A method of inoculating wet and dry seeds with plant growth-promoting bacteria (PGPB) using alginate microbeads as a substrate and Azospirillum brasilense as the model PGPB was developed. The microbeads were produced by low pressure spraying of an alginate solution mixed with liquid bacterial culture suspended in a very rich medium through a small nozzle resulting in small-diameter droplets. These droplets, when sprayed into a slowly stirred solution of CaCl2, immediately hardened into microbeads at diameters ranging between 100 and 200 µm. Although the process killed part of the entrapped bacteria, the remaining bacteria residing in the microbeads were sufficient [>1011 colony-forming units (CFU) g-1 inoculant] for seed inoculation. Further, it was found that the bacterial population in the inoculant could be enhanced by secondary multiplication in the same medium for an additional 16 h. It was found that the microbeads could be used either wet or dry. Dry inoculant was produced using dry air at 38°C, creating a powdery substance loaded with >109 CFU g-1 beads. Alternatively, dry microbeads were produced using a standard freeze-drying procedure. This dry preparation was easily attached to dry seed surfaces with the addition of 1% alcohol-diluted lecithin or with 0.5% synthetic paper adhesive (Resistol). The bacteria were slowly released from the microbeads in amounts ranging from 104 to 106 CFU g-1 depending on the type (wet or dry, with or without skim milk) and the time of incubation (the longer the incubation period, the smaller the amount of bacteria released with time). The wet and dry inoculants enhanced the development of wheat and tomato seedlings growing in unfertile soil, and biodegraded within 15 days in moist soil.  相似文献

Soil degradation by deforestation and introduction of exotic grasses is a grave consequence of land‐use change in tropical regions during the last decades. Soil restoration following natural succession (i.e., passive restoration) is slow because of low tree establishment. Introduction of tree plantings by human intervention (i.e., active restoration) results in a promising strategy to accelerate forest succession and soil recovery in tropical region. The present research was carried out to explore the restoration of soil properties after cattle exclusion and of grazing combined with native tree planting introduction (legumes and nonlegumes) in a tropical pasture in Veracruz, southeast Mexico. Results indicate that land‐use changes decreased soil C and N pools in both litter and mineral soil. In addition, soil heterogeneity increased by land‐use changes at both temporal and spatial scales. In the short term, passive succession (i.e., cattle exclusion) favors the recovery of C and N content in labile soil pools, indicated by the increase in litter C and N masses as well as C and N concentrations in soil microbial biomass. Soils under active restoration showed trends to recover the N cycling, such as a greater accumulation of N in litter, in soil total N concentrations, soil microbial biomass N concentrations, rates of net N transformations, and extractable water and microbial biomass C:N ratios mainly under legumes species. BACKGROUND: Biopesticides containing Cry insecticidal proteins from the bacterium Bacillus thuringiensis (Bt) are effective against many lepidopteran pests, but there is a lack of Bt‐based pesticides for efficient control of important coleopteran pests. Based on the reported increase in Bt toxin oligomerization by a polypeptide from the Cry3Aa receptor cadherin in Tenebrio molitor (Coleoptera: Tenebrionidae), it was hypothesized that this cadherin peptide, rTmCad1p, would enhance Cry3Aa toxicity towards coleopteran larvae. To test this hypothesis, the relative toxicity of Cry3Aa, with or without rTmCad1p, against damaging chrysomelid vegetable pests of China was evaluated. RESULTS: Cry3Aa toxicity was evaluated in the spotted asparagus beetle (Crioceris quatuordecimpunctata), cabbage leaf beetle (Colaphellus bowringi) and daikon leaf beetle (Phaedon brassicae). To assess the effect of rTmCad1p on Cry3Aa toxicity, neonate larvae were fed Cry3Aa toxin alone or in combination with increasing amounts of rTmCad1p. The data demonstrated that Cry3Aa toxicity was significantly increased in all three vegetable pests, resulting in as much as a 15.3‐fold increase in larval mortality. CONCLUSION: The application of rTmCad1p to enhance Cry3Aa insecticidal activity has potential for use in increasing range and activity levels against coleopteran pests displaying low susceptibility to Bt‐based biopesticides. Introduction

Hypertrophic cardiomyopathy (HCM) has a variable prognosis; left atrial size, presence of clinical signs and left ventricular systolic function have been shown to predict outcomes. Mitral annular plane systolic excursion (MAPSE) and tricuspid annular plane systolic excursion (TAPSE) assess longitudinal ventricular systolic function and are decreased in cats with HCM. The aim of the study was to ascertain whether MAPSE and TAPSE have prognostic value in HCM and if cats with pleural effusion have lower MAPSE and TAPSE than cats with pulmonary oedema.

Animals

One hundred eighty-four client-owned cats diagnosed with HCM.

Methods

This is a retrospective study. Echocardiography was used to diagnose HCM (end-diastolic left ventricular wall thickness ≥ 6 mm) and to measure MAPSE and TAPSE. Survival information was obtained.

Results

No multivariable model including MAPSE or TAPSE could be generated in this population. Cats with pleural effusion ± pulmonary oedema had lower MAPSE measured at the interventricular septum (MAPSE IVS) and TAPSE, compared with cats with pulmonary oedema only. MAPSE IVS was the only factor predicting pleural effusion on multivariable regression model.

Conclusions

Lower MAPSE and TAPSE were not independently associated with outcomes on multivariable analysis. Cats with pleural effusion ± pulmonary oedema had lower TAPSE and MAPSE IVS than cats with pulmonary oedema, and MAPSE IVS was the only predictive factor associated with the development of pleural effusion in this population.  相似文献

OBJECTIVES: To report the clinical findings associated with pulsus alternans in English cocker spaniels with dilated cardiomyopathy and to review the phenomenon of mechanical alternans, usually found in patients with severe left ventricular systolic dysfunction. METHODS: The case records of 10 English cocker spaniels with dilated cardiomyopathy and pulsus alternans were reviewed. All dogs had been referred in moderate to severe congestive heart failure. Pulsus alternans was clinically recognised by palpation of a regular alternation of femoral pulse amplitude and confirmed by echocardiography in all cases. Pulsus alternans was an intermittent finding in all 10 dogs. RESULTS: Phonocardiographic recordings were obtained in three dogs to document variation in the intensity of the heart sounds, with one case exhibiting absence of the second heart sound in alternate beats. M-mode echocardiography performed in all dogs showed alternation of the pattern of mitral valve diastolic motion. Doppler echocardiography showed marked alternation in stroke volume in the aortic outflow in all cases. It also showed alternation in mitral regurgitation and in the velocity of early ventricular filling. CLINICAL SIGNIFICANCE: Pulsus alternans may be more prevalent in English cocker spaniels than in other breeds, and because of its intermittent nature, its incidence may be higher than that previously reported. It has diagnostic relevance as it usually indicates severe myocardial depression. Echocardiography allows non-invasive detection of mechanical alternans and provides further insights into its pathophysiology and clinical significance.  相似文献
