Embryonic stem (ES) cells can differentiate spontaneously into various lineages in vitro. However, spontaneous commitment of ES cells to the adipocyte lineage is rare. In the present study, bone morphogenic protein-4 (BMP-4) is described as a factor inducing adipocyte differentiation from ES cells at a high rate. For this reason, ES-cell-derived embryoid bodies (EBs) in suspension cultures were exposed to different doses of BMP-4 for 5 days before they were plated onto gelatin-coated tissue culture plates. Moreover, the effect of serum-containing and serum-free media in three different combinations was assessed. Plated EBs, stained with Sudan Black and processed for transmission and scanning electron microscopy, were observed daily for adipocyte formation. Treatment with BMP-4 resulted in the appearance of adipocyte clusters in EBs' outgrowth, depending on the doses applied. Early in differentiation, many small fat droplets were observed in adipocytes, while later on they coalesced and formed a few large fat droplets. Adipocyte clusters had a fibrillar and vascular stroma, and each adipocyte was surrounded with a reticular external lamina. Furthermore, the appearance and development of adipocytes and their changes following 2-3 weeks of starvation mimicked live adipose tissue. In fact, understanding the biological activity of growth and differentiation factors is needed to regulate and direct stem cell differentiation to specific cell types in vitro.  相似文献

Hematopoietic stem cells in the bone marrow give rise to lymphoid progenitors, which subsequently differentiate into B and T lymphocytes. Here we show that the proto-oncogene LRF plays an essential role in the B versus T lymphoid cell-fate decision. We demonstrate that LRF is key for instructing early lymphoid progenitors in mice to develop into B lineage cells by repressing T cell-instructive signals produced by the cell-fate signal protein, Notch. We propose a new model for lymphoid lineage commitment, in which LRF acts as a master regulator of the cell's determination of B versus T lineage.  相似文献

A recent dispersal of modern humans out of Africa is now widely accepted, but the routes taken across Eurasia are still disputed. We show that mitochondrial DNA variation in isolated "relict" populations in southeast Asia supports the view that there was only a single dispersal from Africa, most likely via a southern coastal route, through India and onward into southeast Asia and Australasia. There was an early offshoot, leading ultimately to the settlement of the Near East and Europe, but the main dispersal from India to Australia approximately 65,000 years ago was rapid, most likely taking only a few thousand years.  相似文献

  1. Most of the Bornean endemic freshwater mussel (Unionida) species known to date have not been recorded by science for the past 50 years or more, owing to a lack of research effort and presumed population losses.
  2. This study assessed current patterns and recent changes in the diversity and distribution of freshwater mussels in north-eastern Borneo. Physical surveys and interviews were conducted at 24 sites, and anecdotal evidence for current or historical presence of mussels was collected for a further 13 sites.
  3. Native species, i.e. Schepmania sp. and Khairuloconcha sahanae gen. & sp. nov., were only found in one small stream of the Kinabatangan River basin within the Gomantong Forest Reserve, whereas the non-native Sinanodonta cf. woodiana was common across the study area.
  4. Molecular phylogenetics (five genes) of the native taxa, including comparative material from West Kalimantan and Sarawak, revealed: (i) the presence of a new genus with two new, rare species: Khairuloconcha lunbawangorum sp. nov. in the Limbang River basin and K. sahanae in the Kinabatangan River basin; (ii) that Khairuloconcha and Ctenodesma form the Bornean endemic tribe Ctenodesmini trib. nov.; and (iii) that Schepmania represents another Bornean endemic tribe Schepmaniini trib. nov.
  5. Both Khairuloconcha gen. nov. species are known from a single stream each and are apparently restricted to forest stream habitats where they occur at very low densities. Schepmania appears to have a severely contracted range in the Kinabatangan and adjacent basins. We urgently call for full protection of the currently known sites of K. lunbawangorum and K. sahanae, and development of an action plan to save the Bornean freshwater mussel fauna.

