ABSTRACT

Laboratory experiments were conducted to investigate how incorporation of the winter grasses foxtail (Alopecurus aequalis) and milk vetch (Astragalus sinicus) into flooded paddy soil affects the germination of summer weeds and soil inorganic nitrogen (N) levels. Polyethylene terephthalate (PET) vessels (10.0 × 8.0 × 15.0 cm) were filled with 0.7 kg air-dried soil that had been collected from organic paddy fields located in Tochigi, Japan. Foxtail and milk vetch were then independently incorporated into the soil at rates of 0% (untreated control), 0.25%, 0.5%, 1%, and 1.5%, with four repetitions per treatment. The PET vessels were placed in a growth chamber (Biotron) set to day/night temperatures and humidities of 27/18°C and 70%/65%, respectively, and with an illuminance of 40,000 lx (12 h) for 28 days, and the total number of weeds and changes in the electrical conductivity (EC) of the surface water were investigated throughout this period, while changes in soil inorganic N were determined at the ends of the experiments. The incorporation of both foxtail and milk vetch significantly suppressed the germination of summer weeds by 56% and 50%, respectively, at incorporation rates of 1% and by 78% and 77%, respectively, at incorporation rates of 1.5% compared with their respective controls. There was a high and significant positive correlation between both the rate of winter grass incorporation and the degree of growth suppression and EC. Incorporation of foxtail at rates of 0.25% and 0.5% significantly increased the inorganic N contents of the sub-layer by an average of 68% compared with the control but had no significant effect in the top layer, whereas the incorporation of milk vetch at a rate of 1% significantly increased the inorganic N contents of both the top layer and sub-layer by 200% and 316%, respectively, compared with the control. Furthermore, the effect of milk vetch on inorganic N significantly increased with an increase in the rate of incorporation. These findings demonstrate that incorporation of the winter grasses foxtail and milk vetch into flooded organic paddy soil can suppress weed growth and act as a source of inorganic N.  相似文献

Aquaporins (AQPs) function as water channels in many types of cells involved in fluid transport. More than 10 isoforms have been identified, and these are differentially expressed in many types of mammalian cells in the body. Six AQPs (AQP0, AQP1, AQP3, AQP4, AQP5, and AQP9) have been identified in the eyes of humans and/or rodents. The unique permeability characteristics and distribution of AQPs indicate their diverse roles in the regulation of water homeostasis in the eye. The aim of this study was to investigate the localisation of AQPs in normal canine eyes, with AQP0 protein expressed in the crystalline lens and retina. Although AQP1 mRNA was detected in various areas of the canine eye, its protein expression was limited to the cornea, iris and ciliary body. AQP4 was identified in the iris, retina and optic nerve. AQP3 and AQP5 were found in the cornea and conjunctiva, and their expression was particularly high in the limbus. AQP3 and AQP5 were present in the nictitating membrane indicating that they play a role in water transport within the membrane. The observations suggested that several subtypes of the AQP family are involved in the regulation of water homeostasis in the canine eye.  相似文献

Trees of the family Dipterocarpaceae are the dominant trees in Southeast Asian tropical forests where they play an important ecological role and are also important commercially. An experiment was conducted to determine the effect of ectomycorrhizal fungi on the growth of dipterocarp species in peat soils. Seedlings of Shorea pinanga were inoculated with spores of two ectomycorrhizal fungi, Pisolithus arhizus and Scleroderma sp. were grown in pots containing sterilized peat soil for 7 months. The percentage of ectomycorrhizal colonization on S. pinanga exceeded 86%. Colonization of S. pinanga roots by ectomycorrhizal fungi resulted in increased shoot height, stem diameter, number of leaves, and shoot fresh and dry weight. Survival rates of S. pinanga were greater for inoculated seedlings than control seedlings. These results suggest that inoculation of ectomycorrhizal fungi can improve the early growth of S. pinanga grown in tropical forests and that this technique will accelerate the rehabilitation of degraded dipterocarp forests.  相似文献

