A 19-month-old Standardbred colt was donated to the University of Pennsylvania School of Veterinary Medicine with a suspicion of intersexuality. The anal−genital distance and penis were normal, and there was no evidence of intersexuality, but the colt was bilaterally cryptorchid. Several aspects of the colt's behavior appeared unusual, including general temperament and behavior described as sympathetically dull and affable. With herd mates, the colt appeared slow to perceive or to learn the usual intraspecies social cues. An atypical gait characterized by intermittent unnatural shuffle of the hind limbs, sliding them along in short rhythmic strides for 3 to 10 seconds at a time was noted at times when a horse might normally transition from a slow walk to a fast walk or a slow trot. Occasionally the colt exhibited slight protrusion of the tongue through the teeth and lips with jaw movements and smacking of the tongue against the teeth as if struggling to retract the tongue to the normal position. Evaluation of the karyotype combined with fluorescent in situ hybridization (FISH) revealed an abnormal male karyotype showing trisomy of chromosome 27 (65, XY + 27). The colt was euthanized at 24 months of age, and a necropsy revealed no significant abnormalities. This case of trisomy was not associated with developmental abnormalities described in other rare reports of trisomy in horses; however, some features were strikingly similar to that of humans with trisomy 21. This study examined the sensitivity, specificity, predictive values, and classification accuracy of 3 commonly used screening tests for failure of passive transfer: the sodium sulfite turbidity test, the zinc sulfate turbidity test, and re-fractometry relative to serum immunoglobulin G1, (IgG1) concentrations determined by radial immunodiffusion. Serum samples were obtained from 242 calves ranging from 1 to 8 days of age. Using a serum concentration of 1,000 mg/dL IgG1 to define adequate passive transfer, the zinc sulfate test had a sensitivity of 1.00 and a specificity of 0.52 in the detection of inadequate passive transfer. The endpoint of the test appeared to be higher than desired; calves testing negative had mean serum IgG1 concentration of 955 mg/dL and a large proportion of calves with adequate passive transfer were misclassified as positive for failure of passive transfer. Using the qualitative zinc sulfate test, the percentage of calves correctly classified with regard to passive transfer status was less than that observed with either the sodium sulfite test or refractometry. The sensitivity of the sodium sulfite assay was 0.85 at a 1+ endpoint and 1.00 at a 2 or 3+ endpoint. The specificity of the sodium sulfite assay varied from 0.87 at a 1+ endpoint and 0.56 at a 2+ endpoint. The sensitivity and specificity of refractometry varied from 0.01 to 1.00 depending on the choice of endpoint. Refractometry correctly classified the largest proportion of calves with regard to their passive transfer status at test endpoints of 5.0 and 5.5 g/dL, 83% and 82% respectively. The highest percentages of calves correctly classified occurred with the sodium sulfite test using a 1+ endpoint (86.30%) and refractometry using a 5.0 g/dL endpoint (83.00%). A regression equation was developed that permitted calculation of an optimal endpoint for refractometric determinations of total serum protein concentration. A serum protein concentration of 5.2 g/dL was equivalent to 1,000 mg/dL serum IgG1. Optimal selection of tests for passive transfer status in calves will be governed by the prevalence of failure of passive transfer, test performance, and the anticipated costs of classification errors. 相似文献

