Saliva flow induced by 6-gingerol (pungent), hydroxy-α/β-sanshools (tingling), and citric acid (sour) was measured, and the time-dependent changes in the whole saliva proteome were analyzed by means of 2D-PAGE, followed by tryptic in-gel digestion and MALDI-TOF-MS peptide mass fingerprint analysis. The proteins showing significantly decreased abundance after oral 6-gingerol stimulation were identified as glutathione S-transferase P, the heat shock protein β-1, the heat shock 70 kDa protein 1, annexin A1, and cytoplasmic β-actin, whereas prolactin inducible proteins (PIP), short palate, lung and nasal epithelium carcinoma-associated protein 2 (SPLUNC2), zinc-α-2-glycoproteins (Zn-α-GP), and carbonic anhydrase VI (CAVI) were found with increased abundance. As the effects of this study were observed instantaneously upon stimulation, any proteome modulation is very likely to result from the release of proteins from preformed vesicles and not from de novo synthesis. The elevated levels of SPLUNC2, Zn-α-GP, and CAVI might be interpreted to trigger innate protective mechanisms in mucosal immunity and in nonimmune mucosal defense and might play an important role during the initial stage of inflammation.  相似文献

Intermittent administration of parathyroid hormone (PTH) is an anabolic therapy for osteoporotic conditions in humans. This study evaluated the effects of equine PTH fragment (ePTH-1-37) administration on bone metabolism in 12 healthy horses. Six horses each were treated once daily for 120days with subcutaneous injections of 0.5μg/kg ePTH-1-37 or placebo. Blood was collected to determine ionized calcium (Ca(++)), total Ca (Ca(T)), inorganic phosphorus, serum equine osteocalcin (eOC), carboxy-terminal telopeptide of type I collagen (ICTP), bone-specific alkaline phosphatase, and carboxy-terminal cross-linked telopeptide of type I collagen. Bone mineral density (BMD) was determined with dual X-ray absorptiometry of the metacarpus and calcaneus. Significantly higher blood Ca(++) and plasma Ca(T) concentrations were measured 5h after ePTH-1-37 administration compared to placebo. Higher serum eOC concentrations were found for ePTH-1-37 treatment at days 90 (P<0.05) and 120 (P=0.05). Significantly higher serum ICTP levels were observed with ePTH-1-37 treatment at days 60 and 90. For both study groups, BMD increased significantly in the calcaneus. Long-term use of ePTH-1-37 seemed to have no negative effects on bone metabolism in healthy horses. The absence of undesirable side effects is the premise to ensure safety for further clinical investigations in horses with increased bone resorption processes.  相似文献

Cell-to-cell interaction via cell contact-dependent pathway is essentially important for maintenance and regulation of corpus luteum (CL) integrity and its physiological actions. The objective of the present study was to evaluate the mRNA expression of the cell adhesion molecules (CAMs) that are constituent factors of gap junctions [connexin (Cx) 43] and adherence junctions (VE-, E-, N-cadherin) in two types of endothelial cells from the mid CL and in CL tissue during the estrous cycle and PGF(2alpha)-induced luteolysis in the cow. Specific mRNA expression for Cx43 and N-cadherin was detected in cytokeratin-positive (CK+) and cytokeratin-negative (CK-) luteal endothelial cells (EC) and fully luteinized granulosa cells (LGC). E-cadherin mRNA was expressed in CK+EC and LGC, but not in CK-EC. VE-cadherin mRNA was expressed in both CK+ and CK-EC. During the estrous cycle, Cx43 mRNA expression was significantly lower in the regressing CL. VE-cadherin expression also tended to increase in the mid CL and increased significantly in the regressing CL. E-cadherin mRNA expression was higher in the early and late CL than in the mid- and regressing CL. N-cadherin mRNA expression gradually increased from the early to late CL followed by a decrease in the regressing CL. During PGF(2alpha)-induced luteolysis, Cx43 mRNA expression appeared to increase, and VE-cadherin and E-cadherin mRNA significantly increased at 24 h. N-cadherin mRNA expression decreased 2 and 4 h after PGF(2alpha) administration. Collectively, expression of the mRNAs for CAMs was different in the two types of luteal endothelial cells and fully luteinized granulosa cells and changed independently in the CL during the estrous cycle and PGF(2alpha)-induced luteolysis in the cow. The results suggest that CAMs play physiological roles in cell-to-cell communication to regulate both gap and adherence junctions during CL development and regression in the cow.  相似文献