It has been suggested that plasma membrane (PM) lipids play a major role in aluminum (Al) tolerance; however, no direct investigations have been carried out using PM lipids from root-tips. Here we report a new technique for PM isolation as an alternative to the laborious two-polymer phase partitioning method that is commonly applied, as follows: 1) separation of protoplasts from 1-cm root-tip portions by enzymatic digestion, 2) attachment of the purified protoplasts to glass plates coated with polylysine, 3) preparation of PM ghosts by successive burst of the attached protoplasts using three separate buffer solutions (25 mM PIPES, 5 mM EDTA, and 2 mM MgCl2, at pH 7.0) with slow stirring for 60 s. The PMs were confirmed to be devoid of organelle membranes by fluorescence microscopy, thin layer chromatography (TLC) and western blot analysis. The PM lipids obtained were found to be useful for studies on their differential permeability and lipid composition between lines of triticale or cultivars of maize under Al stress.  相似文献

The present study describes the isolation, cloning and characterization of adipogenic progenitor cells from rat skeletal muscle. Among the obtained 10 clones, the most highly adipogenic progenitor, 2G11 cells, were further characterized. In addition to their adipogenicity, 2G11 cells retain myogenic potential as revealed by formation of multinucleated myotubes when co‐cultured with myoblasts. 2G11 cells were resistant to an inhibitory effect of basic fibroblast growth factor on adipogenesis, while adipogenesis of widely used preadipogenic cell line, 3T3‐L1 cells, was suppressed almost completely by the same treatment. In vivo transplantation experiments revealed that 2G11 cells are able to possess both adipogenicity and myogenicity in vivo. These results indicate the presence of bipotent progenitor cells in rat skeletal muscle, and suggest that such cells may contribute to ectopic fat formation in skeletal muscle.  相似文献

Zinc (Zn) is an essential nutrient for human beings, and most Zn intake occurs through vegetables or cereals such as rice (Oryza sativa L.) grains. Recently, we detected Zn as well as cadmium, which may be partitioned to rice grains, in the phloem saps from the uppermost internodes of rice and in the xylem saps from the cut stems at early grain-filling. To quantify Zn transport to the grains via the phloem and xylem, a mathematical model previously developed for cadmium transport to rice grains was applied. We examined the translocation of zinc into the grains of rice plants at early grain-filling by feeding zinc-65 (65Zn) via a root-bathing medium, through culm cuts above and below the flag-leaf nodes, and through the flag leaves. The estimate made using the mathematical model and experimental data for three types of 65Zn transport suggests that the grain Zn may be accumulated predominantly via the phloem through two means of transport, phloem transport of stored Zn from the leaves and, more importantly, xylem-to-phloem transfer at the nodes from Zn as it is being absorbed. The Zn transport via the phloem to the grains is more selective than that of cadmium, a non-nutrient element, as also evidenced by the greater transport of cadmium to the glumes via the xylem.  相似文献

We investigate the effect of Al on K+ uptake by rice roots. Potassium-38 (38K), a positron emitting nuclide (the half-life: 7.61 min), was used to trace K+ behavior. When a rice root was treated with 10μM Al for 24 h, the uptake of 38K in the root was increased in the range of 1 to 2 cm from the root tip compared with that of the control sample. Because the root continued to grow without showing any damage of plasma membrane during the Al treatment, it was suggested that the 38K uptake was not occurred through diffusion into the cells. The uptake of 38K in both treatments, with/without Al, was decreased by VO43- (inhibitor of H+-ATPase on plasma membrane) and DNP (H+ ionophore) treatment, which suggested that the K+ uptake was performed through an active transport, such as H+:K+ transport or H+ gradient promoted by an Al treatment.  相似文献

The influence of Hg on soybean plants under different pH conditions and Hg concentrations was studied. Growth inhibition by Hg was higher in roots than the upper part of the plant, but was highly dependant on pH condition. Growth inhibition of roots was observed when Hg concentration was higher than 1 mg Hg L−1 for pH 4.0 and 5 mg Hg L−1 for pH 6.0. Using 203Hg as a radioactive tracer, the amount of Hg (1 mg Hg L−1) uptake in root was found to be about 1.5 times higher at pH 4.0 than that at pH 6.0; suggesting that Hg when highly accumulated at the lower pH induced inhibition of root growth. Decreased amounts of Hg due to evaporation during the plant growth were very low, but were higher at pH 6.0 than that at pH 4.0. There was hardly any translocation of Hg from roots to the upper parts through the stem within 24 h.  相似文献
