A simple, highly efficient and reproducible two‐step extraction procedure using dilute acetic acid without (AN) and then with sonication (AS) has been developed for the fractionation of wheat flour protein. Approximately 97% of total protein was extracted from a Canadian hard red spring wheat flour; an additional 1.2% protein could be recovered by further extraction with 1% DDT and 50% 1‐propanol (AR). Size‐exclusion HPLC (SE‐HPLC) and flow field‐flow fractionation (flow FFF) showed that the AN extract, which accounted for most of the total extractable protein (AN + AS + AR), consisted primarily of monomeric protein. The AS extract was composed primarily of polymeric proteins. Flow FFF showed that AN polymeric protein, including that eluting at the SE‐HPLC void volume, showed smaller Stokes diameters than AS polymeric protein. Flow FFF profiles of AS SE‐HPLC subfractions showed that the void volume subfraction contained monomeric and small polymeric protein in addition to large polymeric protein, indicating formation of larger complexes through interaction between some or all of the components. AN and AS extracts, as well as SE‐HPLC and flow FFF fractions thereof, showed a fairly wide range of values among 12 Canadian hard red and white spring wheat cultivars. The proportion of total protein in the AS extract and in the larger sized polymeric protein fractions from SE‐HPLC and flow FFF were highly positively correlated to farinograph mixing time. 相似文献
ABSTRACT One would expect foliar applied phosphorus (P) to have higher use efficiencies than when applied to the soil, but limited information is available concerning this. Experiments were conducted in 2002, 2003, and 2004 to determine the effect of foliar applications of P on winter wheat grain yields, P uptake, and use efficiency. Twelve treatments containing varying foliar P rates (0, 1, 2, and 4 kg ha? 1 in 2002 and 2003 and additional 8, 12, 16, and 20 kg ha? 1 in 2004) with and without pre-plant rates of 30 kg ha? 1 were evaluated. Foliar applications of P at Feekes 7 generally increased grain yields and P uptake versus no foliar P. Use efficiency was higher when P was applied at Feekes 10.54. Results from this study suggested that low rates of foliar applied P might correct mid-season P deficiency in winter wheat, and that might result in higher P use efficiencies. 相似文献

in B cell populations undergoing an immune response. Standard classical statistical approaches that use large-sample theory
may lead to misleading conclusions when sample size is very small. The data collection in many biological systems, including
the immune response of vertebrate organisms, is both time consuming and often results in relatively small datasets. Therefore,
this article takes a Bayesian perspective that produces finite-sample estimates of the mutation rates and is flexible in extending

A set of 77 markers was used to describe the genetic diversity in a group of 58 tetraploid wheat accessions. Analysis was performed using 31 neutral SSR markers, 31 SSR/STS markers linked with reported major stem rust resistance genes and 15 SSR markers linked with QTL identified for resistance to Ethiopian stem rust races of Puccinia graminis Pers. f. sp. tritici Eriks. et E. Henn. (Pgt),including Ug99. The material consisted of 32 (Triticum durum s.l. incl. T. aethiopicum Jakubz., Triticum turgidum and Triticum polonicum) landraces and 22 registered T. durum varieties from Ethiopia that were released 1966–2009 and four T. durum varieties from ICARDA. A total of 720 alleles were detected. Considering the three marker sets, the mean number of alleles was higher for major stem rust resistance gene linked markers (9.9) followed by neutral SSR markers (9.2) and markers linked with QTL for stem rust resistance (8.5). Dendrograms derived from UPGMA analysis grouped the accessions into two major clusters. The principal component analysis based on the combination of the three marker sets formed three groups. The 1st group was composed of all the improved varieties, whereas the 2nd and the 3rd group contained the landraces. All the landraces that formed the 3rd group were susceptible to Ethiopian stem rust races of Pgt including Ug99. The information on the extent of the genetic diversity of the improved varieties obtained in this investigation will be helpful for developing appropriate breeding strategies to broadening the genetic base of durum wheat varieties in further breeding programmes. 相似文献
An 11‐year‐old Clydesdale gelding was presented for investigation of left forelimb lameness of 2 weeks' duration. The use of scintigraphic imaging helped to localise the source of lameness to the left proximal humerus. In this report, the clinical and diagnostic imaging features of a primary osseous haemangiosarcoma in a horse are described, along with the challenges of establishing a definitive diagnosis ante mortem. In addition, neoplasia of the appendicular skeleton should be considered a differential cause of lameness in the horse. 相似文献
ABSTRACTPest management professionals in California receive more customer complaints about Argentine ants than for any other urban ant pest. Fipronil, applied as a 30 × 30 cm band around the house foundation, has become the preferred treatment used to control these ants. Unfortunately, fipronil is now showing up in urban waterways at levels that are toxic to aquatic invertebrates. Our recent studies are aimed at mitigating insecticide runoff while still controlling the ant infestations. A high priority is preventing fipronil runoff from the driveway to the street, where it can flow into drains and from there to urban waterways. In this paper, two related studies address these issues. Not treating driveways with fipronil reduced by two to three orders of magnitude its runoff when compared with earlier studies. However, not treating the driveway can reduce efficacy of treatments. Granular bifenthrin, indoxacarb, botanicals, and a thiamethoxam ant bait were tested as supplemental treatments. The gel bait showed the best result as a supplement, but only after 8 weeks. We have reduced fipronil runoff while maintaining efficacy of the ant treatments. 相似文献
There is much interest in quantifying the nutritional value of UK wheat distillers dried grains with solubles (W-DDGS) for livestock species. A study was designed to evaluate caecal parameters (pH, short chain fatty acids (SCFAs) and bacterial diversity) in layer hens fed on balanced diets containing graded concentrations of W-DDGS.
A total of 32 layer hens (Bovans Brown strain at 27 weeks of age) were randomly allocated to one of 4 dietary treatments containing W-DDGS at 0, 60, 120 or 180 g/kg. Each treatment was fed to 8 replicate individually housed layer hens over a 5-d acclimatisation period, followed by a 4-week trial. Individual feed intakes were monitored and all eggs were collected daily for weeks 2, 3 and 4 of the trial, weighed and an assessment of eggshell “dirtiness” made. All hens were culled on d 29 and caecal pH and SCFAs measured. Polymerase chain reaction denaturing gradient gel electrophoresis of the bacterial 16 S rDNA gene was used to assess total bacterial diversity of luminal caecal content from hens fed the 0 and 180 g W-DDGS/kg diets. Unweighted pair group method with arithmetic mean (UPGMA) dendrograms were generated from DGGE banding patterns.
Increasing W-DDGS dietary concentrations resulted in a more acidic caecal environment. Caecal SCFAs were unaffected by diet aside from a quadratic effect for molar proportions of isobutyric acid. Diversity profiles of the bacterial 16S rRNA gene from luminal caecal contents were unaffected by W-DDGS inclusion.
The results of the current study suggest that W-DDGS can be successfully formulated into nutritionally balanced layer diets (supplemented with xylanase and phytase) at up to 180 g/kg with no detrimental effects to the caecal environment.