This study evaluated the effect of methionine supplementation, predation risk and their interaction on gut histology, whole-body cortisol levels, and intestinal gene expression in zebrafish. A total of 360 one-year-old animals were maintained under two environmental conditions and fed diets containing different methionine sources. Fish were fed either a control diet (CTL, without methionine supplementation), a diet supplemented with dl -methionine (DLM), or a diet supplemented with methionine dipeptide (MM) in the absence (AP) of a predator or in the presence of the predator (PP) for 48 h or 20 days. Predator-induced stress for 20 days resulted in lower body weight. Zebrafish fed methionine-supplemented diets had higher weight gain than control fish. We found no effect of predation stress or methionine supplementation on cortisol level. Predation risk and methionine supplementation showed no interaction effect on dipeptide transporter gene expression. After 48 h of predation pressure, zebrafish had higher mRNA expression of SOD2, CAT and GPX1 in the gut. After 20 days of exposure to the predator, zebrafish fed methionine-supplemented diets had lower expression of GPX1, SOD2 and CAT than those diet CTL. A comparison of animal gains and vegetation trends was made from 2002–2008 between a continuous season-long stocking (SLS) system and a modified intensive–early stocking system (IES) with late-season grazing (IES 1.6× + 1; 1.6 times the number of animals of the SLS system from May 1 to July 15, and 1 times the number of animals of SLS from July 15 to October 1) on shortgrass native rangeland of western Kansas. The continuous season-long stocked system placed animals at a density of 1.37 ha · steer?1 from May through October, or 2.63 animal unit months (AUM) · ha?1, whereas the intensive–early stocked system with late-season grazing (3.33 AUM · ha?1) stocked pastures at 0.85 ha · steer?1 from May through the middle of July, and then stocked pastures at 1.37 ha · steer?1 for the remainder of the grazing season by removing the heaviest animals mid-July each yr. Average daily gains (0.78 vs. 0.70 kg · d?1, P = 0.039) and total animal gain (58 vs. 52 kg, P = 0.042) were different between the continuous season-long stocked and the intensive–early stocked animals during the first half of the grazing season. No difference was found between average daily gain (0.61 vs. 0.62 kg · d?1, P = 0.726) and total animal gain (48 vs. 49 kg, P = 0.711) for the continuous season-long stocked and intensive–early stocked with late-season grazing animals during the last half of the season. Total individual animal gain (106 vs. 101 kg, P = 0.154) and average daily gain (0.70 vs. 0.66 kg · d?1, P = 0.152) was not different between the continuous season-long stocked and the intensive–early stocked system animals that were on pasture the entire grazing season. Total beef gain on a land-area basis (96 vs. 77 kg · ha?1, P = 0.008) was greater for the modified intensive–early stocked system with late-season grazing with greater animal densities. Drastic changes have occurred in Mongolia’s grazing land management over the last two decades, but their effects on rangelands are ambiguous. Temporal trends in Mongolia’s rangeland condition have not been well documented relative to the effects of long-term management changes. This study examined changes in grazing land use and rangeland biomass associated with the transition from the socialist collective to the current management systems in the Tsahiriin tal area of northern Mongolia. Grazing lands in Tsahiriin tal that were formerly managed by the socialist collective are now used by numerous nomadic households with their privately owned herds, although the lands remain publicly owned. Grazing pressure has more than tripled and herd distribution has changed from a few spatially clustered large herds of sheep to numerous smaller herds of multiple species. Landsat image–derived normalized-difference vegetation index estimates suggest that rangeland biomass significantly decreased (P < 0.001) from the collective to the postcollective periods. The observed decrease was significantly correlated with changes in the grazing management system and increased stocking density (P < 0.001), even when potential climate-induced changes were considered. Furthermore, field- and Satellite Pour l’Observation de la Terre imagery–based rangeland assessments in 2007 and 2008 indicate that current rangeland biomass is low. Spatial pattern analyses show that the low biomass is uniform throughout the study site. The observed decrease in rangeland biomass might be further accelerated if current grazing land use continues with no formal rangeland management institution or organized, well-structured efforts by the local herding households. 相似文献

To determine if different pathotypes of the avian polyomavirus (APV) exist and to compare the genomes of APVs originating from different geographic areas, dates, and species of birds, the partial sequences of 18 APVs were determined. New viral sequences were compared with three published APV sequences. Two of the new viruses had identical sequences. Forty point mutations were found at 31 loci. A 27-bp deletion was found in the VP2 and VP3 open reading frames of one virus. A duplication of the putative origin of replication and adjacent enhancer region was previously reported in one APV. Smaller duplications involving the origin in one APV and a second enhancer region in another were discovered. All duplications were in tissue culture-adapted viruses, suggesting they occurred during the isolation process. Excluding duplications and the deletion, maximum variation between viruses was small (11 bp). A maximum parsimony tree was constructed that contained three major branches. The three earliest isolates were on separate branches. The European viruses were confined to branch I, but APVs from the United States were on all three branches. Lovebird, budgerigar, and macaw APVs were also on each of the three branches, suggesting that species-specific pathotypes have not developed. Most nonsynonymous mutations occurred in a small portion of the VP2 and VP3 open reading frames, demonstrating a selection for these mutations. That a glycine at VP2 221 will inhibit virus replication in chicken embryo fibroblasts (CEFs) has been previously reported. In contrast, six of seven of the new APVs isolated in CEFs had a glycine at VP2 221. 相似文献

ObservationsA 22‐month‐old male neutered Coton De Tulear dog was presented for upper gastrointestinal endoscopy under general anesthesia. The anesthetic plan included premedication with intramuscular meperidine (4 mg kg?1) but meperidine was inadvertently administered at ten‐fold this dose. Within 5 minutes, the dog was unresponsive to external stimulation, and by 10 minutes post‐injection developed generalized signs of central nervous system (CNS) excitement. Initial therapy included inspired oxygen supplementation, and single intravenous (IV) doses of diazepam (0.68 mg kg?1) and naloxone (0.03 mg kg?1) to no effect. A second dose of diazepam (0.46 mg kg?1, IV) abolished most of the signs of CNS excitement. General anesthesia was induced and the endoscopy performed. Time to extubation was initially prolonged, but administering naloxone (final dose 0.1 mg kg?1, IV) to effect enabled extubation. After naloxone, the dog became agitated, noise sensitive, and had leg and trunk muscle twitches. Diazepam (0.30 mg kg?1, IV) abolished these signs and the dog became heavily sedated and laterally recumbent. Naloxone administration was continued as a constant rate infusion (0.02 mg kg?1 hour?1, IV) until approximately 280 minutes post‐meperidine injection, at which time the dog suddenly sat up. Occasional twitches of the leg and trunk muscles were observed during the night. The dog was discharged the next day appearing clinically normal.ConclusionsGiven that the CNS excitatory effects of normeperidine are not a μ opioid receptor effect, the use of naloxone should be considered carefully when normeperidine excitotoxicity is suspected. Benzodiazepines may be beneficial in ameliorating clinical signs of normeperidine excitotoxicity. 相似文献
