, complete darkness and a long day photoperiod of 16 h light:8 h darkness, on the somatic and gonadal growth of the European sea urchin Paracentrotus lividus (19.5–23.0 mm) was investigated using the commercial UrchinPlatter? System over a 6‐month period (5 March to 5 September). Hatchery‐produced P. lividus were transported to the Aquaculture Fisheries Development Centre (AFDC, University College, Cork UCC). Before arrival at the AFDC, sea urchins were reared on a diet of Laminaria digitata. Females were the predominant species of the animal group, displaying a reproductive Stage III (growing stage) where gametogenesis was commencing. Results show that darkness supports higher somatic growth than the photoperiod treatment. Feeding rates were higher for sea urchins reared under darkness with gonadal growth increasing for both experimental treatments. Individuals reared under darkness had a higher per cent change in gonad index from the initial sample taken at the beginning of the experiment.  相似文献

Adventitious shoot regeneration and protoplast isolation and culture were examined from leaf explants of in vitro shoot cultures of several cauliflower (Brassica oleracea var. botrytis) cultivars, sourced from Europe and Australia, was investigated with the aim to develop improved nuclear and plastid transformation protocols for this vegetable crop. Eight out of 10 cultivars regenerated shoots from at least 79% of leaf explants. Mesophyll protoplasts from leaves gave high yields and division frequencies. Growth of shoot cultures in large glass vessels with vented lids was the key factor in obtaining high protoplast division frequencies of up to 71% and at least 70% of protoplast calluses regenerating shoots.  相似文献

Context

Biodiversity in arid regions is usually concentrated around limited water resources, so natural resource managers have constructed artificial water catchments in many areas to supplement natural waters. Because invasive species may also use these waters, dispersing into previously inaccessible areas, the costs and benefits of artificial waters must be gauged and potential invasion- and climate change-management strategies assayed.

Objectives

We present a network analysis framework to identify waters that likely contribute to the spread of invasive species.

Methods

Using the Sonoran Desert waters network and the American bullfrog (Lithobates catesbeianus)—a known predator, competitor, and carrier of pathogens deadly to other amphibians—as an example, we quantified the structural connectivity of the network to predict regional invasion potential under current and two future scenarios (climate change and management reduction) to identify waters to manage and monitor for invasive species.

Results

We identified important and vulnerable waters based on connectivity metrics under scenarios representing current conditions, projected climate-limited conditions, and conditions based on removal of artificial waters. We identified 122,607 km2 of land that could be used as a buffer against invasion and 67,745 km2 of land that could be augmented by artificial water placement without facilitating invasive species spread.

Conclusions

Structural connectivity metrics can be used to evaluate alternative management strategies for invasive species and climate mitigation.

Although smoke exposure has been associated with the development of smoke taint in grapes and subsequently in wine, to date there have been no studies that have demonstrated a direct link. In this study, postharvest smoke exposure of grapes was utilized to demonstrate that smoke significantly influences the chemical composition and sensory characteristics of wine and causes an apparent 'smoke taint'. Verdelho grapes were exposed to straw-derived smoke for 1 h and then fermented according to two different winemaking treatments. Control wines were made by fermenting unsmoked grapes. Sensory studies established a perceivable difference between smoked and unsmoked wines; smoked wines were described as exhibiting 'smoky', 'dirty', 'earthy', 'burnt' and 'smoked meat' characteristics. Quantitative analysis, by means of gas chromatography-mass spectrometry, identified guaiacol, 4-methylguaiacol, 4-ethylguaiacol, 4-ethylphenol, eugenol, and furfural in each of the wines made from smoked grapes. However, these compounds were not detected in the unsmoked wines, and their origin is therefore attributed to the application of smoke. Increased ethanol concentrations and browning were also observed in wines made from grapes exposed to smoke.  相似文献
