The effect of strenuous exercise on the mRNA concentrations of interleukin-12p35 subunit (IL-12p35), interferon-gamma (IFN-gamma) and interleukin-4 (IL-4) in equine pulmonary and peripheral blood mononuclear cells (PBMCs) was investigated. We hypothesized that strenuous exercise would suppress the expression of IL-12p35, IFN-gamma and augment the expression of IL-4. Eleven horses were randomly divided into two groups, a stall-confined control group (n=5) and an exercise-conditioned treatment group (n=6). Bronchoalveolar and PBMCs were obtained from horses in the treatment group prior to the commencement of a 9-week conditioning program and 24h after the completion of a maximum exercise test conducted in week 12. Samples were obtained simultaneously from control horses. Differential counts were performed on the bronchoalveolar lavage cells. Real-time PCR was performed on the pulmonary and PBMCs to quantitate cytokine expression using equine-specific primers and Taqman probes. Target gene expression was normalized to 18s rRNA expression. With the exception of IL-4 in the BALF cells, mRNA for the three cytokines was detected in the mononuclear cells from all horses at both sampling times. There were no significant differences in the cytokine mRNA concentrations between the two groups of horses at either of the sampling times. These findings demonstrate that strenuous treadmill exercise does not exert a deleterious effect on gene expression for IL-12p35, IFN-gamma or IL-4 when assessed in horses 24h following the intense physical activity.  相似文献

OBJECTIVE: To identify factors associated with abortions of mares during late gestation attributed to mare reproductive loss syndrome (MRLS). DESIGN: Case-control study. ANIMALS: 282 broodmares from 62 farms in central Kentucky, including 137 mares that had late-term abortions (LTAs) associated with MRLS, 98 mares from the same farms that did not abort, and 48 mares that aborted from causes other than MRLS. PROCEDURE: Farm managers were interviewed to obtain data on a wide range of management practices and environmental exposures for the mares. Data for case and control horses were compared to identify risk factors for a mare having a MRLS-associated LTA (MRLS-LTA). RESULTS: Several factors increased the risk of mares having MRLS-LTAs, including increased amount of time at pasture, less time in a stall, feeding concentrate on the ground, higher proportion of diet derived from grazing pasture, being fed in pasture exclusively during the 4-week period prior to abortion, access to pasture after midnight during the 4-week period prior to abortion, and drinking from a water trough or not having access to water buckets or automatic waterers. CONCLUSIONS AND CLINICAL RELEVANCE: Analysis indicates that exposure to pasture predisposed mares to having MRLS-LTAs and stillborn foals. Methods for limiting exposure to pasture (keeping mares in stalls longer) during environmental conditions similar to those seen in 2001 should reduce the risk of mares having MRLS-LTAs.  相似文献

This study was designed to evaluate if the immunosuppression typically observed during the immediate periparturient period (3 weeks before and after calving) in dairy cows influences the effectiveness of diagnostic tests for the detection of Johne's disease; and, if providing additional energy to the cows during this period would minimize any immunosuppressive effects. Twelve dairy cows naturally infected with Mycobacterium paratuberculosis were fitted with rumen cannulas in late gestation and assigned to treatment groups: control, n = 6; or stuffed, n = 6. Cows in the control group were allowed to consume feed ad libitum. Cows assigned to the stuffed treatment group were also fed ad libitum but received additional total mixed ration by manually stuffing their rumens with refused feed to maintain a dry matter intake of 2% body weight/day before calving and 2.5% body weight/day after calving. Parturition had a significant impact on immune function with significant reductions in M. paratuberculosis-specific antibodies detected in the serum and milk regardless of treatment group. Similarly, in vitro immunoglobulin production was decreased at calving for both treatment groups. In addition, stuffing cows modulated cell-mediated immune function by reducing antigen-specific lymphocyte proliferation and interferon-gamma production after calving. Shedding of M. paratuberculosis in the milk was apparent in 58% (7/12) of cows after parturition with no difference noted between control and stuffed animals. Parturition had no major effect on fecal shedding of cows regardless of treatment. These data suggest that parturition had a significant effect on immune function parameters including diagnostic tests for paratuberculosis. Furthermore, providing additional energy to cows with Johne's disease did not preclude immunosuppressive effects during the periparturient period.  相似文献

