The native soils of Galicia (NW Spain) exhibit a biochemical equilibrium such that total soil N is a function of five biochemical
and microbiological parameters: microbial biomass C, mineralized N, phosphomonoesterase, β-glucosidase and urease activities.
To investigate whether the ratio of the total N calculated from biochemical soil properties (Nc) and the total N as measured
by the Kjeldahl method (Nk; Nc/Nk) can be used as an index of soil quality, we determined these variables and consequently
the ratio in three kinds of disturbed soils: an artificially Cu-contaminated soil, two lignite mine soils, and a number of
arable soils. In none of the studied soils did the individual biochemical parameters respond consistently to the factors influencing
soil quality, but in all cases soil degradation was reflected by the Nc/Nk value, which differed more or less markedly from
100%. Nc/Nk can therefore be used for the rapid evaluation of soil degradation, since it distinguishes among biochemically
balanced soils, soils in a transient state of high microbiological and biochemical activity and degraded soils. It can also
serve as a reliable basis for the rapid calculation of the "ecological dose" (ED50) of soil pollutants. The use of Nc/Nk as an objective index of the biochemical quality of soils is recommended.

In this study polyphenolic compounds extracted from olive fruits of five registered cultivars were analyzed. A solid-liquid extraction (LSE) procedure with Extrelut cartridge (diatomaceous earth) using different eluents was developed to obtain polyphenolic compounds. HPLC-DAD and HPLC-MS methods were applied for the quali-quantitative analysis of each fraction obtained from LSE. The results of this work show that the LSE procedure with diatomaceous earth cartridge supplies a rapid and reproducible fractioning method able to obtain a quantitative recovery of all compounds and to collect fractions directly analyzed by HPLC. A comparison among different cultivars shows significant quantitative differences in some polyphenols, such as verbascoside, anthocyanic compounds, and oleuropein derivatives. 相似文献

Near-infrared reflectance spectroscopy (NIRS) was examined as a possible alternative to high-performance liquid chromatography (HPLC) for the analysis of ginsenosides from American ginseng (Panax quinquefolium) root powder (n = 26). NIR spectra were collected over 400-2500 nm. For each sample and individual ginsenoside quantified by HPLC, spectral data were regressed against the chemical data to develop prediction equations. The spectral prediction equations produced high correlation coefficient (1-VR) values and low standard errors of cross validation (SECV) values for the determination of individual and total ginsenosides. The contents of individual ginsenosides, Rb(1), Rb(2), Rc, Rd, Re, Rg(1), Ro, m-Rb(1), m-Rb(2), m-Rc, m-Rd, and total ginsenosides (X +/- SECV) were (1.29+/-0.18)%, (0.273+/-0.096)%, (0.298+/-0.052)%, (0.091+/-0.027)%, (1. 015+/-0.114)%, (0.116+/-0.018)%, (0.25+/-0.040)%, (0.776+/-0.116)%, (0.197+/-0.074)%, (0.239+/-0.083)%, (0.143+/-0.042)%, and (4.393+/-0.283)%, respectively. The (1-VR) values of cross validation were 0.877, 0.872, 0.955, 0.834, 0.899, 0.919, 0.325, 0.849, 0.902, 0.877, 0.871, and 0.963, respectively. Fresh strawberries (Fragaria x ananassa Duch.), raspberries (Rubus idaeus Michx.), highbush blueberries (Vaccinium corymbosum L.), and lowbush blueberries (Vaccinium angustifolium Aiton) were stored at 0, 10, 20, and 30 degrees C for up to 8 days to determine the effects of storage temperature on whole fruit antioxidant capacity (as measured by the oxygen radical absorbing capacity assay, Cao et al., Clin. Chem. 1995, 41, 1738-1744) and total phenolic, anthocyanin, and ascorbate content. The four fruit varied markedly in their total antioxidant capacity, and antioxidant capacity was strongly correlated with the content of total phenolics (0.83) and anthocyanins (0.90). The antioxidant capacity of the two blueberry species was about 3-fold higher than either strawberries or raspberries. However, there was an increase in the antioxidant capacity of strawberries and raspberries during storage at temperatures >0 degrees C, which was accompanied by increases in anthocyanins in strawberries and increases in anthocyanins and total phenolics in raspberries. Ascorbate content differed more than 5-fold among the four fruit species; on average, strawberries and raspberries had almost 4-times more ascorbate than highbush and lowbush blueberries. There were no ascorbate losses in strawberries or highbush blueberries during 8 days of storage at the various temperatures, but there were losses in the other two fruit species. Ascorbate made only a small contribution (0.4-9.4%) to the total antioxidant capacity of the fruit. The increase observed in antioxidant capacity through postharvest phenolic synthesis and metabolism suggested that commercially feasible technologies may be developed to enhance the health functionality of small fruit crops. 相似文献

Milled oat groat pearlings, trichomes, flour, and bran were extracted with methanol and the fractions tested in vitro for antioxidant capacity against low-density lipoprotein (LDL) oxidation and R-phycoerythrin protein oxidation in the oxygen radical absorbance capacity (ORAC) assay. The oxidative reactions were generated by 2,2'-azobis(2-amidinopropane) HCl (AAPH) or Cu(2+) in the LDL assay and by AAPH or Cu(2+) + H(2)O(2) in the ORAC assay and calibrated against a Trolox standard to calculate Trolox equivalents (1 Trolox equivalent = 1 TE = activity of 1 micromol of Trolox). The antioxidant capacity of the oat fractions was generally consistent with a potency rank of pearlings (2.89-8.58 TE/g) > flour (1.00-3.54 TE/g) > trichome (1.74 TE/g) = bran (1.02-1.62 TE/g) in both LDL and ORAC assays regardless of the free radical generator employed. A portion of the oat antioxidant constituents may be heat labile as the greatest activity was found among non-steam-treated pearlings. The contribution of oat tocols from the fractions accounted for <5% of the measured antioxidant capacity. AAPH-initiated oxidation of LDL was inhibited by the oat fractions in a dose-dependent manner, although complete suppression was not achieved with the highest doses tested. In contrast, Cu(2+)-initiated oxidation of LDL stimulated peroxide formation with low oat concentrations but completely inhibited oxidation with higher doses. Thus, oats possess antioxidant capacity most of which is likely derived from polar phenolic compounds in the aleurone. 相似文献

