Perennial ryegrass (Lolium perenne) is a perennial crop used in temperate regions as forage. In L. perenne breeding programs, persistency is an important trait. Poor persistency results in sward degradation and associated yield and nutritive value losses. Breeders assess persistency of accessions using visual scoring in field plots during the 2nd or 3rd growing season. This evaluation system is easy and cheap but is not free from human bias. In this study, the correlation between the scoring done by different breeders was only 0.243. As an alternative we have developed a methodology to assess persistency of L. perenne breeding materials based on vegetation indices (VIs) derived from Unmanned Aerial Vehicle (UAV) imagery. The VIs Excess green (ExG2), Green Leaf Index and Normalized green intensity (GCC) were found to provide consistent results for flights carried out under different light conditions and were validated by ground reference information. The correlation between the VIs and the percentage of ground cover extracted from on-ground imagery was 0.885. To test the implementation of the method we compared the ExG2 value based approach to selection with a visual score based selection methodology as applied by two breeders. The breeding decisions of Breeder A agreed well with decisions based on ExG2 values (74.6%), but those of Breeder B displayed a lower agreement (54.0%). In contrast, agreement between decisions based on different flights was very high (91.6%). The methodology was validated for general applicability. In summary, the results demonstrate that basing persistency selection in L. perenne breeding programs on ExG2 values from UAV imagery is likely to be more objective in comparison to the currently-used visual scoring method.  相似文献

Genome-wide association studies (GWAS) are useful to facilitate crop improvement via enhanced knowledge of marker-trait associations (MTA). A GWAS for grain yield (GY), yield components, and agronomic traits was conducted using a diverse panel of 239 soft red winter wheat (Triticum aestivum) genotypes evaluated across two growing seasons and eight site-years. Analysis of variance showed significant environment, genotype, and genotype-by-environment effects for GY and yield components. Narrow sense heritability of GY (h 2  = 0.48) was moderate compared to other traits including plant height (h 2  = 0.81) and kernel weight (h 2  = 0.77). There were 112 significant MTA (p < 0.0005) detected for eight measured traits using compressed mixed linear models and 5715 single nucleotide polymorphism markers. MTA for GY and agronomic traits coincided with previously reported QTL for winter and spring wheat. Highly significant MTA for GY showed an overall negative allelic effect for the minor allele, indicating selection against these alleles by breeders. Markers associated with multiple traits observed on chromosomes 1A, 2D, 3B, and 4B with positive minor effects serve as potential targets for marker assisted breeding to select for improvement of GY and related traits. Following marker validation, these multi-trait loci have the potential to be utilized for MAS to improve GY and adaptation of soft red winter wheat.  相似文献

The two most widely grown bromegrass species in North America are smooth bromegrass, a hay type grass, and meadow bromegrass, a pasture type grass. Hybrid populations between these two species have been developed through hybridization and recurrent selection. The objective of this study was to determine the genetic relatedness of the hybrid bromegrass population S‐9073M to its parental populations using random amplified polymorphic DNA (RAPD) markers and to determine genetic variation within and between populations. Individuals from each of the three populations were genotyped at 43 polymorphic RAPD loci. One of the RAPD fragments was meadow bromegrass‐specific. Cluster analysis showed three groups representing the two parental populations and the hybrid population. An analysis of molecular variance (AMOVA), showed that the hybrid population had the highest within‐population variation, followed by meadow bromegrass and smooth bromegrass. The interpopulation genetic distance (phi‐statistic =Φst) was highest between meadow and smooth bromegrass and lowest between smooth and hybrid bromegrass. The hybrid population was genetically intermediate to smooth bromegrass and meadow bromegrass, but closer to smooth bromegrass, possibly reflecting the selection criteria used in the development of this hybrid.  相似文献

