Atrophic rhinitis (AR) is a widespread and economically important disease of swine caused by Bordetella bronchiseptica and Pasteurella multocida. It can be controlled by vaccination. This study investigates the effect of altering the composition (adjuvants and/or addition of formalin-inactivated P. multocida toxin, fPMT) of conventional vaccines on the serological profile and on protection against AR in swine. A significantly higher B. bronchiseptica specific antibody titre was detected for vaccines with novel immunostimulants, the best being Montanide IMS 1313 (1:630 compared to 1:274 obtained with alum). The highest B. bronchiseptica antibody titre was demonstrated for a combination of B. bronchiseptica--fPMT, while PMT antibody titre was highest for monovalent fPMT (both adjuvanted with IMS 1313). The AR-specific antibodies were transmitted from dams to their offspring in similar titres and with the same hierarchy of effectiveness. After a B. bronchiseptica--P. multocida bacterial challenge, piglets from dams vaccinated with fPMT combined with B. bronchiseptica or B. bronchiseptica--P. multocida bacterins showed the lowest nasal lesions scores (4.5 and 3.2, respectively, out of a possible maximum score of 18). These combinations, both of which were adjuvanted with IMS 1313, gave the best protection against experimentally induced AR. Our results show that the adjuvant and the antigen composition of the vaccine strongly affect seroconversion, and that the AR-specific antibody titre does not necessarily correlate with the degree of protection.  相似文献

Gastrointestinal parasites are important infectious causes of diarrhoea in captive non-human primates (NHP). However, prevalence data of gastrointestinal parasites in zoological gardens are scarce. Therefore, a cross-sectional survey was conducted to estimate the occurrence of gastrointestinal parasites in NHP of four zoological gardens in Belgium. Between August 2004 and April 2006, 910 faecal samples were collected from 222 animals housed in 39 groups. The 31 species involved were representatives of prosimians, New World (NW) monkeys, Old World (OW) monkeys and apes. Because individual sampling was impossible, a statistical simulation was performed to estimate a sufficient sample size. All samples were microscopically examined after an acetic acid-ether concentration. Differences in host species susceptibility were examined by non-parametric tests. Entamoeba spp. (44%) and Giardia spp. (41%) were the most prevalent species. Other parasites detected were Endolimax nana (36%), Chilomastix mesnili (21%), Balantidium coli (13%), Trichuris spp. (10%), Iodamoeba bütschlii (5%) and Strongyloides spp. (5%). Parasites for which a significant difference in susceptibility at the level of host taxonomy was noted were Entamoeba spp. (p<0.001) and C. mesnili (p<0.05). Samples containing Entamoeba spp. were the most prevalent in OW monkeys (p<0.0083). Samples collected from OW monkeys contained the highest number of parasite species (p<0.0083).  相似文献

Extrudate expansion of cereal‐based products is largely dependent on the molecular interactions and structural transformations that proteins undergo during extrusion processing. Such changes strongly influence the characteristic rheological properties of the melt. It is possible to modify rheological properties of wheat flour during extrusion processing, in particular shear viscosity, with cysteine. The objective of this work was to further develop an understanding of the molecular interactions and structural transformations of wheat flour from dynamic oscillatory rheological measurements. Temperature and frequency sweeps were conducted in the linear viscoelastic range of the material. Changes in the storage modulus (G′), the loss modulus (G″) and the loss tangent (tan δ) of 25% moisture wheat flour disks as a function of cysteine concentration (0–0.75%) were monitored. Molecular weight between cross‐links (Mc) and the number of cross‐links (Nc) per glutenin molecule were determined from frequency sweep data. Increasing cysteine concentration broke cross‐links by decreasing G′ maximum and increasing tan δ values. Molecular weight between cross‐links increased and the number of cross‐links decreased. G′ values from temperature sweeps showed a similar trend. This information leads to a better understanding of the viscoelastic behavior of wheat flour doughs during extrusion cooking and elucidation of protein‐protein reaction mechanisms and other interactions in extruded cereal‐based snack foods.  相似文献