Length of sample storage can become significant in sensory studies due to panel fatigue limitations and samples needed for a reasonable expectation of finding significant differences. In roasted peanut sensory studies samples are stored between -10 and -23 degrees C to prevent or retard changes. Studies of up to 13 months' duration have examined stability and slow-rate sensory changes. Sweet taste was relatively stable, whereas bitter and tongue burn attributes increased slightly. Stale taste increased, suggesting lipid oxidation was taking place even at -23 degrees C. Painty attribute did not increase until stale was >3. An increase in fruity attribute was unexpected. With increases in fruity and stale attributes a decrease in roasted peanut was expected. However, storage at -23 degrees C seems to stabilize the roasted peanut lability when compared to storage at -10 degrees C. Fruity and stale interactions with roasted peanut and lability of roasted peanut were shown to be three separate and identifiable effects on roasted peanut. 相似文献

Precipitation events were collected during the period from October 1996 to April 1997 from two sites in the city of Amman, Jordan. The samples were analyzed for major cations (Na+, K+, Ca2+, Mg2+ and NH4+), major anions (F-, Cl-, NO3- and SO42-), in addition to heavy metals (Cu2+, Cd2+, Zn2+, and Pb2+). High levels of Ca2+ and SO42- were observed. The results were comparable with some regional values and they indicate that about 58% of Na+ and Cl- are of marine origin, while Ca2+, K+ and a large fraction of SO42- are of soil dust origin. Heavy metals were detectable in most of the samples, zinc showed the highest concentrations. Significant correlation was obtained between zinc and lead, which may indicate automobile emission source. 相似文献

Microbial activity is affected by changes in the availability of soil moisture. We examined the relationship between microbial activity and water potential in a silt loam soil during four successive drying and rewetting cycles. Microbial activity was inferred from the rate of CO2 accumulating in a sealed flask containing the soil sample and the CO2 respired was measured using gas chromatography. Thermocouple hygrometry was used to monitor the water potential by burying a thermocouple in the soil sample in the flask. Initial treatment by drying on pressure plates brought samples of the test soil to six different water potentials in the range -0.005 to -1.5MPa. Water potential and soil respiration were simultaneously measured while these six soil samples slowly dried by evaporation and were remoistened four times. The results were consistent with a log-linear relationship between water potential and microbial activity as long as activity was not limited by substrate availability. This relationship appeared to hold for the range of water potentials from ?0.01 to ?8.5 MPa. Even at ?0.01 MPa (wet soil) a decrease in water potential from ?0.01 to ?0.02 MPa caused a 10% decrease in microbial activity. Rewetting the soil caused a large and rapid increase in the respiration rate. There was up to a 40-fold increase in microbial activity for a short period when the change in water potential following rewetting was greater than 5 MPa. Differences in microbial activity between the wetter and drier soil treatments following rewetting to the original water potentials are discussed in terms of the availability of energy substrate. 相似文献

Penetration resistance, bulk density, soil water content and root growth of oats were intensively studied in a tilled and an untilled grey brown podzolic loess soil. Bulk density and penetration resistance were higher in the top layer of the untilled soil compared with the tilled soil. In the latter, however, a traffic pan existed in the 25–30 cm soil layer which had higher bulk density and penetration resistance than any layer of the untilled soil. Above the traffic pan, rooting density (cm root length per cm3 of soil) was higher but below the pan it was lower than at the same depth in the untilled soil. Root growth was linearly related to penetration resistance. The limiting penetration resistance for root growth was 3.6 MPa in the tilled Ap-horizon but 4.6-5.1 MPa in the untilled Ap-horizon and in the subsoil of both tillage treatments. This difference in the soil strength-root growth relationship is explained by the build up of a continuous pore system in untilled soil, created by earthworms and the roots from preceding crops. These biopores, which occupy < 1% of the soil volume, can be utilized by roots of subsequent crops as passages of comparatively low soil strength. The channeling of bulk soil may counteract the possible root restricting effect of an increased soil strength which is frequently observed in the zero tillage system. 相似文献

Summary Three mollisols, typical of the Palouse winter wheat region of eastern Washington and northern Idaho, were analyzed for microbial biomass, total C and total N after 10 years of combined tillage and rotation treatments. Treatments included till, no-till and three different cereal-legume rotations. All crop phases in each rotation were sampled in the same year. Microbial biomass was monitored from April to October, using a respiratory-response method. Microbial biomass, total C and total N were highest under no-till surface soils (0–5 cm), with minimal differences for tillage or depth below 5 cm. Microbial biomass differences among rotations were not large, owing to the relative homogeneity of the treatments. A rotation with two legume crops had the highest total C and N. Microbial biomass was significantly higher in no-till surface soils where the current crop had been preceded by a high-residue crop. The opposite was true for the tilled plots. There was little change in microbial biomass over the seasons until October, when fresh crop residues and rains had a strong stimulatory effect. The seasonal pattern of biomass in no-till surface soils reflected the dry summer/winter rainfall climate of the region. The results of this study show that numerous factors affect soil microbial biomass and that cropping history and seasonal changes must be taken into account when microbial biomass data are compared.Scientific paper no. 7634 相似文献