The carotid body of the goat was found to be a small oval or rounded parenchymatous organ. It was characterized by its profound vascularity. Delicate septa divided the parenchyma into small feebly denned lobules. Electron microscopy revealed that the parenchyma comprised type I cells, type II cells, nerve endings, axons and fenestrated dilated capillaries. Type I cells were characterized with electron dense-cored vesicles. They showed variations in size and concentration of the dense-cored vesicles and number of mitochondria. The possibility that these variations are reflections of different stages of activity is discussed. Type II cells were less numerous than type I cells, relatively small and devoid of dense-cored vesicles. They usually surrounded small groups of type I cells and associated nerve endings and axons. Presumptive afferent nerve endings characterized with many clear vesicles, occasional large granular vesicles and varying numbers of slender mitochondria, lay apposed to type I cells. Nerve endings of this kind showed afferent and efferent synaptic junctions with type I cells. Presumptive sympathetic efferent endings were occasionally seen within the lobules but never lay apposed to type I cells or afferent nerve ending.  相似文献

The objective of this study was to evaluate the effects of quantitative feed restriction, along with dietary supplementation with a probiotic blend (Protexin) as a natural growth promoter, on the performance, water consumption, mortality rate and carcass traits of meat‐type quails. A total of 250 1‐day unsexed quails were randomly allocated to five equal groups in a completely randomized design. The first group (A) fed a basal diet without any restriction (24 hr/day); the second group (B1) fed the basal diet for 20 hr/day; the third group (B2) fed the basal diet enriched with probiotic (0.1 g/kg diet) for 20 hr/day; the fourth group (C1) fed the basal diet for 16 hr/day; and the fifth group (C2) fed the basal diet enriched with probiotic (0.1 g/kg diet) for 16 hr/day. Birds were fed ad‐libitum from 0–14 days of age, and then the feed restriction regimes started from 14 till 28 days of age. Results showed that quails in the control‐group consumed more feed and water than the other treatment groups (p < .01), however their body weights did not differ (p > .05) compared with the other treated groups. The best feed conversion values were achieved in quails supplemented with probiotic blend (B2 and C2) in comparison with the other groups (p < .01). Feeding probiotic had a positive effect on bird health which reduced the mortality rate. Further, mortality rate was significantly reduced (p < .05) by feed restriction, with or without probiotic supplementation. No carcass parameters were significantly affected (p > .05) by treatments. Our results show that quail could be reared under a feed restriction system, for 4–8 hr daily, along with dietary supplementation of probiotic as growth promoter for better growth performance.  相似文献

1. Three experiments, each involving over 400 pullets, were conducted into the use of whole grain rearing diets. They were concerned with the cereal to be used, the age from which such diets may be used, and the value of a vitamin‐trace element supplement. Each experiment was performed on a brown‐egg and a white‐egg strain.

2. Whole maize was found to be unsuitable.

3. Whole wheat or whole barley diets should not be introduced before the age of 10 weeks to avoid unacceptably high mortality due to cannibalism. Thereafter the principal results are reduced body weight at caging and retarded sexual maturity.

4. The addition of a vitamin‐trace element supplement had some effect in reducing mortality when whole grain diets were introduced before 10 weeks of age.  相似文献

Using a local Algerian cultivar of chickpea, from 9 sites in Eastern Algeria, along a sub-humid to arid gradient, we isolated 60 nodulating isolates, among which 19 were assigned to Mesorhizobium species and 41 to Ensifer meliloti. Trapping revealed great differences among sites for their ability to induce nodules on plant, but no correlation with chemical and physical characterizations of soil samples could be found. Based on 16S rRNA gene sequences, 16S-23S IGS, recA, nodC and nifH sequences, Mesorhizobium isolates were assigned to at least 5 different species, among which one had never been described as nodulating chickpea before. The symbiotic diversity is conversely low, and shared by several species, reflecting horizontal gene transfer. The ability of Mesorhizobium genus to recombine and exchange symbiotic clusters among different species must play a great role in this ability to be associated with chickpea. The question to what extent the symbiotic species would end in this genus is discussed.  相似文献
