Neuroaxonal dystrophy (NAD) was examined in two Papillon dogs and a mix breed dog between Papillon and Chihuahua. In addition, cerebellar cortical abiotrophy (CCA) in a Papillon dog, which had similar clinical and magnetic resonance imaging (MRI) features to those of NAD, was also investigated. The common clinical symptoms of all dogs affected with NAD and CCA, were pelvic limb ataxia and cerebellar ataxia including intention tremor, head tremor, and hypermetria in the early onset. These clinical signs were progressed rapidly, and two dogs with NAD were euthanized by owner's request and the other two died by aspiration pneumonia. MRI examinations and gross observations at necropsy revealed moderate to severe cerebellar atrophy in all cases of NAD and CCA. The most typical histological change of NAD was severe axonal degeneration with marked spheroid-formation in the dorsal horn of the spinal cords, the nuclei gracilis, cuneatus, olivalis and its circumference in the medulla oblongata. The spheroids were characterized as large eosinophilic or granular globes within the enlarged myelin sheaths, sometimes accompanied by moderate accumulation of microglias and/or macrophages. In contrast, such spheroid formation was minimal in the brain of CCA. In the cerebellum, mild to moderate loss of the Purkinje and granular cells were recognized in three dogs with NAD, whereas these changes were more prominent in a dog with CCA. Although the clinical signs and MRI findings relatively resembled between NAD and CCA, the histopathological features considered to be quite differ, suggesting distinct pathogenesis and etiology. Since both NAD and CCA are proposed as the autosomal recessive hereditary disorders, careful considerations might be needed for the breeding of Papillon and Chihuahua dogs. 相似文献

A Pomeranian dog, 1 year- and 8 month-old neutered female, was presented with persistent
respiratory distress and recurrent generalized demodicosis. Physical examination revealed
cyanosis, rough respiratory sounds, multifocal alopecia and dermal erosions on the dorsal
side of the forelimbs, perineal area and skin around the eyes. A severe diffuse
interstitial lung pattern was observed on thoracic radiographs. The blood examination
revealed neutrophilia and hypoglobulinemia. Serum immunoglobulin concentrations of IgG and
IgA were low. Histopathological examination revealed severe diffuse interstitial pneumonia
with Pneumocystis carinii infection. Severe lymphoid depletion was
observed in the spleen and other organs with lymphoid follicles consisted mainly of
CD3-positive T cells and few cells of B-cell lineage. B-cell hypoplasia with subsequent

Studies in vivo and in vitro suggest that curcumin is a neuroprotective agent. Experiments were conducted to determine whether dietary supplementation with curcumin has neuroprotective effects in a mouse model of Parkinson’s disease (PD). Treatment with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) significantly induced the loss of dopaminergic cells in the substantia nigra and deletion of dopamine in the striatum, which was attenuated by long-term (7 weeks) dietary supplementation with curcumin at a concentration of 0.5% or 2.0% (w/w). Although curcumin did not prevent the MPTP-induced apoptosis of neuroblasts in the subventricular zone (SVZ), it promoted the regeneration of neuroblasts in the anterior part of the SVZ (SVZa) at 3 days after MPTP treatment. Furthermore, curcumin enhanced the MPTP-induced activation of microglia and astrocytes in the striatum and increased the expression of glial cell line-derived neurotrophic factor (GDNF) and transforming growth factor-β1 (TGFβ1) in the striatum and SVZ. GDNF and TGFβ1 are thought to play an important role in protecting neurons from injury in the central and peripheral nervous systems. These results suggest that long-term administration of curcumin blocks the neurotoxicity of MPTP in the nigrostriatal dopaminergic system of the mouse and that the neuroprotective effect might be correlated with the increased expression of GDNF and TGFβ1. Choroid plexus tumor (CPT) is a primary intracranial neoplasm of the choroid plexus epithelium in the central nervous system. In the current World Health Organization classification, CPT is classified into two categories; choroid plexus papilloma (CPP) and carcinoma (CPC). In the present study, we investigated immunohistochemical expressions of N-cadherin, E-cadherin and β-catenin in 5 canine CPT cases (1 disseminated CPC, 2 CPCs and 2 CPPs). One CPP case was positive for N-cadherin and β-catenin, but negative for E-cadherin. The disseminated CPC case was positive for E-cadherin and β-catenin, but negative for N-cadherin. The other cases were positive for the three molecules examined. These results suggest that loss of the N-cadherin expression might associate with the spreading of CPC cells. 相似文献