Nitrate resulting from nitrogen fertilisers used in Agriculture is a widespread contaminant of shallow groundwater and causes adverse effects on human, animal and ecosystem health. In order to evaluate the full extent of groundwater nitrate contamination, and how it might evolve in time, it is essential to understand controls on aquifer assimilative capacity. This level of understanding will also help to better target policies and incentives aimed at controlling the amount of nitrate entering downstream water systems.The potential for nitrate attenuation in groundwater was assessed by examining the concentration and distribution pattern of electron donors such as dissolved organic carbon (DOC), ferrous iron, and redox indicators such as dissolved oxygen (DO) and Eh in 57 monitoring bores on the lower Burdekin coastal floodplain, one of Queensland's and Australia's premier irrigation districts. Nitrate concentrations ranged from 0.1 to 14.4 mg/L NO3-N but were mostly undetectable in bores close to the coast. Groundwater age dates suggest that while there are nitrate ‘hot spots’ in certain areas, some or most of the nitrate is being consumed on its way to the ocean. Low nitrate concentrations were coupled with high ferrous concentrations. The low DO concentrations (<2 mg/L) and high ferrous concentrations found in 55% of the bores indicate that redox conditions are suitable for nitrate attenuation by either denitrification or dissimilatory nitrate reduction to ammonium. The reducing environment may be associated with the high DOC concentrations (up to 82 mg C/L) found in these groundwaters. Furthermore, high levels of ferrous iron found in the Ayr area combined with the wide spread geographical distribution of DOC indicate that these areas have a high potential for sustaining geochemical processes that reduces nitrate levels. The distribution of geochemical indicators also suggests that the shallower depths (<15 m) of the groundwater systems have more potential for nitrate reduction than the deeper depths. The map identifying areas within the lower Burdekin with most potential for denitrification is a valuable first step in helping to understand and manage the fate of nitrate entering the groundwater.  相似文献

Genetic impacts under selective breeding of agricultural crops have been frequently investigated with molecular tools, but inadequate attention has been paid to assess genetic changes under long-term genetic improvement of plant traits. Here we analyzed allelic changes with respect to wheat trait improvement in 78 Canadian hard red spring wheat cultivars released from 1845 to 2004 and screened with 370 mapped SSR markers. The improvements in quality, maturity, yield, disease, stem rust, leaf rust, sawfly resistance, and agronomy were considered. A total of 154 (out of 370) loci with significant allelic changes across 21 chromosomes were detected in the 78 wheat cultivars separated into improved versus non-improved groups for eight traits. The number of significant loci for improving a trait ranged from four for quality to 68 for yield and averaged 35. Many more loci with significant allelic reduction for improving a trait were detected than those with significant allelic increase. Selection for early maturity introduced more alleles, but improving the other traits purged more alleles. Significantly lower numbers of unique alleles were found in the cultivars with improved traits. The distributions of unique allele counts also varied greatly across the 21 chromosomes with respect to trait improvement. Significant SSR variation between two cultivar groups was observed for improvement in seven traits, but not in stem rust. The proportional SSR variation residing between two groups ranged from 0.014 to 0.118. The proportional SSR variations within the improved cultivar groups consistently were much lower than those within the non-improved groups. These findings clearly demonstrate the association between allelic changes and wheat trait improvements and are useful for understanding the genetic modification of the wheat genome by long-term wheat breeding.  相似文献

Carrot cell walls have been shown to contain significant quantities of esterified p-hydroxybenzoic acid, which is presumed to be esterified to cell wall polymers. The purpose of this study was to investigate the distribution of p-hydroxybenzoic acid and related phenolics among carrot cell wall polysaccharides. Cell wall material was prepared from fresh carrot root tissues and extracted sequentially with water, imidazole, cyclohexane- trans-1,2-diamine- N, N, N', N'-tetraacetate, Na 2CO 3, and KOH (0.5, 1, and 4 M) to leave a cellulose-rich residue. The fractions were analyzed for their carbohydrate and phenolic acid components. Selected soluble fractions were subfractionated further by graded precipitation in ethanol. The majority of the polymer fractions comprised pectic polysaccharides, with varying quantities of neutral sugars (arabinose and galactose). Hemicellulosic polymers were generally found only in the strong alkali extracts (4 M KOH). p-OH-benzoic acid was the predominant phenolic ester and was associated with most fractions analyzed; p-OH-benzaldehyde was also detected in the fractions at much lower levels. Principal components analysis of the chemical data indicated that the p-OH-benzoic acid was associated predominantly with the branched pectic polysaccharides, in contrast to the p-OH-benzaldehyde. The possible roles and functional properties of these phenolics are discussed.  相似文献

Analysis of wheat bran and spent grain shows that concentrations of ferulate and diferulates offer considerable scope to modify the cross-linking of feruloylated polysaccharides and hence the mechanical properties of these residues. In solution ferulic acid (FA) can be readily polymerized by horseradish peroxidase, but when esterified to a polysaccharide, the profile of diferulates becomes restricted. This situation also exists in muro and suggests structural constraints may limit the availability of FA for cross-linking. At relatively low polysaccharide concentration, (approximately 3%), steric restrictions were apparent in gels prepared using isolated polysaccharides. Mechanical properties such as swelling also appear to be fixed at these relatively low polysaccharide concentrations. This limits the potential to modify mechanical properties in muro through oxidoreductase activity. To modify mechanical properties such treatments will need to focus on increasing the "flexibility" of the cell wall matrix and the accessibility of enzymes to the cell wall matrix.  相似文献