We characterized the changes in blood glucose concentrations in healthy cats exposed to a short stressor and determined the associations between glucose concentrations, behavioral indicators of stress, and blood variables implicated in stress hyperglycemia (plasma glucose, lactate, insulin, glucagon, cortisol, epinephrine, and norepinephrine concentrations). Twenty healthy adult cats with normal glucose tolerance had a 5-minute spray bath. Struggling and vocalization were the most frequent behavioral responses. There was a strong relationship between struggling and concentrations of glucose and lactate. Glucose and lactate concentrations increased rapidly and significantly in all cats in response to bathing, with peak concentrations occurring at the end of the bath (glucose baseline 83 mg/dL, mean peak 162 mg/dL; lactate baseline 6.3 mg/dL, mean peak 64.0 mg/dL). Glucose response resolved within 90 minutes in 12 of the 20 cats. Changes in mean glucose concentrations were strongly correlated with changes in mean lactate (r = .84; P < .001) and mean norepinephrine concentrations (r = .81; P < .001). There was no significant correlation between changes in mean glucose concentrations and changes in mean insulin, glucagon, cortisol, or epinephrine concentrations. Struggling and lactate concentrations were predictive of hyperglycemia. Gluconeogenesis stimulated by lactate release is the likely mechanism for hyperglycemia in healthy cats in this model of acute stress. Careful handling techniques that minimize struggling associated with blood collection may reduce the incidence of stress hyperglycemia in cats.  相似文献

Ph3SnCl, Ph3SnAc (Brestan), Cy3SnOH (Plictran), and (Bu3Sn)2O (TBTO) markedly reduce the weight of the spleen and the gain in body weight of young mice feeding for 7 days on diets containing 260 μequiv organotin/kg diet. With mature mice feeding for 4 days on diets containing 780 μequiv organotin/kg, these compounds result in a loss in spleen weight and body weight and an alteration in the blood composition (reduction in lymphocytes and total leucocytes and an increase in erythrocytes, hemoglobin level, and hematocrit value). Four other organotins, [(PhCMe2CH2)3Sn]2O (Vendex), PhSnCl2, Ph2SnCl2, and Ph4Sn, are much less toxic based on these parameters. Poisoning by these dietary triorganotins was not related to cerebral edema or alterations in adrenal epinephrine level, liver nonprotein -SH groups, or the activity of digestive enzymes (intestine and stomach proteases and intestine amylase and invertase). The most sensitive indices of triorganotin toxicity appear to be those associated with changes in the lymphatic tissues and blood composition.  相似文献

This study was designed to compare the glucose, insulin, and glucagon responses to consumption of high‐soluble β‐glucan compounds from oats and barley. After an initial medical evaluation that included blood and urine testing, 11 men and 11 women, nondiabetics, 35–57 years, were selected. Subjects consumed a controlled diet for three days. On the third day of five successive periods, subjects consumed 1 g/kg of body weight of carbohydrate as glucose or 0.66 g/kg of body weight pudding (predominantly sucrose) and 0.33 g/kg of body weight as oat bran, barley flour, oat or barley extract (Nu‐trimX) in a Latin square design. Order of treatment was randomly assigned. Glycemic responses were calculated using the trapezoid method. Data were analyzed using mixed procedure analysis of variance program. Glucose responses to oats, barley, and both extracts, and areas under the curve were significantly lower than responses to the glucose solution (P < 0.0001). Insulin responses for the barley extract were lowest and were significantly lower than for glucose solution. Oat and barley extracts retain the beneficial effects of the grains from which they are extracted. High‐soluble fiber barley is more effective than standard oats. Oat and barley carbohydrate‐based fat substitutes can provide a useful addition to menus to control plasma glucose responses.  相似文献

A study was made of the growth and survival of postlarvae and spat of Argopecten nucleus and Nodipecten nodosus produced in the laboratory and exposed to different conditions of settlement and culture. An evaluation was made of the different types of collection substrates (onion-bags, artificial-grass, fibreglass-plates), two types of settlement inducers (epinephrine, low temperature), and two culture environments (sea, laboratory). The results showed that larger numbers of postlarvae were obtained in artificial-grass collectors than on fibreglass-plates or in the onion-bags. The growth of the A. nucleus postlarvae was not affected by the collector type, while that of N. nodosus was higher on fibreglass-plates, and lesser in the onion-bags. Both thermal and chemical stimulation of the competent larvae resulted in recovery of a greater percentage of postlarvae. The growth of postlarvae of A. nucleus was better in groups exposed to the two methods of settlement induction, while those of N. nodosus remained unaffected by this treatment. The survival of the postlarvae and spat was greater in the laboratory than at sea except in the case of A. nucleus postlarvae attached to collectors, whose survival was similar in both environments. The growth of postlarvae and spat was greater in the sea than in the laboratory.  相似文献