Depositions originating from a central Slovak Al smelter may increase metal solubility in adjacent soils because they contain F (mainly HF). The reason for fluoro‐mobilization of metals may be the formation of soluble fluoro‐metal complexes or the mobilization of organic matter and subsequent formation of organo‐metal complexes. The objectives of our work were (1) to assess the extent of metal mobilization by fluoride in a Slovak Lithic Eutrochrept affected by the emissions of an Al smelter and (2) to model the dissolved metal species with the help of a chemical equilibrium model (MINEQL+). The O (Moder), A, and B horizons were equilibrated with solutions at F concentrations of 0, 0.9, 2.7, and 9.0 mmol l—1. In the extracts, the concentrations of Al, Ca, Cd, Cr, Cu, Fe, K, Mg, Mn, Ni, Pb, Zn, dissolved organic carbon (DOC), free and complexed F, and the pH and electrical conductivity (EC) were determined. The heavy metal concentrations in the O horizon (Cd: 0.99, Cr: 18.0, Cu: 44, Ni: 26, Pb: 110, and Zn: 84 mg kg—1) were 2.5 to 9 times larger than those in the A and B horizons. The concentrations of H2O‐soluble F decreased from the O (261 mg kg—1) to the A (103 mg kg—1) and B horizon (92 mg kg—1). In batch experiments increasing addition of F increased the equilibrium concentrations of Al, Cr, Cu, Fe, Ni, Pb, and DOC in all samples, of Cd in the A, and of K in the B horizon. At the same time the concentrations of complexed F and pH increased whereas EC decreased. Chemical equilibrium modelling indicated that the mobilizing effect of F resulted from the formation of fluoro‐Al complexes and organo‐complexes of all other metals.  相似文献

The objective of this study was to test the suitability of a simple approach to identify the direction from where airborne heavy metals reach the study area as indication of their sources. We examined the distribution of heavy metals in soil profiles and along differently exposed transects. Samples were taken from 10 soils derived from the same parent material along N-, S-, and SE-exposed transects at 0—10, 10—20, and 20—40 cm depth and analyzed for total Al, Cd, Cr, Cu, Fe, Mn, Ni, Pb, and Zn concentrations. The heavy metal concentrations at 0—10 cm were larger than background concentrations in German arable soils except for Cr (Cd: 0.6—1.8 mg kg—1; Cr: 39—67; Cu: 40—77; Ni: 87—156; Pb: 48—94; Zn: 71—129; Fe: 26—34 g kg—1; Mn: 1.1—2.4). Decreasing Cd, Cu, Mn, and Pb concentrations with increasing soil depth pointed at atmospheric inputs. Aluminum and Ni concentrations increased with soil depth. Those of Fe, Cr, and Zn did not change with depth indicating that inputs at most equalled leaching losses. The Pb accumulation in the surface layer (i.e. the ratio between the Pb concentrations at 0—10 to those at 20—40 cm depth) was most pronounced at N-exposed sites; Pb obviously reached Mount Križna mainly by long-range transport from N where several industrial agglomerations are located. Substantial Cd, Cu, and Mn accumulations at the S- and SE-exposed sites indicated local sources such as mining near to the study area which probably are also the reason for slight Cr and Zn accumulations in the SE-exposed soils. Based on a principal component analysis of the total concentrations in the topsoils four metal groups may be distinguished: 1. Cr, Ni, Zn; 2. Mn, Cd; 3. Pb (positive loading), Cu (negative loading); 4. Al, Fe, indicating common sources and distribution patterns. The results demonstrate that the spatial distribution of soil heavy metal concentrations can be used as indication of the location of pollution sources.  相似文献

With increasing demand for sustainable production, the need to effectively utilise site-based resources increases. One such resource is the remaining crop residues, both above and below ground, after harvest. In order to assess the magnitude of this resource, this study determined the plant nutrient contribution of residues from 17 different crops in seven different regions of Slovakia over a nine-year period. The soil profile 0.0–0.3?m was taken into account at belowground residues sampling. The results showed that winter rape, mustard, corn maize, triticale, winter rye with straw and sunflower left the most residues, exceeding 8 tons of dry biomass per hectare. Root crops (sugar beet, potatoes) and peas left less than 3 tons of residual dry biomass per hectare. Using these data, the nutrient potential coefficient, i.e. the amount of nutrients left by production of one ton of the main product was calculated by polynomial regression. With these coefficients and the known yield of a given crop, it was possible to calculate how much nitrogen, phosphorus and potassium each crop leaves in the soil after harvest. It was found that the amount of nitrogen left ranged from 20 to 132?kg, phosphorus from 2 to 24?kg and potassium from 13 to 218?kg per hectare. This has to be taken into account when calculating the fertiliser requirement of the subsequent crop in order to achieve better resource utilisation, thereby reducing the risk of eutrophication and improving farm profits by reducing expenditure on fertiliser.  相似文献

