Micro-fading spectrometry is a technique that combines visible reflectance spectroscopy and accelerated light aging testing. Therefore, it is a useful tool for determining the light-stability of dyes applied on textile substrates in a relatively short amount of time. Traditional accelerated light aging methods usually require controlling many variables such as time and intensity of illumination, correct positioning of the sample during irradiation, and reproducible spot selection for each subsequent spectrocolorimetric measurement. The use of micro-fade testing as an alternative way of conducting light-fastness assessments of dye/textile systems has been explored. The results indicate that the technique is suitable for testing the color stability of cultural heritage materials and industrially manufactured textiles since it is a direct and non-contact method that takes into account the aforementioned experimental variables.  相似文献

A comparison between traditional extraction techniques (hydrodistillation and organic solvent extraction) and supercritical fluid extraction was made for two different populations and crops of Artemisia absinthium L., cultivated in the field and aeroponically. The composition of the extracts, volatile and non volatile oils, was analyzed by GC-MS and HPLC-DAD, respectively. The antifeedant and phytotoxic activity of the extracts was tested on insect pests (Spodoptera littoralis, Myzus persicae and Rhopalosiphum padi) and plants (Lactuca sativa and Lolium perenne). The supercritical extracts exhibited stronger antifeedant effects than the traditional ones (up to 8 times more active) with moderate selective phytotoxic effects on L. perenne root growth (<50% inhibition).  相似文献

The fall armyworm, Spodoptera frugiperda (Lepidoptera: Noctuidae), is one of the most important pests of corn in Latin America. This insect presents two strains with behavioural and genetic differences. In Mexico, both strains are present, and at least two different FAW populations have been reported within the corn-strain. The objective of this study was to gather evidence of whether pheromonal communication varies among different S. frugiperda populations infesting corn in Mexico. First, we investigated any qualitative or quantitative difference in the composition of sex pheromones among populations; second, we studied whether male antennal responses to pheromone components vary among populations; and, finally, we investigated whether males from a region can discriminate between a synthetic pheromone blend characteristic of their region and blends formulated with the ratio of pheromone compounds emitted by females from other populations. Sex pheromone components were sampled by solid-phase microextraction and identified by gas chromatography coupled to mass spectrometry. Females from all populations consistently released three compounds: (Z)-9-tetradecenyl acetate (Z9-14:OAc), (Z)-11-hexadecenyl acetate (Z11-16:OAc) and (Z)-7-dodecenyl acetate (Z7-12:OAc). Our results showed that Z9-14:OAc and Z7-12:OAc evoked the highest and most variable antennal responses among the populations studied compared to Z11-16:OAc. However, males did not discriminate between local pheromone blends and those formulated from other populations in a field test. These results show that although there is geographic variation in the ratio of pheromone components and in the peripheral reception of them, males were not differentially attracted to different pheromone blends in the field.  相似文献

The process for extracting sugarcane juice (Saccharum officinarum) represents the point of greatest contamination in sugarcane mills. Sodium dithiocarbamate also known as metam-sodium or MS is added to inhibit the growth of microorganisms especially Leuconostoc mesenteroides which is responsible for forming polysaccharides. Metam-sodium, upon decomposition, produces highly toxic byproducts. According to literature, under acidic conditions, MS is hydrolyzed resulting in methylamine (MA), CH3NH2, and carbon disulfide (CS2), and in dilute alkaline solutions, MS produces an oxidation reaction characterized by the formation of elemental sulfur (S) and methyl isothiocyanate (MITC). In this paper, it was studied how MS decomposes to MITC and/or MA considering the effects of the matrix (methanol and water); of temperature (4 and 25 °C); of processing time (0, 1, 2, 3, 4 days); and of pH (4.0, 4.5, 7.0). A second experimental design considering the effects of the matrix (water and sugarcane juice); of temperature (4, 25, 35, 45 °C); of processing time (30, 300 min); and of pH (4.0, 4.5, 7.0) was derived from the results obtained considering MITC and/or MA formation. According to the statistical analysis of these results (p < 0.05), the order of the influential factors was as follows: time > matrix > pH > temperature. Results also indicated that the water matrix at pH = 4.5 and 45 °C had the lowest degradation rate (k), with a value of 8.82 day?1, while for the sugarcane juice matrix at the same pH and temperature conditions was larger, with a k value of 30.07 day?1. These results show that the matrix is also important for the degradation of dithiocarbamate to MITC and to MA.  相似文献

Recombinant adenoviral vectors have emerged as an attractive system for veterinary vaccines development. However, for poultry vaccination a very important criterion for an ideal vaccine is its low cost. The objective of this study was to test the ability of chicken CD154 to enhance the immunogenicity of an adenoviral vector-based vaccine against avian influenza virus in order to reduce the amount of antigen required to induce an effective immune response in avian. Chickens were vaccinated with three different doses of adenoviral vectors encoding either HA (AdHA), or HA fused to extracellular domain chicken's CD154 (AdHACD). Hemagglutination inhibition (HI) assay and relative quantification of IFN-γ showed that the adenoviral vector encoding for the chimeric antigen is able to elicit an improved humoral and cellular immune response, which demonstrated that CD154 can be used as a molecular adjuvant allowing to reduce in about 50-fold the amount of adenoviral vector vaccine required to induce an effective immune response.  相似文献

