Uncoupling protein 1 (UCP-1) was believed to be an exclusive protein found in the brown adipose tissue of small rodents and humans; however, recent studies show that the expression of UCP-1 protein has been found in the sebaceous glands of the mouse tail and human skin. There are a few reports about the presence of UCP-1 in the sebaceous glands of other rodents, such as the Sunda porcupine (Hystrix javanica), a wild spiny rodent commonly found in Indonesia with a large sebaceous gland. The aim of this study was to identify the presence of UCP-1 in the sebaceous glands on the skin of the Sunda porcupine. The skin from three regions (thoracodorsal, lumbosacral and apex caudal) of eight adult Sunda porcupines was used to detect UCP-1-immunopositive cells through immunohistochemistry. All three regions were found immunopositive to anti-UCP-1 antibody in the sebaceous gland of quill and hair follicles, and the epidermal layer in quill and hair follicles with various intensities. The result of immunohistochemistry revealed that the thoracodorsal and apex caudal region was the most intense immunoreaction followed by the lumbosacral region. These findings proved that the presence of UCP-1 was also identified in the sebaceous glands of other rodent (Hystrix javanica) and regions of the body, which has not been reported previously.  相似文献

It is well known that some caspases in apoptosis is involved in determinant of terminal differentiation and maturation of various cells. Our previous study ultrastructurally clarified the differentiation into M cells from immature microvillous epithelial cells and the redifferentiation from M cells to microvillous epithelial cells in the follicle-associated epithelium (FAE) of rat Peyer's patch. In this study, the difference of epithelial apoptosis between the FAE of Peyer's patch and intestinal villi was immunohistochemically investigated in rat jejunoileum. As a result, cleaved caspase-3 was limited to several epithelial cells at the tip of FAE, whereas almost all of the epithelial cells were cleaved caspase-3 positive in intestinal villi. Cleaved caspase-9 was detected only in a few exfoliating or exfoliated epithelial cells of both FAE and intestinal villi. Nuclear DNA-fragmentation was detected only in several epithelial cells of the tip of FAE, while it was expressed from the middle regions in the intestinal villi. The DNase I expression of the epithelial cytoplasm was much weaker in FAE than in intestinal villi. Bcl-x expression was restricted in the apical cytoplasms of epithelial cells in the FAE, whereas it was restricted in whole cytoplasms in villous epithelial cells. These findings suggest that the progression of the apoptotic process in the epithelial cells of FAE is later than in the intestinal villi, so that the possibility of epithelial differentiation might be remained in the FAE, unlike in the intestinal villi.  相似文献

The relationship between the charge characteristics and the dispersibility of soils from reclaimed land (highly dispersive) and adjacent forest area (physically stable) was investigated.

To evaluate the amount of soil charges quantitatively, the measurement based on cation and anion adsorption (ion adsorption method) was attempted in two ways, where special attention was paid to the solution concentration. In the conventional ion adsorption method, the treatments with concentrated solutions (1 mol L-1) were included in the process. In the modified method, soils were treated with dilute solutions (5 mmol L-1), in taking account of the soil solution concentration in humid temperate regions. The amount of charges derived from the modified method was smaller than that from the conventional one, especially in the high pH zone. In a preliminary experiment, the extraction of polyvalent cations from soils was found to be significantly affected by the salt concentration. Thus, the use of solutions with an excessively high concentration was considered to result in an error in the estimation of the active charges in soils which would contribute to the dispersion behavior in the field.

The applicability of the modified method for analyzing the dispersion behavior of soils was evaluated by relating the dispersibility of silts and clays at different pHs with the charge characteristics determined under the same ionic strength. The dispersion ratio of silts of the reclaimed land reached maximum values at pH 5.5, above which the ratio decreased, whereas the ratio of clays began to increase in the pH range above 6. The dispersion ratio of silts of the forest area increased in the pH range above 6, whereas that of clays remained constant at a low level in all the pH ranges below 7. The changes in the dispersibility of silt plus clay fractions corresponded to total charge characteristics determined by the modified method. Thus, the evaluation of soil charges by the modified ion adsorption method was considered to be effective to predict soil dispersibility under field conditions, indicating that it could replace electrokinetic analyses such as electrophoresis in studies on the dispersion/flocculation behavior of soils.  相似文献

为探究生物刺激素产品在春季花椰菜绿色防控生产中的应用效果,2019年在北京市大兴区开展4种生物刺激素对花椰菜生产的影响试验,比较植株长势、结球率、单球直径、单球质量、品质等指标,以筛选适合推广使用的生物刺激素。结果表明:苗期施用大丽轮枝孢激活蛋白(处理2)可以促进花椰菜苗期生长,推荐用量为苗期施用大丽轮枝孢激活蛋白5000倍液,定植后30 d施用12000倍液;定植后施用大丽轮枝孢激活蛋白(处理6)可以提高花椰菜的产量,推荐定植后7 d和结球前各施用1次大丽轮枝孢激活蛋白6000倍液;采用以上处理方法可以增加花椰菜的维生素C含量,提高品质和安全性。  相似文献

To confirm ability forming the basement membrane of the regenerated laminar epidermis (rLE) in chronic laminitis, expression of type VII and type XVII collagen mRNAs in the rLE was studied applying sequences of two type of murine collagens. On northern blot analysis, complement DNA (cDNA) probes adjusted from the murine type VII and type XVII collagen could hybridize with the equine mRNAs, and each signal was detected as single-bands at approximately 9.5 kb and 5.6 kb, respectively. Contrasting with the expression level of equine glyceraldehyde-3-phosphate dehydrogenease mRNA, the band of type VII collagen mRNA in laminitis was stronger than normal, but the type XVII collagen mRNA in laminitis was less than normal. By in situ hybridization, positive signals in response to the murine type VII and type XVII collagen mRNA probes could be detected in the equine laminitic rLE region. From these results, it is concluded that the keratinocytes constructing the rLE in chronic stage of laminitis can express type VII and type XVII collagen mRNAs and these expression patterns were different from the normal.  相似文献