Soil organic matter (SOM) is a key factor in ecosystem dynamics. A better understanding of the global relationship between environmental characteristics, ecosystems and SOM chemistry is vital in order to assess its specific influence on carbon cycles. This study compared the composition of extracted SOM in 18 topsoil samples taken under tundra, taiga, steppe, temperate forest and tropical forest using pyrolysis-GC/MS. Results indicate that SOM from cold climates (tundra, taiga) still resembles the composition of litter, evidenced by high quantities of levosugars and long alkanes relative to N-compounds and a clear odd-over-even dominance of the longer alkanes. Under temperate conditions, increased microbial degradation generally results in a more altered SOM chemistry. SOM formed under temperate coniferous forests shows an accumulation of aromatic and aliphatic moieties, probably induced by substrate limitations. Tropical SOM was characterized by an SOM composition rich in N-compounds and low in lignins, without any accumulation of recalcitrant fractions (i.e. aliphatic and aromatic compounds). Lignin composition moreover varies according to vegetation type. Results were validated against 13 new samples. The humic signature of topsoil organic matter formed under different biomes indicates a dominating effect of (i) SOM input composition related to vegetation, and (ii) SOM breakdown reflecting both climate and input quality. No evidence was found for a chemically stabilized SOM fraction under favorable decomposition conditions (temperate or warm climate with broadleaved vegetation).  相似文献

According to the European Food Safety Authority, salmonellosis is still one of the main causes of infectious foodborne gastroenteritis in humans. Broilers are an important source of salmonellosis after eggs and pork. Between 1987 and 1999 the trend of human salmonellosis incidence in Belgium increased constantly. However, from 2000 until 2005 a decrease in human cases was observed, probably following the sanitary measures implemented in the poultry breeder and laying sector. In order to decrease human infections it is essential to tackle the problem at the farm level to minimize cross-contamination from farm to fork. This paper seeks to answer two questions: (i) given the Salmonella status of the farm at a certain occasion (equal to the sampling time of the flock), what are the risk factors that the farm will be Salmonella positive at a following occasion? And (ii) what are the risk factors for a farm to be persistently positive for two consecutive flocks? We used surveillance data on 6824 broiler flocks studied for Salmonella infectivity from 2005 to 2006 in Belgium. The farms were tested regularly (3 weeks before slaughter of each broiler flock) for the presence of Salmonella based on multiple faecal samples per flock on a farm yielding clustered data. Generalized estimating equations, alternating logistic regression models, and random-intercept logistic regression models were employed to analyse these correlated binary data. Our results indicated that there are many factors that influence Salmonella risk in broiler flocks, and that they interact. Accounting for interactions between risk factors leads to an improved determination of those risk factors that increase infection with Salmonella. For the conditional analysis, the risk factors found to increase the risk of Salmonella infection on a farm at a current occasion given the previous Salmonella status included: Salmonella infection of day-old chicks (of the current flock); a previously infected flock even though the farm was equipped with a hygiene place to change clothes prior to entering the broiler house; having temporary workmen when there was a separation between birds of different species; and separating birds of different species in the Walloon region relative to the Flanders region. Sanitary measures such as a cleaning and disinfecting procedure conducted by an external cleaning firm, applying the all-in all-out procedure, and hand washing decreased the risk despite their interaction with other factors. From the joint analysis, the most important factors identified for increased risk for persistent Salmonella on a farm involved the interaction between having temporary workmen when there were poultry or farmers in contact with foreign poultry or persons, and the interaction between having temporary workmen when there were poultry or farmers in contact with external poultry or persons.  相似文献