In order to describe the normal bacterial flora in vaginas of Criollo Limonero cows, 51 healthy multiparous cows, at least 90-day postpartum, were selected. Duplicated swabs (N = 102) were taken from the vaginal fornix of cows to perform aerobic and anaerobic cultures as well as conventional biochemical tests. Out of 102 swabs, bacterial growth was obtained in 55 (53.9%) while the remaining 47 (46.1%) did not exhibited any bacterial growth. Of the 55 bacterial growths, 23 (41.8%) were aerobic whereas 32 (58.1%) were anaerobic. Likewise, 29 (52.72%) of bacterial growths were pure and 26 (47.27%) were mixed. Under both aerobic and anaerobic conditions, Gram positive bacteria were predominant (81.82% and 73.08%, respectively) over Gram negative bacteria (18.18% and 26.92%, respectively). Isolated bacteria were Arcanobacterium pyogenes (22.92%), Staphylococcus aureus (15.63%), Staphylococcus coagulase negative (17.71%), Erysipelothrix rhusiopathiae (6.25%), Bacteroides spp. (13.54%), and Peptostreptococcus spp. (7.29%). In conclusion, normal vaginal bacterial flora of Criollo Limonero cows was predominantly Gram positive and included A. pyogenes, S. aureus, coagulase negative Staphylococcus, E. rhusiopathiae, Bacteroides spp., and Peptostreptococcus spp. In Criollo Limonero cattle, adaptive aspects such as development of humoral and physical mechanisms for defense, and bacterial adaptation to host deserve research attention.  相似文献

The amount and composition of crop residues added to soil in agroecosystems can influence decomposition processes and soil organic matter levels. This study aimed to evaluate residues (quantity and quality) of different corn cultivars commonly used in Brazilian cropping systems. The experiment was conducted for two seasons (2005/2006 and 2006/2007) in Rolândia, Paraná State, Brazil. Ten corn cultivars that represent five degrees of breeding development (i.e., landrace, commercial variety, double cross, triple cross, and single cross hybrids) were evaluated. At harvest, carbon (C) and nitrogen (N) of non-yield residue and grain were determined. Except for grain C, other measures (grain N concentration, residue C and N concentration, and C:N ratio) varied among cultivars. In general, the hybrids had higher residue C and lower residue N concentrations than the landraces and commercial varieties. Findings suggest that breeding selection may have altered residue production and composition, which may influence soil C dynamics.  相似文献

Plant genotypes within species differ widely in tolerance to excess manganese (Mn) that may occur in acid soils, or in neutral or alkaline soils having poor aeration caused by imperfect drainage or compaction. However, Mn tolerance mechanisms in plants are largely unknown. Silicon (Si) is reported to detoxify Mn within plants, presumably by preventing localized accumulations of Mn associated with lesions on leaves. Because Mn is paramagnetic, electron paramagnetic resonance (EPR) spectroscopy, shows promise as a tool for characterizing toxic and non‐toxic forms of Mn in tolerant and sensitive plants. The objective of our study was to use EPR to: i) determine the chemical/ physical state of Mn in Mn‐tolerant and ‐sensitive snapbean cultivars; and ii) characterize the protective effects of Si against Mn toxicity. Manganese‐sensitive Wonder Crop 2 (WC) and Mn‐tolerant Green Lord (GL) cultivars of snapbean were grown at pH 5.0, in a greenhouse, in a modified Steinberg solution containing: Mn=0.05mg.L‐1 (optimal); Mn=1.0mgL‐1 (toxic); Mn=1.0 mg L‐1 plus Si=4 mg L‐1; and Mn=0.05 mg L‐1 plus 4 mg Si L‐1. All trifoliate leaf samples exhibited a 6‐line EPR signal that is characteristic of hexaaquo Mn2+. In both cultivars, a higher EPR Mn2+ signal‐intensity generally correlated with lower total leaf mass, higher total Mn concentrations and more pronounced symptoms of toxicity. Tolerance to excess Mn coincided with lower Mn2+ signal intensity. Silicon treatments ameliorated Mn toxicity symptoms in both genotypes, decreased total leaf Mn concentrations, and decreased EPR Mn2+ signal intensity. Results suggest that Mn toxicity is associated with reduced electron transport and accumulation of oxidation products in leaves. Amelioration of Mn toxicity by Si is regarded as connected with a reduction in this Mn‐induced process. Results indicated that EPR spectroscopy can be useful in investigating the biochemical basis for differential Mn tolerance in plants. The EPR observations might also help plant breeders in developing Mn‐tolerant cultivars.  相似文献

Nicandra physaloides, a common weed in South America, was found to be infected by an isolate of Tomato severe rugose virus (ToSRV), a bipartite begomovirus. The plants developed severe yellow rugose mosaic and were collected in São Paulo State, Brazil. This isolate of ToSRV was transmitted by Bemisia tabaci B biotype from infected plants of N. physaloides to healthy plants of N. physaloides and tomato in a glasshouse. This is the first report of natural infection of N. physaloides by ToSRV in Brazil.  相似文献