Cutting rot of chrysanthemum was found on cuttings of cv. Jimba No.2 in 2008. The cuttings were imported, then transplanted in Aichi Prefecture. Root development was not initiated in about 30% of the cuttings. The cut stem ends developed black discolouration and decay. When healthy cuttings were the fungus isolated from diseased cuttings, these cuttings developed the same disease symptoms. The characteristics and morphology of the fungal culture were identical to those of Plectosporium tabacinum. We propose that the new disease be named cutting rot of chrysanthemum.  相似文献

We investigated the profiles of circulating levels of inhibin A and total inhibin in beef cows with follicular cysts in relation to the patterns of follicular development and circulating gonadotropins and steroid hormones. Turnover of follicular waves was monitored in five cows every 2 days for 70 days from 10 days after detection of estrus without ovulation. The mean interwave intervals were 19.6 ± 1.0 days (n = 18 waves with cysts from the five cows). Circulating levels of inhibin A were approximately 170 pg/ml before emergence of follicular waves with cysts and increased (P < 0.05) concomitantly with follicle emergence. High concentrations of inhibin A (greater than 300 pg/ml) were noted for 7 days during the growth phase of cystic follicles, but inhibin A levels decreased gradually when development of the cysts reached a plateau. This profile of inhibin A was similar to those of total inhibin and estradiol, but was inversely related to the changes in plasma FSH concentrations. LH pulse frequency and mean concentrations of LH in cows with cysts were higher than those observed in the luteal phase of normal cyclic cows. These results indicate that the capacity to secrete inhibin, as well as estradiol, is maintained in cystic follicles, the growth of which is extended by LH secretion at levels greater than those seen in the normal luteal phase. Inhibin A plays an important role in the extension of interwave intervals by suppressing recruitment of a new cohort of follicles.  相似文献

The purpose of present study was to determine annual changes in serum progesterone (P(4)) concentrations and to clarify basic reproductive characteristics, such as breeding season, estrous cycle, and puberty in female bharals (Pseudois nayaur). Blood was collected from 9 female bharals once or twice weekly for approximately one year. Serum P(4) concentrations were determined by radioimmunoassay. Serum P(4) concentrations showed remarkable and cyclic changes between November/December (winter) and May/June (late spring). The mean estrous cycle was 24.9 +/- 0.5 days. Chasing insistently to other females and discharge of mucus from the vulva were observed around the time when the serum P(4) concentrations began to increase. The chasing behavior and discharge of mucus were considered to be external indicators of estrus in female bharals. Serum P(4) concentrations of a pregnant female had non-cyclic changes, and the values remained high. In this study, all 37 deliveries were between April and September, and about 70% of these were concentrated in May and June. The conception month determined on the day of birth was between October and April for all animals, and the most common month was in December (54%). This month corresponded to an early stage of the period when the serum P(4) concentrations changed cyclically. These results indicate that many female bharals become pregnant at the beginning of the breeding seasons and, if they do not become pregnant, the estrous cycle, about 25 days in length, is repeated.  相似文献

Pluripotent stem cells, such as embryonic stem cells (ESCs) and induced pluripotent stem cells (iPSCs) are able to differentiate into all cell lineages of the embryo proper, including germ cells. This pluripotent property has a huge impact on the fields of regenerative medicine, developmental biology and reproductive engineering. Establishing the germ cell lineage from ESCs/iPSCs is the key biological subject, since it would contribute not only to dissection of the biological processes of germ cell development but also to production of unlimited numbers of functional gametes in vitro. Toward this goal, we recently established a culture system that induces functional mouse primordial germ cells (PGCs), precursors of all germ cells, from mouse ESCs/iPSCs. The successful in vitro production of PGCs arose from the study of pluripotent cell state, the signals inducing PGCs and the technology of transplantation. However, there are many obstacles to be overcome for the robust generation of mature gametes or for application of the culture system to other species, including humans and livestock. In this review, we discuss the requirements for a culture system to generate the germ cell lineage from ESCs/iPSCs.  相似文献

This study evaluated the effects of condensed barley distillers soluble (CBDS) on growth rate, rumen fermentation, plasma metabolite, and myofiber properties, and gene expression related to metabolism in the skeletal muscles of Japanese Black calves, compared with soybean meal and corn. Twenty‐four calves were divided into four groups: fed 5% CBDS based on the hay dry matter weight (low CBDS) and fed soybean meal and corn at the same nutrition level (control); and fed 15% CBDS based on the hay dry matter weight (high CBDS) and fed soybean meal and corn at the same nutrition level (high soy). The daily gain was larger in the low (p = 0.08) and high (< 0.05) CBDS groups compared with the control group. In the CBDS‐fed groups, plasma β‐hydroxybutyric acid concentrations were significantly higher at 6 months of age (< 0.05), the percentage of type I myofibers was significantly lower and their diameters were significantly larger at 9 months of age (< 0.05), and carnitine palmitoyltransferase 1b mRNA expression was significantly lower (< 0.05) and citrate synthase mRNA expression tended to be lower (low; p = 0.06, high; = 0.05) compared with control group. Thus, feeding CBDS promotes growth and leads to animals with more glycolytic and less oxidative muscle metabolism.  相似文献