Soil characteristics regulate various belowground microbial processes including methanogenesis and, consequently, affect the structure and function of methanogenic archaeal communities due to change in soil type which in turn influences the CH4 production potential of soils. Thus, five different soil orders (Alfisol, Entisol, Inceptisol, Podzol and Vertisol) were studied to assess their CH4 production potential and also the methanogenic archaeal community structure in dryland irrigated Indian paddy soils. Soil incubation experiments revealed CH4 production to range from 178.4 to 431.2 μg CH4 g-1 dws in all soil orders as: Vertisol<Inceptisol<Entisol<Podzol<Alfisol. The numbers of methanogens as quantified using real-time quantitative polymerase chain reaction (qPCR) targeting mcrA genes varied between 0.06 and 72.97 (×106 copies g-1 dws) and were the highest in Vertisol soil and the least in Alfisol soil. PCR-denaturing gradient gel electrophoresis (DGGE)-based approach targeting 16S rRNA genes revealed diverse methanogenic archaeal communities across all soils. A total of 43 DGGE bands sequenced showed the closely related groups to Methanomicrobiaceae, Methanobacteriaceae, Methanocellales, Methanosarcinaceae, Methanosaetaceae and Crenarchaeota. The composition of methanogenic groups differed among all soils and only the Methanocellales group was common and dominant in all types of soils. The highest diversity of methanogens was found in Inceptisol and Vertisol soils. Methane production potential varied significantly in different soil orders with a positive relationship (p?<?0.05) with methanogens population size, permanganate oxidizable C (POXC) and CO2 production. In the preceding paper (1), the authors had determined the infrared absorption spectra of humic acids obtained from various soils of different origin, and concluded that they were, according to their infrared characteristics, classified into four groupes, i.e., A,B,Rp, and P types. After that, the authors conducted some experiments of the infrared spectra of soil components in order to get some information on the nature of so-called humin, and forms of soil clay and clay humus complex. The infrared spectra of silica, sesquioxides, clay minerals and humates of aluminium, iron and calcium were also determined for comparison. Abstract The effect of the addition of bromacil (pesticide) or/and a sewage sludge on the urease, phosphatase and dehydrogenase activities of soil was studied. Urease and phosphatase activities increased initially with the addition of bromacil. This effect disappeared after 28 d of soil incubation. The increase in the urease and phosphatase activities caused by the addition of sewage sludge was more pronounced than that of bromacil. The combined addition of sewage sludge and bromacil also led to an increase in the activity of both hydrolases. Dehydrogenase activity was affected negatively by the addition of bromacil and positively by the sewage sludge addition. When sewage sludge and bromacil were added simultaneously, dehydrogenase activity was higher than when sewage sludge alone was added. 相似文献

ABSTRACTExcessive application of animal manure to farmland leads to phosphorus (P) loss into the surrounding water. Manure is incinerated to convert it to P-rich ash as a slow-release P fertilizer. However, the potential P loss and P availability for plants from cattle manure ash (CMA) have not been fully understood. The aims of this study were to determine the P release mechanism from CMA and to propose appropriate application rates that mitigate P loss and increase available P to soil in Fukushima, where the soil is deficient in nutrients after the replacement of cesium-137-contaminated soil with sandy mountain soil. Different P fractions in CMA were sequentially extracted with H2O, 0.5 M NaHCO3, 0.1 M NaOH, and 1 M HCl. Phosphorus contents in different fractions of CMA were in the order of HCl–P > NaHCO3–P > H2O–P > NaOH–P. Water-soluble P release of CMA was also determined by kinetic experiments for 120 h. Results showed that total water-soluble P accounted for a maximum of 2.9% of total P in CMA over 120 h due to recalcitrant P compounds formed through incineration. The Fukushima sandy soil amended with CMA at three application rates, 94, 157, and 314 mg P kg?1 (corresponding to 300, 500, 1000 kg P2O5 ha?1) was incubated for 56 days. Cattle manure compost and KH2PO4 were applied at 157 mg P kg?1 for comparison. Phosphorus release in water and CaCl2 solution from ash-amended soil was significantly lower than those from compost and KH2PO4-amended soil at the same P application rate of 157 mg P kg?1 (p < 0.05). Available P in ash-amended soil, determined by Fe-oxide impregnated strips, was not significantly different from those in compost-amended soil after day 7 and KH2PO4-amended soil on day 56 at the same P application rate. Thus, CMA reduces P losses from soil to the surrounding water while it increases P availability for plants. In comparison of different rates of CMA, P release in water or CaCl2 was significantly greater at 314 mg P kg?1 than at 94 or 157 mg P kg?1, while the percentage of available P to total P was the lowest at the highest application rate (p < 0.05), suggesting that the best application rates were 94 and 157 mg P kg?1 in this experiment. 相似文献

In a previous paper (1), a modification of the chloroform fumigation method (2) was proposed for estimating microbial biomass-nitrogen (N) in submerged soil. It was found that a large part of the living bacteria in the submerged soil was killed within about 12 h of the fumigation and was actively mineralized by subsequent anaerobic incubation. In this paper, the following factors which influence the estimated amount of microbial biomass-N were examined: a) inoculation to fumigated soil, b) duration of anaerobic incubation, c) storage and sieving pretreatments of soil, and d) effect of fumigation on soil organic matter. 相似文献

A direct competitive enzyme-linked immunosorbent assay (dc-ELISA) was developed for residue analysis of azoxystrobin in garden crops, for which the maximum residue limits (MRLs) are 0.5-50 mg/kg in Japan. For hapten synthesis, an ethyl carboxyl group was introduced to the 4-position of the 2-cyanophenoxy group in azoxystrobin, and its cyano group was changed to a methyl group. An anti-azoxystrobin monoclonal antibody was prepared from mice immunized with hapten-keyhole limpet hemocyanin conjugate. The dc-ELISA using prepared antibody showed 50-250-fold higher sensitivity compared to the MRLs. The working range of the dc-ELISA was 10-200 ng/mL. The dc-ELISA showed high specificity to azoxystrobin. When methanol extracts from nine kinds of garden crops spiked with azoxystrobin ranging near the MRLs were analyzed, the determined results by the dc-ELISA agreed well with the results of their controls. In addition, azoxystrobin spiked in garden crops homogenates was satisfactorily extracted by methanol solution and easily analyzed. The recovery rate of dc-ELISA was 96-109% and correlated well with the results obtained by HPLC analysis. 相似文献