A 6-week-old female Simmental calf was evaluated for acute non-ambulatory tetraparesis. Physical and laboratory examinations revealed no clinically relevant abnormalities. Neurological findings were consistent with acute, progressive and painful cervical myelopathy. Radiographs displayed a fractured odontoid process (dens axis) and vertebral step misalignment at the fracture site. A traumatic origin was suspected. Advanced diagnostic imaging was considered to allow better planning of potential surgical stabilisation and to exclude any additional lesions of the cervical vertebral column. However, during trailer transportation to the advanced diagnostic imaging and surgery site, the calf deteriorated neurologically and was humanely euthanised. Magnetic resonance imaging (MRI) and computed tomography (CT) were performed immediately post-mortem for scientific reasons. The MRI examination reflected the radiographic findings and confirmed severe spinal cord compression at the fracture site. In addition, a T2W-hyperintense signal change within the paravertebral soft tissue dorsal to the fracture site was indicative of a traumatic event. CT identified the fracture site at the synchondrosis between the odontoid process and the body of the axis, and this finding was confirmed by post-mortem examination. Advanced diagnostic imaging and post-mortem examination did not identify any other cervical lesion. In summary, this calf was diagnosed with a traumatic odontoid process synchondrosis fracture, which has not been reported previously in calves but presents a challenging and well-known fracture type in young children. This case report indicates that the odontoid process synchondrosis is a potential predisposed injury site and that traumatic odontoid process synchondrosis fractures should be considered as a potential differential in calves with acute cervical pain and/or signs of a cervical myelopathy.  相似文献

Purpose

The invasive plant, Impatiens glandulifera (common English name: Himalayan Balsam), is now found in many river catchments in most European countries. Its preference for damp, nutrient-rich soils, along with its intolerance to cold weather and rapid dieback, has implicated it in promoting soil erosion along the riparian zone. Despite the implication, its influence on the sediment dynamics of river systems remains unconfirmed. This communication reports the preliminary findings of ongoing work to investigate a possible link between I. glandulifera and accelerated erosion rates in inland river systems.

Materials and methods

Erosion pins, a micro-profile bridge, and a digital caliper were employed to measure changes in the soil surface profile (SSP) at six separate locations, each contaminated with I. glandulifera, along the riparian zone of a small watercourse in northwest Switzerland. Changes in SSP were also measured at an identical number of nearby locations supporting natural vegetation, in order to establish baseline erosion conditions. Soil surface profiles at all 12 locations were re-measured on seven separate occasions, from October 2012 to May 2013. This covers the time before dieback occurred to the germination and seasonal regrowth of new plants.

Results and discussion

A total of 720 individual SSP measurements were recorded during the above monitoring period. Increasingly negative values relative to initial values were documented at most transects, indicating a net reduction in soil surface elevations. This is interpreted as evidence of the removal (i.e., erosion) of surface material. Paired samples statistical analysis of the data indicate that erosion from contaminated sites was significantly greater than erosion from topographically comparable reference sites (t?=?5.758; P?<?0.05; N?=?359) supporting natural vegetation.

Conclusions

The results provide tentative yet compelling evidence that I. glandulifera promotes soil erosion along the riparian zone of the watercourse investigated. Given the unrelenting spread of this notoriously invasive plant throughout inland river systems in many countries, the likelihood of greater quantities of nutrient-rich sediment entering into aquatic environments may steadily reduce water quality in all affected catchments. An absence of effective control measures capable of halting or even slowing its rate of invasion may make it increasingly difficult for affected European Union member states to meet and then maintain key water quality standards set by the Water Framework Directive (WFD) when fully implemented in 2015.  相似文献

Surface sealing and runoff generation on soils derived from loess and pleistocene deposits An attempt is undertaken to bring together existing knowledge on the occurence of surface sealing, on subprocesses and factors controlling surface sealing, and on hydraulic properties of seals, with special reference to the soil conditions prevailing in Germany. A conceptional model is proposed that relates different subprocesses such as aggregate breakdown, aggregate deformation, particle rearrangement and different factors such as soil cover, microrelief, stone cover, and macropores to surface sealing and runoff generation. We conclude that loess soils and soils formed on pleistocene deposits are particularly prone to surface sealing, and seals formed on these soils can induce runoff at rainfall intensities of 5 mm h?1.  相似文献
